Mice have been used as a model for studying many forms of transplantation, including corneal transplantation. We describe in this report a murine model for both acute and late-term corneal transplantation.
Corneal transplantation is the most common form of organ transplantation in the United States with between 45,000 and 55,000 procedures performed each year. While several animal models exist for this procedure and mice are the species that is most commonly used. The reasons for using mice are the relative cost of using this species, the existence of many genetically defined strains that allow for the study of immune responses, and the existence of an extensive array of reagents that can be used to further define responses in this species. This model has been used to define factors in the cornea that are responsible for the relative immune privilege status of this tissue that enables corneal allografts to survive acute rejection in the absence of immunosuppressive therapy. It has also been used to define those factors that are most important in rejection of such allografts. Consequently, much of what we know concerning mechanisms of both corneal allograft acceptance and rejection are due to studies using a murine model of corneal transplantation. In addition to describing a model for acute corneal allograft rejection, we also present for the first time a model of late-term corneal allograft rejection.
Hornhinnetransplantation är en av de mest framgångsrika och vanliga typer av transplantation som utförts på människor. Orsakerna till att denna operation utförs är en följd av skada, infektionssjukdomar 1, eller andra former av icke-infektiös hornhinnans sjukdomar 2. Siffror från Eye Bank Association of America visar att över 46.000 utfördes under 2011 (se hemsida: restoresight.org/eye_banks/eye_banks.html). En indikation på dess framgång är att ett år felfrekvens för allogena hornhinnan transplantat varierar från 10 till 15% och 5 år framgången är över 70% 3-8. Eftersom många studier har visat, är framgången för korneala allogena transplantat direkt relaterad till det faktum att ögat är ett immunologiskt privilegierat ställe. Faktorer som ansvarar för hornhinnor status som en immun privilegium anges med brist på både blod- och lymfkärl i hornhinnan, en relativ brist på antigenpresenterande celler, faktorer som produceras av hornhinnan som suppress immuneffektorfunktioner funktionerna här 9-15, lågt uttryck av MHC-antigener 16, och uttrycket av FasL 17-20.
Men trots dessa faktorer predisponerande dessa transplantat för framgång, de genomgår avslag 3-7. Därför att förstå de mekanismer som förmedlar denna avvisande samt testa olika behandlingar för att förhindra avstötning är av avgörande betydelse. Därför beskriver vi här en musmodell av hornhinnetransplantation som har varit i bruk i över 20 år för att studera hornhinnetransplantation i en kontrollerad experimentell miljö. Sedan transplantationssvar involverar många olika faktorer som arbetar i konsert som kommer ultimata avgör om den transplanterade vävnaden misslyckas eller lyckas, är det inte möjligt att förstå vikten av dessa faktorer i några in vitro-modell. Därför är studier med intakta djur som krävs för att avgöra vilka faktorer som är viktiga för antingen framgång eller failure av transplanterad vävnad.
Medan andra arter av djur har använts för att studera hornhinnetransplantation, har den murina modellen flera fördelar vid jämförelse med användning av andra arter. Den första är att det finns många stammar av möss som uttrycker vissa transgener eller har gen-inriktade sakna uttryck av specifika immunologiska faktorer vars funktion i transplantation bättre kan studeras. Dessutom finns det många reagens (både rekombinanta faktorer och antikroppar som neutraliserar faktorer) som är specifika för möss och som inte finns för många andra djurarter. På grund av förekomsten av dessa faktorer, har denna modell använts i stor utsträckning för att identifiera relevanta faktorer som akuta hornhinnan transplantations svar 15, 17,18,20 -29. Dessutom har många av de faktorer som hornhinnetransplantation är också kända för att vara funktionell i transplantation av andra vävnader.
Den murinmodell av hornhinnetransplantation beskrivs här gör det möjligt för forskaren att studera mänsklig hornhinnan allograftavstötning i en modell som förutsäger vilka faktorer som bäst förknippas med både avslag 15,17,18,20, 26-30 och acceptans 21-25 av hornhinnan allotransplantat.
