Mice have been used as a model for studying many forms of transplantation, including corneal transplantation. We describe in this report a murine model for both acute and late-term corneal transplantation.
Corneal transplantation is the most common form of organ transplantation in the United States with between 45,000 and 55,000 procedures performed each year. While several animal models exist for this procedure and mice are the species that is most commonly used. The reasons for using mice are the relative cost of using this species, the existence of many genetically defined strains that allow for the study of immune responses, and the existence of an extensive array of reagents that can be used to further define responses in this species. This model has been used to define factors in the cornea that are responsible for the relative immune privilege status of this tissue that enables corneal allografts to survive acute rejection in the absence of immunosuppressive therapy. It has also been used to define those factors that are most important in rejection of such allografts. Consequently, much of what we know concerning mechanisms of both corneal allograft acceptance and rejection are due to studies using a murine model of corneal transplantation. In addition to describing a model for acute corneal allograft rejection, we also present for the first time a model of late-term corneal allograft rejection.
Hoornvliestransplantatie is een van de meest succesvolle en voorkomende soorten transplantaties uitgevoerd bij mensen. De redenen waarom deze operatie wordt uitgevoerd zijn een gevolg van letsel, infectieziekten 1, of andere vormen van niet-infectieuze ziekten hoornvlies 2. Uit cijfers van de Eye Bank Association of America geven aan dat meer dan 46.000 werden uitgevoerd in 2011 (zie website: restoresight.org/eye_banks/eye_banks.html). Een indicatie van het succes dat één jaar uitval allogene corneale transplantaten van 10 tot 15% en 5 jaar het succes dan 70% 3-8. Zoals veel studies hebben aangetoond, is het succes van corneale transplantaten direct gerelateerd aan het feit dat het oog een immunologisch begunstigde plaats. Factoren die verantwoordelijk zijn voor de cornea hoedanigheid immune privilege site zijn indien zowel bloed- en lymfevaten in de cornea, een relatieve afwezigheid van antigen presenterende cellen, factoren die door het hoornvlies dat te onderdrukkens immune effector funtions 9-15, lage expressie van MHC antigenen 16, en de expressie van FasL 17-20.
Ondanks deze factoren predisponeren deze enten voor succes, ze ondergaan afwijzing 3-7. Een goed begrip van de mechanismen die deze afwijzing bemiddelen alsook het testen van verschillende therapieën om afstoting te voorkomen, is van cruciaal belang. Daartoe beschrijven wij hier een muizenmodel van hoornvliestransplantatie die in gebruik voor meer dan 20 jaar hoornvliestransplantatie bestuderen in een gecontroleerde experimentele omgeving. Aangezien transplantatie responsen vertonen talrijke factoren daarbij samenwerken die uiteindelijk bepalen of de getransplanteerde weefsel of valt, is het niet mogelijk om het belang van deze factoren in een in vitro model te begrijpen. Bijgevolg zijn studies met behulp van intacte dieren nodig om te bepalen welke factoren belangrijk om ofwel succes of failu zijnre van getransplanteerde weefsel.
Terwijl andere diersoorten zijn gebruikt voor hoornvliestransplantatie studie, het murine model heeft verschillende voordelen ten opzichte van het gebruik van andere soorten. De eerste is het bestaan van vele stammen van muizen die bepaalde transgenen tot expressie of zijn gen-gerichte expressie van specifieke immunologische factoren waarvan de functie in transplantatie beter worden bestudeerd ontbreekt. Daarnaast zijn er vele reagentia (zowel recombinante factoren en antilichamen die factoren neutraliseren) die specifiek zijn voor muizen die niet bestaan voor vele andere diersoorten. Vanwege het bestaan van deze factoren, is dit model uitgebreid gebruikt om relevante factoren die bij acute corneale transplantaatafstoting reacties 15, 17,18,20 -29 identificeren. Bovendien zijn veel van de bij hoornvliestransplantatie factoren bekend functioneel transplantatie van andere weefsels zijn.
Het muizenmodel van hoornvliestransplantatie hier beschreven kan de onderzoeker menselijke hoornvlies allograft afstoting bestuderen in een model dat voorspellend is welke factoren zijn best geassocieerd met zowel afstoting 15,17,18,20, 26-30 en 21-25 acceptatie van corneale allografts.
