We present methods for the construction of electrodes to simultaneously record extracellular neural activity and release multiple neuroactive substances at the vicinity of the recording sites in awake mice. This technique allows the detailed analysis of putative local synaptic inputs to the neuron of interest.
Forskjeller i aktiviteten til nevrotransmittere og nevromodulatorer, og dermed ulike nevrale responser, kan finnes mellom bedøvet og våken dyr. Derfor er fremgangsmåter som tillater manipulering av synaptiske systemer i våken dyr nødvendig for å bestemme bidraget av synaptiske innganger til neuronal behandling upåvirket av anestetika. Her presenterer vi metodikk for bygging av elektroder til å samtidig registrere ekstracellulære nevral aktivitet og slipper flere neuroaktive stoffer i nærheten av innspillingsplasser i våken mus. Ved å kombinere disse prosedyrene, utførte vi microiontophoretic injeksjoner av gabazine å selektivt blokkere GABA A-reseptorer i nevronene i dårligere colliculus hode-behersket mus. Gabazine hell modifisert neurale responsegenskaper, slik som frekvensrespons området og stimulus-spesifikk tilpasning. Derfor viser vi at våre metoder er egnet for recording single-enhet aktivitet og for dissekere rollen av spesifikke nevrotransmitterreseptorer i auditiv prosessering.
Den største begrensningen av den beskrevne fremgangsmåte er den forholdsvis korte opptakstiden (~ 3 timer), som bestemmes av nivået av tilvenning til dyret som innspillingen. På den annen side, kan flere innspillingen utføres i det samme dyr. Fordelen med denne teknikken fremfor andre eksperimentelle prosedyrer anvendt for å manipulere nivået av neurotransmisjon eller neuromodulation (slik som systemiske injeksjoner eller bruken av optogenetic modeller), er at medisineffekt er begrenset til de lokale synaptiske innganger til målet neuron. I tillegg er tilpasset fremstilling av elektroder tillater justering av spesifikke parametere i henhold til det neurale strukturen og typen av nervecelle av interesse (slik som tupp motstand for å forbedre signal-til-støy-forholdet av opptak).
Samspillet mellom nevrale eksitasjon og hemming er grunnleggende for behandlingen av en sensorisk informasjon. Det er også kjent at anestesi har en sterk innvirkning på dynamikken i kortikale aktivering og tidsmessig mønster av synaptiske innganger 2,3. For eksempel har det blitt observert at bedøvelsesmidler endre varigheten av visuelt-fremkalte responser i kortikale neuroner 3,4. Videre er forholdet mellom stimulerende og hemmende synaptiske innganger annerledes i bedøvede og våken dyr 4,5, endre både fremkalt og spontane aktivitet prisene 6,7. Ved å måle synaptic conductances, Haider og kolleger fire funnet at hemming matchet eksitasjon i amplitude under narkose, mens under våkenhet, hemming var sterkere enn eksitasjon. Disse funnene be utvikling av eksperimentelle prosedyrer for å studere effekten av spesifikke synaptiske innganger på sensorisk prosessering i våken dyr.
<p class = "jove_content"> Den kontrollerte utstøting av ladede neuroaktive stoffer ved påføring av små strøm injeksjoner (i størrelsesorden nA) har blitt mye brukt for å undersøke bidraget av synaptiske innganger og rollen til antatte cellereseptorer i sensorisk prosessering 8-13 . Denne teknikken, kjent som microiontophoresis, tillater anvendelsen av narkotika i nærheten av den innspilte nervecellen, noe som bidrar til en rask og begrenset effekt. Denne fremgangsmåten er mer egnet for å studere virkningene av lokale neuroaktive stoffer, sammenlignet med den utbredte effekten fremkalt av andre eksperimentelle manipulasjoner slik som systemiske injeksjoner, mikrodialyse eller bruken av optogenetic teknikker. Vanligvis er en piggy-back elektrodekonfigurasjon 14,15 brukes til å samtidig spille inn målet nervecellen og levere neuroaktive stoffer av interesse. Den består av en innspilling elektrode festet til en multibarrel pipette som bærer de neuroaktive stoffer. Modifikasjoner av original prosedyre beskrevet av havey og Caspary 14 er gjennomført. For eksempel kan en wolframelektrode, i stedet for en glass en, brukes til å registrere den 16 nevral aktivitet. Tidligere publiserte metoder for produksjon av wolfram elektroder 17,18 involverer tre generelle trinn: elektrolytisk etsning av wolfram ledning tips, glass isolasjon og justering av spissen eksponering for å møte opptakskravene.En interessant og emergent felt i auditiv nevrovitenskap er studiet av stimulus-spesifikk tilpasning (SSA 19). SSA er en spesifikk nedgang i det nevrale respons på repeterende lyder som ikke generalisere til andre, sjelden presentert lyder. Betydningen av SSA befinner seg i sitt potensielle rolle som en neural mekanisme underliggende avvik deteksjon i hørsels hjernen, samt en mulig neuronal korrelat for sent mismatch negativity del av auditive evoked potensial 20,21. SSA occurs fra IC opp til auditiv cortex 19,22-24. GABA A-formidlet inhibering har vist seg å fungere som en forsterkningsstyremekanisme på SSA 7,16,25, som også er blitt vist å være påvirket av anestesi 26. Her presenterer vi en protokoll som kombinerer de tidligere beskrevne fremgangsmåter for registrering av én enhet aktivitet av IC-nevroner før og under påføringen av en selektiv antagonist av GABA-A-reseptorene i våken mus. Først beskriver vi produksjon av piggy-back elektroder og neste, de kirurgiske og innspillingsmetoder. For å teste effekten av medikamentfrigjøring, vi sammenlignet mottakelig feltet, så vel som nivået av SSA av IC-nevroner før og under microiontophoretic utstøting av gabazine.
Den microiontophoresis av neuroaktive stoffer i våken dyr er en kraftfull teknikk for å undersøke og dissekere rollen av spesifikke synaptiske innganger på aktiviteten til enkeltnerveceller 40,41. Enda viktigere, gjør denne prosedyren fastsettelse av virkningen av nevrotransmittere og nevromodulatorer på nevrale kretser uten eventuelle forstyrrelser av bedøvelse.
