Protokollet som beskrivs här syftar till att förklara och förkorta de många hinder i vägen för den intrikata vägen som leder till modifierade nukleosidtrifosfater. Följaktligen underlättar detta protokoll både syntesen av dessa aktiverade byggstenar och deras tillgänglighet för praktiska tillämpningar.
Den traditionella strategin för införandet av kemiska funktionaliteter är användningen av fast-fas-syntes genom att lägga lämpligt modifierade fosforamidit prekursorer till den framväxande kedjan. Men de villkor som används under syntesen och begränsningen till ganska korta sekvenser hindra tillämpligheten av denna metod. Å andra sidan, är modifierade nukleosidtrifosfater aktiverade byggstenar som har använts för den milda inledningen av ett stort antal funktionella grupper i nukleinsyror, en strategi som banar väg för användning av modifierade nukleinsyror i en bred palett av praktiska tillämpningar såsom funktionell märkning och generering av ribozymer och DNAzymes. En av de stora utmaningarna är bosatt i intrikat av den metod som leder till isolering och karakterisering av dessa nukleosidanaloger.
I denna video artikeln presenterar vi ett detaljerat protokoll för syntes av these modifierade analoger med hjälp av fosfor (III)-baserade reagenser. Dessutom är det förfarande för biokemisk karakterisering röjas, med särskild tonvikt på primerextensionsprodukter reaktioner och TdT svans polymerisation. Denna detaljerade protokoll kommer att vara till nytta för att tillverka av modifierade dNTP och deras vidare användning i kemisk biologi.
5'-nukleosidtrifosfater ((d) NTP) representerar en klass av viktiga biomolekyler som är inblandade i otaliga processer och funktioner som sträcker sig från att vara den universella valutan av energi till regulatorer av cellmetabolism. Förutom deras roll i dessa fundamentala biologiska förändringar, har sina modifierade motsvarigheter fram som en mångsidig och mild plattform för införandet av funktionella grupper i oligonukleotider, en metod som fint kompletterar den automatiska fastfassyntesen som brukar tillämpas 1,2. I själva verket, förutsatt att (d) NTP kan fungera som substrat för RNA-och DNA-polymeraser 3, en mängd funktionella grupper bland aminosyrorna 4-13, boronsyror 14,15, nornbornene 16, diamondoid liknande rester 17, sidokedjor för organocatalysis 18, gallsyror 19 och till och med oligonukleotider 20 kan föras in i oligonukleotider.

Införandet av ändringar i nukleinsyror är av intresse för många praktiska tillämpningar, inklusive utvecklingen av antisens-och antigenmedel 42,43, märkning och funktionell märkning av oligonukleotider 41, och i arbetet med att utvidga den genetiska alfabetet 44-46. Kemiska förändringar och funktionella grupper vanligen införes i nukleinsyror genom användning av standard-och automatiserade fastfas-syntesprotokoll.
