Ce protocole vidéo montre une méthode pour générer des transgéniques<em> Xenopus laevis</em> Par l'introduction de transgènes dans les noyaux des spermatozoïdes suivie par transplantation nucléaire dans les œufs non fécondés.
L'intégration stable de produits de gène cloné dans le génome de Xenopus est nécessaire de contrôler l'heure et le lieu d'expression, d'exprimer des gènes à des stades ultérieurs du développement embryonnaire, et de définir comment les amplificateurs et les promoteurs régulent l'expression génique au sein de l'embryon. Le protocole démontré ici peuvent être utilisés pour produire efficacement transgéniques Xenopus laevis embryons. Cette approche implique la transgénèse trois parties: 1. Noyaux spermatozoïdes sont isolés des adultes X. testicule laevis par traitement avec lysolécithine, qui perméabilise la membrane plasmique des spermatozoïdes. 2. Extraire des oeufs est préparé par centrifugation à faible vitesse, ajout de calcium pour provoquer l'extrait de progresser vers l'interphase du cycle cellulaire, et une centrifugation à haute vitesse pour isoler cytosol interphase. 3. Transplantation nucléaire: les noyaux et extraire sont combinés avec l'ADN plasmidique linéarisé d'être présenté comme le transgène et une petite quantité d'enzyme de restriction. Lors d'une réaction très court, extrait d'oeuf décondense partie de la chromatine des spermatozoïdes et l'enzyme de restriction génère cassures chromosomiques qui favorisent la recombinaison du transgène dans le génome. Les noyaux des spermatozoïdes traités sont ensuite transplantées dans les œufs non fécondés. L'intégration du transgène se produit habituellement avant le premier clivage embryonnaire tels que les embryons qui en résultent ne sont pas chimériques. Ces embryons peuvent être analysées sans qu'il soit nécessaire de se reproduire à la prochaine génération, permettant la génération rapide et efficace des embryons transgéniques pour des analyses de promoteur et de la fonction des gènes. Adulte X. laevis résultant de cette procédure également propager le transgène dans la lignée germinale et peut être utilisé pour générer des lignées d'animaux transgéniques à des fins multiples.
Pour chaque construction transgénique à être testé, nous avons généralement greffe noyaux dans les œufs 500-1000; à cette échelle, nous pouvons générer des embryons transgéniques exprimant jusqu'à 10 différentes constructions par jour, selon le nombre de femelles sont induits à pondre des oeufs. Agarose dishes for injection: In a 60 mm plastic petri dish, lay a small 35mmX35mm weigh boat on molten 2.5% agarose in water 0.1XMMR to create a depression with an agarose-coated bottom for filling with eggs. Once agarose has hardened, wrap in parafilm and store at 4°C until use. Make 2-3 dishes in advance for each transgenic reaction you plan to do.
Infusion pump: We use a single syringe infusion pump from Harvard Apparatus, equipped with a 3 cc syringe/needle filled with mineral oil (Sigma M-8410). Blunt the syringe needle tip (to keep it from perforating the tubing) and attach the fine tygon tubing. Run the pump at ~10nl/sec; this assumes that the time the needle is in each egg will be no greater than 1 sec. Pump should be pre-run for several minutes prior to starting transgenesis for the day to assure that the plunger for the syringe is flush with the piston and that steady positive flow of oil out of the tubing is occurring.
Needles for nuclear transfers. Using a micropipette puller, generate needles with long, sloping tips. Clip these with a forcep under a dissecting microscope equipped with an ocular micrometer to obtain an ~80 micron opening with a beveled shape.
