We beschrijven het gebruik van optogenetics en elektrofysiologische opnames voor selectieve manipulaties van hippocampal theta oscillaties (5-10 Hz) in gedragend muizen. De werkzaamheid van de entrainment ritme wordt gecontroleerd met behulp van lokale veld potentieel. Een combinatie van opto- en Farmacogenetische inhibitie adressen de efferent uitlezing van hippocampal synchronisatie.
Uitgebreide gegevens over relaties van neuraal netwerk oscillaties naar gedrag en organisatie van neuronale kwijting over hersengebieden vragen om nieuwe instrumenten te selectief manipuleren ritmes van de hersenen. Hier beschrijven we een aanpak projectie-specifieke optogenetics te combineren met de extracellulaire electrofysiologie voor HiFi-besturingselement van hippocampal theta oscillaties (5-10 Hz) in gedragend muizen. De specificiteit van de entrainment optogenetic wordt bereikt door gericht op channelrhodopsin-2 (ChR2) voor de GABAergic bevolking van mediale septal cellen, cruciaal betrokken in de generatie van hippocampal theta oscillaties, en een lokale gesynchroniseerd activering van een subset van remmende septal afferents in de hippocampus. De doeltreffendheid van het optogenetic ritme besturingselement wordt gecontroleerd door een gelijktijdige controle van het lokale veld potentiële (LFP) over lamina van de CA1-gebied en/of neuronale kwijting. Laten we zien met behulp van deze gemakkelijk uitvoerbaar voorbereiding werkzaamheid van verschillende optogenetic stimulatie protocollen voor inductie van theta oscillaties en voor het manipuleren van de frequentie en de regelmaat. Tot slot, een combinatie van de theta rhythm control met remming van de projectie-specifieke adressen de uitlezing van bepaalde aspecten van de hippocampal synchronisatie door efferent regio’s.
Neuronale activiteit bij zoogdieren wordt gecoördineerd door netwerk trillingen, die het bijstaan van informatieoverdracht binnen en tussen hersenen regio’s1,2,3,4. Ritmes van de hersenen omvatten oscillaties variërend van erg traag ( 200 Hz) frequenties. Een grote hoeveelheid bewijs ondersteunt betrokkenheid van netwerk oscillaties in diverse hersenfuncties, inclusief cognitie5,6,7,8,9,10 , aangeboren gedrag11,12 , alsmede neuropsychiatrische aandoeningen zoals de ziekte van Parkinson en epilepsie13,14,15. Selectieve en stoffelijk nauwkeurige methoden voor experimentele manipulatie van netwerk oscillaties zijn daarom essentieel voor de ontwikkeling van fysiologisch plausibel modellen van synchronisatie en voor de vaststelling van causale verbanden met gedrag.
Netwerksynchronisatie is bemiddeld door diverse biologische substraten en processen, variërend van moleculaire identiteit van ionenkanalen en hun kinetiek tot neuromodulatie van prikkelbaarheid en verbinding met het netwerk. Het biologische ontwerp van ritme generatoren16 is gebleken voor vele ritmes van de hersenen, verschillende aspecten van welke (b.v., frequentie en amplitude) vaak zijn veroorzaakt door de dynamiek van verschillende celtypes en netwerken. Bijvoorbeeld, zijn remmende interneuronen gericht op de waarin van de belangrijkste cellen de belangrijkste spelers over frequentiebanden en hersenen regio’s17,18, met inbegrip van theta19,20, gamma20 , 21, en rimpel (140-200 Hz)22 oscillaties. Op zijn beurt wordt fase synchronisatie van verre cellen gewaarborgd door het robuuste feed-forward signalering van piramidale cellen, die het afvuren van interneuronen reset. Een cruciale parameter van trillingen, de grootte van de gesynchroniseerde neuronale bevolking, is nauw verwant aan de amplitude van de gemeten LFP trilling en, ten minste voor snelle schommelingen, afhankelijk van het excitatory station naar interneuronen2. Daarentegen langzamer oscillaties, zoals delta en theta ritmes, worden gegenereerd door lange-afstands terugkerende lussen, gevormd door cortico-thalamus23,24 en hippocampal-mediale septal projecties25, 26,27, respectievelijk. Oscillaties in dergelijke schakelingen zijn teweeggebracht door interacties van signaal doorgeven vertragingen, prikkelbaar reacties en hun frequentie voorkeur in deelnemende cellen28,29,30, 31 , 32. remmende projecties van GABAergic parvalbumin (PV)-positieve cellen van het mediale septum (MS) te interneuronen in de hippocampus25,33, de parahippocampal regio’s en de Entorinale cortex26 essentieel voor de generatie van theta oscillaties in de mediale temporale kwab. Dus, fysiologische mechanismen van netwerk oscillaties en neuronale synchronisatie kunnen worden gemanipuleerd met behulp van optogenetics met een real-time precisie.
