Stroke är den tredje vanligaste dödsorsaken och den vanligaste orsaken till funktionshinder i världen, med en beräknad kostnad på nära $ 70 miljarder USA under 2009 1,2. Den intraluminal mellersta cerebral artär ocklusion (kortex) modellen utvecklades av Koizumi 4 1986 för att simulera detta effektfulla mänskliga patologi hos råtta. En ändring av kortex metoden senare presenterades av Longa 3. Båda teknikerna har ofta använts för att identifiera molekylära mekanismer för hjärnskador till följd av ischemisk stroke och potentiella terapeutiska metoder 5. Denna relativt icke-invasiv metod hos råttor har utvidgats till att använda i möss att dra nytta av transgena och stammar knockout 6,7. Att modellera fokal cerebral ischemi, är en intraluminal sutur avancerat via det interna halspulsådern för att täppa till basen på MCA. Dra tillbaka sutur efter en viss tid härmar spontan reperfusion, men sutur kan också vara permanent behållas. Denna video kommer att demonstrera de två stora metoder för att utföra intraluminal kortex förfarande i möss i en stegvis, samt att ge insikter för potentiella nackdelar och fallgropar. Den ischemisk hjärnvävnad kommer senare att färgas av 2,3,5-triphenyltetrazolium klorid (TTC) för att utvärdera omfattningen av hjärninfarkt 8.
Den kortex modellen är en etablerad modell som replikerar cerebral ischemi i murina arter. Denna modell användes först på råttor och därefter anpassas till möss.
