Alternative splicing regulation has been shown to contribute to the epithelial-mesenchymal transition (EMT), an essential cellular program in various physiological and pathological processes. Here we describe a method utilizing an inducible EMT model for the detection of alternative splicing during EMT.
Alternative splicing plays a critical role in the epithelial-mesenchymal transition (EMT), an essential cellular program that occurs in various physiological and pathological processes. Here we describe a strategy to detect alternative splicing during EMT using an inducible EMT model by expressing the transcription repressor Twist. EMT is monitored by changes in cell morphology, loss of E-cadherin localization at cell-cell junctions, and the switched expression of EMT markers, such as loss of epithelial markers E-cadherin and γ-catenin and gain of mesenchymal markers N-cadherin and vimentin. Using isoform-specific primer sets, the alternative splicing of interested mRNAs are analyzed by quantitative RT-PCR. The production of corresponding protein isoforms is validated by immunoblotting assays. The method of detecting splice isoforms described here is also suitable for the study of alternative splicing in other biological processes.
La transition épithéliale-mésenchymateuse (TEM) est un programme de développement qui anime la morphogenèse d'organes et le remodelage tissulaire cours de l'embryogenèse. Lorsque anormalement activée, EMT favorise la métastase de la tumeur et des organes fibrose 1,2. Études convaincantes ont décrit l'importance de la régulation de la transcription au cours du processus de EMT, définie par plusieurs facteurs de transcription, tels que Twist, Snail, et ZEB, qui répriment l'expression de la adhérentes protéine de jonction E-cadhérine, entraînant la perte d'un cobble- pierre comme la morphologie et le gain d'un phénotype mésenchymateux fusiforme 3-8 épithéliale. Des études récentes à travers l'analyse de l'ensemble du génome d'ARN ont révélé qu'il existe un groupe de gènes dont l'épissage modèles sont associés soit aux phénotypes épithéliales ou mésenchymateuses 9,10. Le travail de notre laboratoire relié fonctionnellement épissage alternatif et EMT. En étudiant la surface de la molécule d'adhésion cellulaire CD44, CD44, nous avons démontré que alternative épissage est étroitement régulée pendant EMT, et plus important encore, que CD44 épissure isoforme de commutation de causalité contribue à EMT 11.
L'épissage alternatif est un modèle répandu et conservé de la régulation des gènes, car jusqu'à 95% des gènes multi-exons humains sont épissage alternatif 12-14. En générant de multiples produits de protéines à partir d'un seul gène, épissage alternatif constitue un mécanisme essentiel de la diversité des protéines, en ajoutant une autre couche de complexité au génome humain. En tant que tel, la dérégulation de l'épissage alternatif pourrait conduire à des effets biologiques profonds causent des maladies humaines. En effet, l'épissage alternatif aberrant dans les maladies a été documentée depuis plus d'une décennie 15-25, y compris les conclusions récentes mutations dans les gènes codant pour les machines de splicéosome sont généralement trouvés dans les syndromes myélodysplasiques 26-28. Par conséquent, le développement de méthodes fiables pour la détection de la i alternativement épissésoforms est d'une grande importance dans l'étude de divers processus biologiques, y compris EMT.
Ici, nous fournissons un protocole pour détecter des changements dans l'épissage alternatif en utilisant un modèle d'EMT inductible. Les méthodes de conception d'amorces de PCR et la détection des isoformes d'épissage sont adaptés non seulement pour l'étude de l'épissage alternatif pendant EMT, mais également pour l'étude de l'épissage alternatif dans d'autres processus biologiques. Enquête épissage alternatif pendant EMT est impératif afin de mieux comprendre les mécanismes de EMT et les métastases de la tumeur, facilitant ainsi le développement de stratégies efficaces pour traiter les métastases du cancer.
La procédure décrite ici permet la détection de l'épissage alternatif dans un modèle d'EMT inductible. Ainsi, les modifications dynamiques de l'isoforme d'épissage expression peuvent être capturées pendant toute la durée de temps de EMT.