Cel type-specifieke optogenetic manipulaties zijn aangevraagd door studies van de hippocampal en corticale oscillaties in vitro34,35,,36,,37,38 en in vivo30,39,40,41,42,43,44,45, met inbegrip van functionele onderzoeken van gamma5,12,36,,46,47,48,49,50, 51,52 en rimpel oscillaties40,53,,54 en slaap spindles55,,56. We hebben onlangs een Cre-afhankelijke ChR2-virus in de lidstaat, een belangrijke regio voor de generatie van het hippocampal theta ritme, van PV-Cre muizen. Met behulp van deze voorbereiding, werden kenmerken van de hippocampal theta oscillaties (frequentie en temporele stabiliteit) gecontroleerd door optogenetic stimulatie van remmende prognoses van de lidstaten in de hippocampus11. Bovendien opgeroepen theta-frequentie optogenetic stimulatie van remmende septo-hippocampal projecties theta ritme tijdens wakker immobiliteit. De optogenetically entrained theta ritme weergegeven eigenschappen van spontane theta oscillaties in de muis LFP en neuronale activiteitenniveaus.
Belangrijkste kenmerken van dit protocol zijn: (1) gebruik van een remmende traject dat is fysiologisch essentieel voor spontane theta oscillaties terwijl het vermijden van aspecifieke effecten op hippocampal prikkelbaarheid; (2) axonale, dat wil zeggen, projectie-specifieke stimulatie te minimaliseren een directe invloed op niet-hippocampal MS-efferents; (3) lokale theta-ritmische lichte stimulatie, zorgen voor een minimale directe inmenging met theta-ritmische septo-hippocampal dynamiek en een wereldwijde bilaterale meevoeren van theta oscillaties; (4) parametrische controle theta oscillaties frequentie en regelmatigheid; en (5) kwantificering van entrainment trouw met hoge temporele resolutie met behulp van LFP om kwantitatieve causaliteit analyse in het gedrag van dieren. Aangezien deze voorbereiding in wezen speelt in op een bekende rol van de septo-hippocampal disinhibitie in theta generatie25,30, hierdoor robuuste controle over verschillende parameters van theta oscillaties in gedragend muizen. Studies waar andere minder onderzochte trajecten en celtypes van het septo-hippocampal circuits waren gemanipuleerd38,39,47,49,50,51 , 52 , 53 , 54 , 55 , 56 , 57 , 58 onthullen verdere mechanismen van de theta-ritme.
Hier voorgesteld hebben we een breed toegankelijke methodologie later te ontlokken hippocampal theta oscillaties in het dier gedraagt. Deze aanpak kan zinvol zijn voor studies van theta ritme van functies in de informatieverwerking en gedrag. Kritieke aspecten van deze methode zijn: (1) de keuze van de opsin en doelgerichtheid van ChR2 aan de axonen van MS cellen in de hippocampus, (2) robuuste optische en elektrische kenmerken van geïmplanteerde optische vezel-draads matrix assemblages om continu stimulatie en de LFP o…
