Primary cilia influence various signaling pathways. The mammalian cochlea is ideal for examining planar cell polarity (PCP) signaling. Cilia dysfunction affects cochlear outgrowth, cellular patterning and hair cell orientation, readouts of PCP. Our goal is to analyze PCP signaling in mouse cochlea via phenotypic analysis, immunohistochemistry and scanning electron microscopy.
In recent years, primary cilia have emerged as key regulators in development and disease by influencing numerous signaling pathways. One of the earliest signaling pathways shown to be associated with ciliary function was the non-canonical Wnt signaling pathway, also referred to as planar cell polarity (PCP) signaling. One of the best places in which to study the effects of planar cell polarity (PCP) signaling during vertebrate development is the mammalian cochlea. PCP signaling disruption in the mouse cochlea disrupts cochlear outgrowth, cellular patterning and hair cell orientation, all of which are affected by cilia dysfunction. The goal of this protocol is to describe the analysis of PCP signaling in the developing mammalian cochlea via phenotypic analysis, immunohistochemistry and scanning electron microscopy. Defects in convergence and extension are manifested as a shortening of the cochlear duct and/or changes in cellular patterning, which can be quantified following dissection from developing mouse mutants. Changes in stereociliary bundle orientation and kinocilia length or positioning can be observed and quantitated using either immunofluorescence or scanning electron microscopy (SEM). A deeper insight into the role of ciliary proteins in cellular signaling pathways and other biological phenomena is crucial for our understanding of cellular and developmental biology, as well as for the development of targeted treatment strategies.
Primaire cilia lange microtubule-gebaseerde aanhangsels die zich uitstrekken vanaf het oppervlak van de meeste zoogdierlijke cellen. Primaire cilia worden vaak verward met beweeglijke cilia, waarvan er altijd meerdere per cel, en waarvan het doel is om de vloeistof over membraanoppervlakken bewegen. Primaire cilia daarentegen vast sensorische rollen en derhalve ook wel sensorische cilia. Eenmaal lang vergeten, dit organel is onlangs herontdekt als gevolg van de associatie met een veelheid van menselijke genetische ziekten 1. Idealiter gepositioneerd als een signalerende organel, heeft de primaire cilium aangetoond talrijke signaalwegen, waarvan vele niet alleen belangrijk in weefsel homeostase en ziekte, maar ook tijdens de ontwikkeling 2 reguleren.
Een van de eerste signaalwegen aangetoond geassocieerd te zijn met cilia disfunctie de Wnt signaling pathway, ook bekend als het vlakke celpolariteit (PCP) pathway <sup > 3. Dit signaalcascade aanvankelijk geïdentificeerd in Drosophila, is van cruciaal belang voor embryogenese; met name convergentie en extensie processen en de juiste oriëntatie van cellen in het vlak van epitheel 4. De sequentiële signalering van een kern set van regulerende eiwitten vertaalt directionele signalen die uiteindelijk leiden tot het cytoskelet herschikkingen en resulteren in de gecoördineerde polarisatie van epitheelcellen in een vlak 5. Het proces van convergentie en uitbreiding is absoluut noodzakelijk voor het cochleair kanaal te verlengen en voor de juiste cellulaire patroonvorming 6. Aangezien dit gereguleerd via activatie van de PCP route, een van de meest opvallende fenotypen van mutanten cochlea PCP is een verkorte cochleair kanaal met ongeorganiseerd sensorische epithelia 7. Evenzo muismutanten die cilia missen, vertonen ook zo'n convergentie en extensie fenotype 8,9, maar precies hoe dit gereguleerd moet nog worden opgehelderd.

Binnen elke chevron-vormige hair bundel, drie rijen stereocilia georganiseerd in een sterk geordende en regelmatige pattern in een trap-achtige manier. Real-microtubule gebaseerde trilharen, kinocilia genoemd, zijn vereist voor de ontwikkeling en oriëntatie van de stereociliary bundels 12. Bij elke haar cel, wordt één kinocilium fysiek verbonden aan de stereocilia bundel, centraal gelegen naast de langste rij stereocilia. De precieze functie van de kinocilium is onduidelijk, en een hypothese is dat de kinocilium 'trekt' de stereocilia in vorm als ze volwassen worden van microvilli 12. In vertebraten, kinocilia in de cochlea alleen aanwezig tijdelijk en intrekken van de haarcellen in muizen voorafgaand aan het begin van het gehoor 11,13,14.Volledig verlies van trilharen in de ontwikkeling van cochlea resulteert in sterk verkorte cochleaire kanalen, verkeerd gevormde of scheef georiënteerde stereociliary bundels, evenals verkeerd gepositioneerd basaalkorrels 8,9. Een functioneel cilium is niet alleen bestaat uit het ciliaire axoneme. Veel eiwitten geassocieerd met ciliafunctie komen in complexen gelokaliseerd op cilia-gerelateerde subdomeinen, zoals de basale lichaam, overgangszone, of ciliaire axoneme 15. De basale instantie, gebaseerd op de moeder centriole van het centrosoom, is een microtubulus organiserende centrum voor microtubules weg van de cilium uitstrekt in het cellichaam en kan intracellulair transport en ciliaire handel reguleren. De ciliaire overgangszone is een ander gebied waar de ciliaire functie is geregeld op het gebied van het organiseren van import en export van trilharen verbindingen 16.
Meerdere studies hebben een verband tussen cilia en niet-canonieke Wnt (PCP signalering) geïdentificeerd, hoewel het precieze mechanisme is onduidelijk 17. Redundantie van ciliaire en PCP genen en de gevoeligheid van de cel polariteit van algemene cellulaire abnormaliteiten, maken het moeilijk om direct een mutatie koppelen aan PCP-specifieke tekorten. Een van de uitlezingen van PCP signalering is de positionering van de basale lichaamstemperatuur en primary cilium, dus scheiden de primaire uit de secundaire gebreken is uitdagend. Sommige studies in zebravis en muis mutanten hebben geen verband tussen de trilharen en Wnt signalering 18-20 voorgesteld. Discrepanties in de gegevens kunnen species, weefsel of tijdelijke-afhankelijke verschillen in ciliaire bijdragen aan Wnt-signalering weerspiegelen. Bovendien zou normaal Wnt reactievermogen blijven, indien basale lichamen blijven functioneren. Een beter inzicht in de rol van ciliaire eiwitten in cellulaire signaalroutes en andere biologische verschijnselen is cruciaal voor ons begrip van cellulaire en ontwikkelingsbiologie en voor de ontwikkeling van gerichte behandelingsstrategieën.
Bij de voorbereiding van cochleaire weefsel voor analyse, er zijn een paar belangrijke punten in gedachten te houden. Ten eerste, de verschillen in genetische achtergrond kan het cochleair fenotype aan te passen, waardoor het noodzakelijk is om te analyseren en te vergelijken alleen nestgenoot controles.
