We beschrijven procedures voor etikettering en genotypering pasgeboren muizen en het genereren van primaire neuronale kweken van hen. De genotypering is snel, efficiënt en betrouwbaar, en maakt geautomatiseerde nucleïnezuur extractie. Dit is vooral handig neonataal dodelijke muizen en culturen die voorafgaande afronding genotyperen vereisen.
Hoge-resolutie analyse van de morfologie en functie van zoogdierneuronen vereist vaak de genotypering van individuele dieren, gevolgd door de analyse van primaire kweken van neuronen. We beschrijven een reeks procedures voor: het labelen van pasgeboren muizen worden gegenotypeerd, snelle genotypering, en tot oprichting van een lage dichtheid culturen van neuronen in de hersenen van deze muizen. Individuele muizen worden gelabeld door tatoeëren, die zorgt voor langdurige identificatie blijvend in de volwassenheid. Genotypering van het beschreven protocol is snel en efficiënt en maakt geautomatiseerde extractie van nucleïnezuur goede betrouwbaarheid. Dit is nuttig in omstandigheden waarin voldoende tijd conventionele genotypering niet beschikbaar, bijvoorbeeld in muizen die lijden neonatale sterfte. Primaire neuronale culturen worden gegenereerd bij een lage dichtheid, die beeldvorming experimenten mogelijk maakt bij een hoge ruimtelijke resolutie. Deze cultuur methode vereist de voorbereiding van gliale feeder lagen voorafgaand aan neuronale plating. De pROTOCOL wordt toegepast in zijn geheel een muismodel van de bewegingsstoornis DYT1 dystonie (AE-Torsina knock-in muizen) en neuronale culturen worden bereid uit de hippocampus, cerebrale cortex en striatum van deze muizen. Dit protocol kan worden toegepast op muizen met andere genetische mutaties, alsmede dieren van andere soorten. Verder kunnen de afzonderlijke componenten van het protocol worden gebruikt voor geïsoleerde deelprojecten. Zo zal dit protocol hebben brede toepassingen, niet alleen in de neurowetenschappen, maar ook in andere gebieden van de biologische en medische wetenschappen.
Knaagdiermodellen van genetische ziekten nuttig bij het vaststellen van de normale fysiologische functies van eiwitten en nucleïnezuren, en de pathofysiologische gevolgen van defecten in deze bewezen. Voorbeelden omvatten muizen deficiënt voor eiwitten die betrokken zijn bij essentiële cellulaire functies, evenals muismodellen van aandoeningen zoals de ziekte van Alzheimer. Toch kunnen bepaalde genetische manipulaties leiden tot neonatale sterfte kort of een paar dagen na de geboorte. In deze gevallen primaire celculturen een belangrijk instrument omdat levende cellen kunnen worden verkregen uit de embryonale of neonatale pups voor sterfte kunnen zij gedurende ten minste enkele weken in vitro, en gedurende deze tijd vroege neuronale ontwikkeling kan worden gevolgd door biochemische, functionele en morfologische experimenten. Voor de primaire kweken, kan het voordelig zijn de neuronen bij lage dichtheid plaat; dit maakt het mogelijk de individuele cellichamen, dendrieten, axonale assen en zenuw beëindiging visualiserennals bij hoge ruimtelijke resolutie. De overleving en differentiatie van neuronen bij lage dichtheid vereist gewoonlijk dat ze uitgeplaat op een gliale feeder layer, samen gekweekt met gliale cellen in afwezigheid van fysiek contact met hen, of gekweekt in medium geconditioneerd door glia 1.
De oprichting van lage dichtheid neuronale culturen gliale feeder lagen kan afhankelijk snelle en betrouwbare genotypering vooraf zijn – binnen een paar uur in tegenstelling tot een paar dagen. Snelheid is vooral belangrijk wanneer de neuronale genotype moet worden aangepast aan die van een gliale feeder layer vooraf voorbereid. Als een praktisch voorbeeld, kan het nodig zijn om te beslissen welke pups waarvan genotype voor gebruik in het genereren culturen, de efficiëntie van een experiment optimaliseren.
Hier laten we zien het werken protocol dat is gebruikt voor een snelle, eenvoudige en betrouwbare muis genotypering in eerdere publicaties 2-6. Muis staarten eneen commercieel verkrijgbare kit gebruikt. Dit protocol omvat één fase extractie van nucleïnezuren uit het weefsel, en vereist noch een nucleïnezuur zuiveringsstap noch het gebruik van een buffer beëindigd ("stop solution). De betrouwbaarheid van deze genotyperingsmethode illustreert waarin de resultaten van een reeks proeven bij verschillen wordt geïntroduceerd ten opzichte van het uitgangsbedrag van de specimens, de leeftijd van de dieren en de lengte van de PCR amplicons. Deze kit biedt de voordelen van geautomatiseerde extractie en betrouwbaarheid.
Omwille van zijn uitgebreid, wordt het gebruik van tatoeage voor identificatie van het genotype muizen lange termijn eveneens aangetoond. Tatoeage wordt bereikt door tatoeage inkt naar de dermis van de huid (onder de epidermis) 7. Een werkwijze wordt beschreven voor het tatoeëren de voetzolen van pasgeboren of 1 dag oude muizen, hoewel tatoeages kunnen worden toegepast op andere delen van het lichaam, zoals staarten en tenen, en Animals van alle leeftijden. Bovendien zullen procedures worden aangetoond plateren en kweken muis neuronen bij een lage dichtheid, gebaseerd op geoptimaliseerde bereiding van verschillende gliale feeder lagen 2,8.
We maken gebruik van een genetische muismodel van de erfelijke neurologische aandoening DYT1 dystonie – een autosomaal-dominante stroming aandoening veroorzaakt door een mutatie in het gen TOR1A (c.904_906delGAG / c.907_909delGAG; p.Glu302del / p.Glu303del) 9. Het gecodeerde eiwit, Torsina, behoort tot de "ATPasen geassocieerd met diverse cellulaire activiteiten" (AAA +) familie van eiwitten, waarvan de leden doorgaans uitgevoerd chaperone-achtige functies, assisteren in: eiwitontvouwende, eiwit-complex demontage, membraantransport en vesikel fusie 10-13. De mutatie leidt tot een in-frame deletie van een codon voor glutaminezuur, en kan leiden tot manifestatie van "early-onset algemene geïsoleerd dystonie '14,15. De padophysiological mechanismen die verantwoordelijk zijn voor deze aandoening blijven slecht begrepen. In een knock-in muismodel, de mutant allel is Tor1a tm2Wtd, hierna genoemd als Tor1a AE. Heterozygote AE-Torsina knock-in muizen zijn levensvatbaar en genetisch bootsen menselijke patiënten met DYT1 dystonie, terwijl homozygote knock-in muizen na de geboorte sterven 16,17, met de latency tot postnatale dood beïnvloed door genetische achtergrond 18. De vroege dood van homozygote knock-in muizen noodzakelijk dat zowel de genotypering van dieren en de instelling van neuronale kweken snel voltooid. Als een ander voorbeeld van genotypering, Tfap2a (transcriptiefactor AP-2α activerende enhancer bindend eiwit 2α) gebruikt. De proteïne gecodeerd door dit gen is belangrijk bij het reguleren meerdere cellulaire processen, zoals proliferatie, differentiatie, overleving en apoptose 19.
The protocol presented here includes procedures for tattooing to label/identify mice, for genotyping mice from tail tips, and for culturing mouse brain neurons at low density. In one round of experiments using 6-8 pups, these procedures typically require ~0.5 hr, ~4 hr and ~2 hr, respectively, at a total of 6-7 hr.
