This protocol provides step-by-step guidelines for setting up competitive mouse bone marrow transplant experiments to study hematopoietic stem/progenitor cell function without prior purification of stem cells by cell sorting.
The gold standard definition of a hematopoietic stem cell (HSC) is a cell that when transferred into an irradiated recipient will have the ability to reestablish blood cell production for the lifespan of the recipient. This protocol explains how to set up a functional assay to compare the HSC capacities of two different populations of cells, such as bone marrow from mice of two different genotypes, and how to analyze the recipient mice by flow cytometry. The protocol uses HSC equivalents rather than cell sorting for standardization and discusses the advantages and disadvantages of both approaches. We further discuss different variations to the basic protocol, including serial transplants, limiting dilution assays, homing assays and non-competitive transplants, including the advantages and preferred uses of these varied approaches. These assays are central for the study of HSC function and could be used not only for the investigation of fundamental HSC intrinsic aspects of biology but also for the development of preclinical assays for bone marrow transplant and HSC expansion in culture.
Hematopoes är en regenerativ process som säkerställer påfyllning av blodkroppar som har gått förlorade på grund av skada, strålning och celldöd. Denna process säkerställs genom hematopoetiska stamceller (HSC) som till stor del bor i den vuxna benmärgen. Dessutom kan hematopoietiska stamceller användas för terapeutiska ändamål i autoimmuna sjukdomar, hematologiska maligniteter och immunbrister 1. Det finns således ett behov av att bättre förstå de mekanismer som reglerar HSC funktion, inklusive deras proliferativa expansion och deras förmåga att nå och ympas mottagaren benmärg efter transplantation. Även nyare studier har rapporterat flera cellytmarkörer, inklusive SLAM familjemedlemmar CD150 och CD48, att prospektivt berika vuxna HSCs och foster HSCs till ca 50% renhet 2-4, guldstandardmått för funktionell HSCs fortfarande ett in vivo återinplantering analys för att bestämma deras förmåga att återupprätta blod cell produktion i en bestrålad värd 5.
In vivo klonåterpopulation analys utvecklades ursprungligen av Till och McCulloch 6 och har sedan dess förfinats och utökats. Ursprungligen angiven, HSCs säkerställa produktionen livslångt blodkroppar genom självförnyelse och differentiering. Överföringen av HSC: er i en bestrålad mottagare kan alltså oss att bedöma deras förmåga att differentiera genom analys av de olika blodcellinjer (T-lymfocyter, B-lymfocyter, granulocyter, monocyter) och deras förmåga till självförnyelse via seriell transplantation. Analysen skulle vanligtvis innebära en jämförelse av funktionaliteten och / eller kvantitet av två populationer av HSC: er, t ex celler som kommer från två möss av olika genotyper eller celler som har behandlats eller inte behandlats med olika faktorer som kan påverka underhåll eller expansion av HSC: er i kultur. Donator chimerismen eller bidrag överförd donator HSCs to blodkroppar kan sedan bestämmas genom flödescytometrianalys i det perifera blodet och benmärgen med hjälp av markörer på cellytan eller andra metoder som kommer att skilja donatorceller från mottagaren, eller värd. De mest använda markörerna är säkerligen två alleler för genen Ptprc eller CD45 leukocytantigen 7 som vi har valt för exemplen nedan.
Den klonala återinsättning analysen kan vara antingen konkurrerande eller icke-konkurrenskraftiga. I en icke-konkurrenskraftig miljö, är kontroll- och test HSCs överförs till separata mottagande möss och utfallet för varje celltyp kommer att vara oberoende av den andra. På en konkurrensutsatt miljö, är funktionen av både test- och kontroll HSCs mätt mot en population av konkurrent HSCs. Protokollet som beskrivs här använder konkurrenskraftig miljö men kan också anpassas för icke-konkurrenssituationer. Båda tillvägagångssätten har sina fördelar och begränsningar, och vi kommer att jämföra dem i detalj idiskussion. Vi beskriver också olika metoder för att säkerställa rättvisa i antalet transplanterade HSCs, förklarar hur man anpassar analysen för kvantifiering av HSC: er genom begränsande utspädning analys (LDA), och ger exempel på både lyckade och misslyckade transplantationer för tolkning av resultaten.
Protokollet som beskrivs här är utformad för att utvärdera den relativa lämplighet donator (test) HSCs mot kända konkurrenten HSCs. Den konkurrenssituation ökar den relativa känsligheten hos analysen (mer sannolikt att upptäcka måttliga sänkningar av stamcells kondition) och ger en intern teknisk kontroll för effekten av strålning och injektion. Det bör dock inte användas som ett absolut mått på HSC kondition, en minskning av konkurrens beredning innebär inte automatiskt att HSCs inte skulle klara sig …
