Deze video toont een protocol methode voor het genereren van transgene<em> Xenopus laevis</em> Door de introductie van transgenen in spermakernen gevolgd door nucleaire transplantatie in onbevruchte eieren.
Stabiele integratie van gekloonde gen producten in de Xenopus genoom is nodig om de tijd en plaats van meningsuiting controle, om genen te uiten in een later stadium van de embryonale ontwikkeling, en om te bepalen hoe enhancers en initiatiefnemers van genexpressie reguleren binnen het embryo. Het protocol blijkt hier kan gebruikt worden om efficiënt te produceren transgene Xenopus laevis embryo's. Deze transgenese wordt in drie delen: 1. Spermakernen zijn geïsoleerd van volwassen X. laevis testis door behandeling met lysolecithin, waarbij het sperma plasmamembraan permeabilizes. 2. Ei extract wordt bereid door lage snelheid centrifugeren, toevoeging van calcium aan het extract zorgen om de voortgang te interfase van de celcyclus, en een high-speed centrifugatie om interfase cytosol isoleren. 3. Nucleaire transplantatie: de kernen en extract worden gecombineerd met de gelineariseerde plasmide DNA worden ingevoerd als het transgen en een kleine hoeveelheid van de restrictie-enzym. Tijdens een korte reactie, ei-extract decondenses gedeeltelijk het sperma chromatine en de restrictie-enzym genereert chromosomale breekt dat recombinatie van het transgen te bevorderen in het genoom. De behandelde spermakernen worden vervolgens getransplanteerd in onbevruchte eieren. Integratie van het transgen gebeurt meestal voorafgaand aan de eerste embryonale klieving zodanig dat de ontstane embryo's niet chimerisch zijn. Deze embryo's kunnen worden geanalyseerd zonder dat te fokken voor de volgende generatie, waardoor een efficiënte en snelle generatie van transgene embryo's voor analyses van de promotor en gen functie. Volwassen X. laevis als gevolg van deze procedure ook propageren het transgen door de kiembaan en kan gebruikt worden om lijnen van transgene dieren voor meerdere doeleinden te genereren.
Voor elke transgene construct te testen, hebben we over het algemeen transplantatie kernen in 500-1000 eieren, op deze schaal, kunnen we het genereren van transgene embryo's uitdrukken tot 10 verschillende constructen per dag, afhankelijk van het aantal vrouwtjes worden aangezet om eieren te leggen. Van deze transplantaties, ongeveer een derde van de eieren aankleven en 60-80% van deze embryo's splitsen normaal verder door gastrulatie. Agarose dishes for injection: In a 60 mm plastic petri dish, lay a small 35mmX35mm weigh boat on molten 2.5% agarose in water 0.1XMMR to create a depression with an agarose-coated bottom for filling with eggs. Once agarose has hardened, wrap in parafilm and store at 4°C until use. Make 2-3 dishes in advance for each transgenic reaction you plan to do.
Infusion pump: We use a single syringe infusion pump from Harvard Apparatus, equipped with a 3 cc syringe/needle filled with mineral oil (Sigma M-8410). Blunt the syringe needle tip (to keep it from perforating the tubing) and attach the fine tygon tubing. Run the pump at ~10nl/sec; this assumes that the time the needle is in each egg will be no greater than 1 sec. Pump should be pre-run for several minutes prior to starting transgenesis for the day to assure that the plunger for the syringe is flush with the piston and that steady positive flow of oil out of the tubing is occurring.
Needles for nuclear transfers. Using a micropipette puller, generate needles with long, sloping tips. Clip these with a forcep under a dissecting microscope equipped with an ocular micrometer to obtain an ~80 micron opening with a beveled shape.
