This article describes methods for site-directed spin labeling and reconstitution of pentameric ligand-gated channels for Electron Paramagnetic Resonance studies. This protocol can be adapted for any membrane protein. The reconstitution method described here can also be used for patch-clamp measurements of macroscopic and single-channel currents in a defined lipid system.
Ion channel gating is a stimulus-driven orchestration of protein motions that leads to transitions between closed, open, and desensitized states. Fundamental to these transitions is the intrinsic flexibility of the protein, which is critically modulated by membrane lipid-composition. To better understand the structural basis of channel function, it is necessary to study protein dynamics in a physiological membrane environment. Electron Paramagnetic Resonance (EPR) spectroscopy is an important tool to characterize conformational transitions between functional states. In comparison to NMR and X-ray crystallography, the information obtained from EPR is intrinsically of lower resolution. However, unlike in other techniques, in EPR there is no upper-limit to the molecular weight of the protein, the sample requirements are significantly lower, and more importantly the protein is not constrained by the crystal lattice forces. Therefore, EPR is uniquely suited for studying large protein complexes and proteins in reconstituted systems. In this article, we will discuss general protocols for site-directed spin labeling and membrane reconstitution using a prokaryotic proton-gated pentameric Ligand-Gated Ion Channel (pLGIC) from Gloeobacter violaceus (GLIC) as an example. A combination of steady-state Continuous Wave (CW) and Pulsed (Double Electron Electron Resonance-DEER) EPR approaches will be described that will enable a complete quantitative characterization of channel dynamics.

I synnerhet, har negativa lipider, kolesterol och fosfolipider visat sig modulera funktionen av pLGIC 8-11. Medan den exakta rollen för dessa lipid beståndsdelar i kanalen funktion är okänd, skulle en fullständig molekylär förståelse av grind kräver att studera dessa kanaler i sin ursprungliga miljö. Platsriktad Spin Labeling (SDSL) och elektronspinnresonans (EPR) spektroskopi är de metoder för val för att studera proteindynamik i ombildade system. EPR-spektroskopi är inte begränsad av molekylstorleken (som är NMR) eller optisk egenskap av provet (som är fluorescensspektroskopi), och därigenom tillåter mätningar av fullängds konstruktioner rekonstituerade i nativa lipid förhållanden. Tekniken är extremt känslig och har krav relativt låga prov (i pico-mol range). Båda dessa aspekter gör tekniken väl lämpad för att studera stora membranproteiner som är svåra att uttrycka i över milligramkvantiteter.
Användningen av EPR-spektroskopi i kombination med ställesriktad spinnmärkning har utvecklats av Wayne Hubbell och kollegor, och har anpassats för att studera en mängd av proteintyper. 12-24 EPR uppgifter har använts för att undersöka sekundärstrukturer, förändringar i proteinet konformation, membraninsättningsdjup, och protein-protein / protein-ligandinteraktioner.
Metoden involverar cystein substitution vid positionerna av intresse genom ställesriktad mutagenes. Att säkerställa platsspecifik märkning, är det nödvändigt att ersätta nativa cysteiner med en annan aminosyra (t.ex.., Serin) för att skapa ett cystein-mindre mall. Den överlägset, är den mest populära spin label en tiol-specifik MTSL: (1-oxyl-2,2,5,5-tetrametyl-Δ3-pyrrolin-3-metyl) methanethiosulfonate som fäster till proteinet genom en disulfidbindning bro. Tack vare sin höga specificitet, relativt liten storlek (något större än tryptofan), och flexiheten av länk regionen har här rotations etikett visat sig ha utmärkt reaktivitet även med en nedgrävd cystein. Vidare, för att maximera reaktiviteten, är märkningsreaktionen av proteinet utförs i detergent-solubiliserad form. Efter separation av överskottet av fria spin-label genom storleksuteslutningskromatografi proteinet rekonstitueras in i liposomer eller dubbelskiktsliknande system med definierad lipidkomposition. I allmänhet är cystein mutagenes tolereras väl i de flesta delar av proteinet, och den relativt lilla storleken på spin-sonden orsakar minimal störning för de sekundära och tertiära strukturer. Det märkta-funktionellt protein utsätts sedan för spektroskopiska mätningar, som i huvudsak ger tre huvudsakliga typer av information: 12,14,15,20,22,23,25-27 spinnsond dynamik genom lineshape-analys; tillgängligheten av proben till paramagnetiska relaxationsmedel; och avstånd fördelning. 27 EPR avstånd mäts med två olika metoder. Den första är baserad på kontinuerlig våg (CW) teknik, där spektral breddning till följd av dipolära interaktioner mellan spin-etiketter (i 8-20 ett avståndsintervall).
