Studien beskriver en kostnadseffektiv metode for identifisering av kilden til fekal / urin forurensning eller forurensning av nitrater i vann med qPCR for de spesifikke kvantifisering av menneskelig / svin / storfe DNA virus, adenoviruses og polyomaviruses, foreslått som MST verktøy.
Mikrobiell forurensning av miljøet representerer en betydelig helserisiko. Har klassisk bakteriell fecal indikatorer vist seg å ha betydelige begrensninger, virus er mer resistente mot mange inaktivering prosesser og standard fecal indikatorene ikke informere om kilden til forurensning. Utvikling av kostnadseffektive metoder for konsentrasjonen av virus fra vann og molekylære analyser forenkler anvendelsen av virus som indikatorer på fekal forurensning og som mikrobiell kilde tracking (MST) verktøy. Adenoviruses og polyomaviruses er DNA virus infiserer spesifikke arter virveldyr, inkludert mennesker, og er vedvarende skilles ut i avføring og / eller urin i alle geografiske områder studert. I tidligere studier, foreslo vi kvantifisering av menneskelig adenoviruses (HAdV) og JC polyomaviruses (JCPyV) av kvantitativ PCR (qPCR) som en indeks for menneskelig fekal forurensning. Nylig har vi utviklet qPCR analyser for de spesifikke kvantifisering av porcine adenoviruses (PAdV) og storfe polyomaviruses (BPyV) som dyr fecal markører for forurensing med følsomheter fra 1-10 genom eksemplarer per reagensrør. I denne studien presenterer vi prosedyren som skal følges for å identifisere kilden til forurensning i vannprøver ved hjelp av disse verktøyene. Som eksempel på representative resultater, er analyse av virus i grunnvann presentere høye nivåer av nitrater vist.
Påvisning av virus i lav eller moderat forurenset vann krever konsentrasjon av virus fra minst flere liter vann i et mye mindre volum, en prosedyre som vanligvis inkluderer to konsentrasjon trinn i serien. Denne noe tungvinte prosedyren og variasjonen hos viral inngang betydelig hinder for samtidig behandling av et stort antall vannprøver.
For å fjerne flaskehalsen forårsaket av to-trinns prosedyrer vi har brukt en ett-trinns protokoll utviklet i tidligere studies og gjelder for et mangfold av vann matriser. Prosedyren omfatter: forsuring av ti-liters vannprøver, flokkulering av skummet melk, gravitasjon sedimentering av flocculated materialer, samling av bunnfall og sentrifugering, resuspendering av bunnfall i 10 ml fosfat buffer. Den viral konsentrere brukes til utvinning av virale nukleinsyrer og de spesifikke adenoviruses og polyomaviruses av interesse er kvantifisert av qPCR. Høyt antall prøver kan samtidig analyseres ved hjelp av denne lavkost konsentrasjon metoden.
Prosedyren har blitt brukt til analyse av badevann, sjøvann og elvevann og i denne studien presenterer vi resultatene analysere grunnvann prøver. Denne high-throughput kvantitative metoden er pålitelig, enkel og kostnadseffektiv.
Prosedyren er beskrevet ville oppfylle vilkårene for en passende metode for rutinemessig miljø og folkehelse laboratorier: reproduserbar, pålitelig, enkel og kostnadseffektiv. Protokollen er enkel, men den må følges nøye. Lav ledningsevne i prøvene uten å legge den ønskede konsentrasjon av kunstig sjøvann salter vil dramatisk redusere utvinning av virus som ville være tilfelle dersom omrøring tid for flokkulering er betydelig redusert (mindre enn 5 timer for eksempel).
