We beschrijven een in vitro protocol co-cultuur darm microbioom en darmvlokken voor een langere periode met een menselijke darm-on-a-chip microphysiological systeem.
Hier beschrijven we een protocol om op lange termijn co-cultuur van multi-species menselijke darm microbiome voeren met microengineered darmvlokken in een menselijke darm-on-a-chip microphysiological apparaat. We herhalen het darmlumen-capillaire weefsel-interface in een microfluïdische apparaat, waar de fysiologische mechanische vervormingen en vloeistof shear stroming voortdurend worden toegepast op peristaltiek na te bootsen. In het lumen microkanaal, de menselijke darm epitheel Caco-2-cellen worden gekweekt om een 'kiemvrij' villus epitheel te vormen en te regenereren kleine darmvlokken. Pre-gekweekte microbiële cellen geënt in het lumen kant om een gastheer-microbe ecosysteem stellen. Na microbiële cellen zich aan het apicale oppervlak van de villi, vloeistofstroom en mechanische vervormingen worden hervat aan een stationair micro waarin vers kweekmedium constant toegevoerd en ongebonden bacteriën (evenals bacteriële afval) continu verwijderd produceren. Na uitgebreide co-cultuur from dagen tot weken, worden meerdere microkolonies totaal willekeurig worden geplaatst tussen de villi en zowel microbiële en epitheelcellen levensvatbaar blijven en functionele ten minste één week in kweek. De co-cultuur protocol kan worden aangepast om een veelzijdig platform voor andere gastheer-microbiome ecosystemen welke in diverse menselijke organen, die in vitro onderzoek van de rol van menselijke microbiome orkestreren gezondheid en ziekte kunnen vergemakkelijken.
De menselijke darm herbergt een verbluffend uiteenlopende reeks microbiële species (<1000 soorten) en een enorm aantal microbiële cellen (10 keer meer dan het humane gastheercellen) en genen (100 keer meer dan het humane genoom) 1. Deze menselijke microbiomes spelen een sleutelrol bij het metaboliseren van voedingsstoffen en xenobiotica, reguleren immuunresponsen en onderhouden intestinale homeostase 2. Niet verrassend, gezien deze diverse functies, de commensal darm microbiome moduleert uitgebreid gezondheid en ziekte 3. Zo is het begrijpen van de rol van de darm microbioom en gastheer-microbe interacties zijn van groot belang voor gastro-intestinale (GI) gezondheid te bevorderen en nieuwe therapieën voor darmziekten 4. De bestaande in vitro modellen darm (bijvoorbeeld statische kweken) beperken gastheer-microbiome co-cultuur een korte periode (<1 dag) omdat microbiële cellen overgroeien en compromis darmwandfunctie 5. Surrogaat diermodellen (bv kiem-vrije 6 of genetisch gemanipuleerde muizen 7) zijn ook niet vaak gebruikt om te studeren gastheer-gut microbioom crosstalk omdat de kolonisatie en stabiele handhaving van de menselijke darm microbiome zijn moeilijk.
Om deze uitdagingen te overwinnen, ontwikkelden we onlangs een biomimetische mens "Gut-on-a-Chip" microphysiological systeem (Figuur 1A, links) op de host-darm microbiome interacties die plaatsvinden in de levende menselijke darm 5,8 emuleren. De darm-on-a-chip micro-inrichting bevat twee evenwijdige microkanalen gescheiden door een flexibel, poreus, extracellulaire matrix (ECM) beklede membraan bekleed met humane intestinale epitheliale Caco-2 cellen nabootsen het darmlumen-capillair weefsel-grensvlak (Figuur 1A , rechts) 9. Vacuüm gedreven cyclische ritmische vervormingen veroorzaken fysiologische mechanische vervormingen dat veranderingen normaal induc na te bootsened door peristaltiek (Figuur 1A, rechts). Interessant wanneer Caco-2-cellen worden gekweekt in de darm-on-a-chip voor meer dan 100 uur, vormen zij spontaan driedimensionale (3D) darmvlokken met tight junctions, apicale borstelzomen, proliferatieve cellen beperkt tot basale crypten, slijmproductie, toegenomen drug metaboliseren activiteit (bijvoorbeeld cytochroom P450 3A4, CYP3A4), en verbeterde glucose heropname 8. In deze "kiemvrij 'micromilieu, was het mogelijk om co-kweek het probioticum Lactobacillus rhamnosus GG of therapeutische vorming van een probiotische bacteriekweek mengsel met menselijke epitheelcellen gedurende twee weken 5,10.
In deze studie beschrijven we het gedetailleerd protocol host-gut microbiome co-kweek uitgevoerd in de darm-on-a-chip apparaat voor een langere periode. Daarnaast bespreken we kritieke problemen en mogelijke uitdagingen in aanmerking komen voor een brede toepassing van deze host-microbiome co-cultuur pROTOCOL.
Inzicht in gastheer-microbiome interacties is van cruciaal belang voor het bevorderen van de geneeskunde; Echter, traditionele celkweekmodellen uitgevoerd in een plastic schaal of een statische wells plaat ondersteunt de stabiele co-kweek van menselijke intestinale cellen met levende darmorganismen langer dan 1-2 dagen vanwege microbiële cellen meestal overgroeien de zoogdiercellen in vitro.
