Dit artikel zal zich richten op het genereren van humane lever endoderm van menselijke embryonale stamcellen populaties.
Ondanks de vooruitgang in het modelleren van menselijke geneesmiddelen toxiciteit, veel stoffen niet tijdens klinische studies als gevolg van onverwachte bijwerkingen. De kosten van de klinische studies substantieel zijn, daarom is het essentieel dat meer voorspellende toxicologie schermen zijn ontwikkeld en vroeg ingezet in de ontwikkeling van geneesmiddelen (Greenhough et al., 2010). Humane hepatocyten vertegenwoordigen de huidige gouden standaard model voor het evalueren van drugs toxiciteit, maar zijn een beperkte hulpbron die variabele functie vertonen. Daarom zijn het gebruik van onsterfelijk gemaakte cellijnen en dierlijk weefsel modellen routinematig ingezet als gevolg van hun overvloed. Hoewel beide bronnen zijn informatief, worden ze beperkt door een slechte werking, soorten variabiliteit en / of instabiliteit in de cultuur (Dalgetty et al. 2009). Pluripotente stamcellen (PSC's) zijn een aantrekkelijk alternatief bron van menselijke hepatocyten achtige cellen (HLCS) (Medine et al. 2010). PSC's zijn in staat om zelfvernieuwing en differentiatie van alle somatische cellen soorten gevonden in de volwassen en daarmee vertegenwoordigeneen potentieel onuitputtelijke bron van gedifferentieerde cellen. We hebben een procedure ontwikkeld die eenvoudig, zeer efficiënt, vatbaar voor automatisering en levert functionele menselijke HLCS (Hay et al., 2008; Fletcher et al. 2008; Hannoun et al. 2010; Payne et al. 2011 en Hay et al. 2011). Wij geloven dat onze technologie zal leiden tot de schaalbare productie van HLCS voor drug discovery, ziekte modelleren, de bouw van extra-lichamelijke-apparaten en eventueel cell-based therapieën transplantatie.
We hebben een eenvoudig, homogeen en zeer reproduceerbaar in vitro model om schaalbare niveaus van menselijke HLCS te genereren. Ons model is gevalideerd door een aantal externe samenwerkende laboratoria. We routinematig karakteriseren van stamcellen afgeleid HLCS het gebruik van onze in-house gereedschapskist van de ontwikkelings-markers en de lever specifieke functionele assays (waarvan de meeste in de handel verkrijgbaar).
