Wir beschreiben die Verwendung der optogenetik und elektrophysiologische Aufnahmen für gezielte Manipulationen der hippocampal Theta Schwingungen (5 bis 10 Hz) im Verhalten Mäuse. Die Wirksamkeit von Rhythmus mitreißen wird mit lokalen Feldes Potenzial überwacht. Eine Kombination von Opto und pharmakogenetischen Hemmung befasst sich mit das ableitenden Auslesen der hippocampalen Synchronisation.
Umfangreiche Daten über Beziehungen des neuronalen Netzes Schwingungen zu Verhalten und Organisation der neuronalen Entlastung über Gehirnregionen fordern neue Tools zum Gehirn Rhythmen gezielt zu manipulieren. Hier beschreiben wir einen Ansatz Projektion-spezifische optogenetik mit extrazellulären Elektrophysiologie für High-Fidelity-Kontrolle der hippocampalen Theta Schwingungen (5 bis 10 Hz) im Verhalten Mäuse kombinieren. Die Besonderheit des optogenetische Entrainment wird erreicht durch gezielte Channelrhodopsin-2 (ChR2) der Gabaergen Bevölkerung von medialen septal Zellen, entscheidend in die Generation der hippocampalen Theta Schwingungen, und eine lokale Aktivierung synchronisiert eine Teilmenge von hemmenden septal afferenzen im Hippocampus. Die Wirksamkeit des Kontrollstreifens optogenetische Rhythmus wird durch eine gleichzeitige Überwachung des lokalen Feldes potenzielle (LFP) über Lamina der CA1-Region und/oder neuronale Entladung überprüft. Anhand dieser leicht umsetzbare Vorbereitung zeigen wir Wirksamkeit verschiedener optogenetische Stimulation-Protokolle für die Induktion von Theta-Schwingungen und für die Manipulation von deren Häufigkeit und Regelmäßigkeit. Schließlich spricht eine Kombination der Theta-Rhythmus Steuerung mit Projektion-spezifischen Hemmung das Auslesen einzelner Aspekte der hippocampalen Synchronisation von ableitenden Regionen.
Neuronaler Aktivität bei Säugetieren wird vom Netzwerk Schwingungen, koordiniert die Informationsübertragung innerhalb und zwischen Gehirn Regionen1,2,3,4zu unterstützen. Gehirn Rhythmen sind Schwingungen von sehr langsam ( 200 Hz) Frequenzen. Ein großer Körper des Beweises unterstützt Beteiligung der Netzwerk-Schwingungen in unterschiedlichen Gehirnfunktionen, einschließlich Kognition5,6,7,8,9,10 , angeborene Verhaltensweisen11,12 , sowie neuropsychiatrischen Erkrankungen wie Parkinson und Epilepsie13,14,15. Selektive und zeitlich präzise Methoden zur experimentellen Manipulation Netzwerk Schwingungen sind daher unabdingbar für die Entwicklung der physiologisch plausible Modelle der Synchronisation und für die kausale Verbindung mit Verhalten.
Netzwerksynchronisation wird durch vielfältige biologische Substrate und Prozesse, Molekulare Identität von Ionenkanälen und deren Kinetik bis hin zu Neuromodulation der Erregbarkeit und Netzwerkkonnektivität vermittelt. Die biologische Gestaltung der Rhythmus, die Generatoren, die16 offenbart worden ist für viele Gehirn Rhythmen, unterschiedliche Aspekte von denen (z.B.Frequenz, Amplitude) oft sind herbeigeführt durch Dynamik der unterschiedlichen Zelltypen und Netzwerke. Zum Beispiel sind hemmende Interneurone, die gezielt die Somata der wichtigsten Zellen die wichtigsten Akteure in Frequenzbänder und Gehirn Regionen17,18, einschließlich Theta19,20, Gamma20 , 21und22 Schwingungen Welligkeit (140 – 200 Hz). Im Gegenzug wird Phase Synchronisation von weit entfernten Zellen durch robuste feedforward Signalisierung von Pyramidenzellen, die den Abschuss von Interneuronen zurückgesetzt gewährleistet. Ein entscheidender Parameter der Schwingungen, die synchronisierte neuronale Bevölkerungszahl ist eng verwandt mit der gemessenen LFP Oszillation Amplitude und mindestens für schnelle Schwingungen hängt der exzitatorischen befahren Interneuronen2. Im Gegensatz dazu langsamere Schwingungen, wie Delta und Theta Rhythmen entstehen durch weiträumige reentrant Loops, gebildet von Cortico-thalamische23,24 und hippocampal Medial septal Projektionen25, 26,27, beziehungsweise. Schwingungen in solche Schaltungen sind durch Wechselwirkungen zwischen Ausbreitung signalverzögerungen, erregbar Antworten und ihre Häufigkeit Vorliebe in teilnehmenden Zellen28,29,30, herbeigeführt 31 , 32. hemmende Projektionen von Gabaergen Parvalbumin (PV)-positiven Zellen der medialen Nasenscheidewand (MS) zu Interneuronen in den Hippocampus25,33, Parahippocampal Regionen und entorhinalen Kortex26 wesentlich für die Erzeugung von Theta-Schwingungen in den medialen Temporallappen. So können physiologische Mechanismen der Netzwerk-Schwingungen und neuronale Synchronisation mit optogenetik mit einer Echtzeit-Präzision manipuliert werden.
Zelle typspezifischen optogenetische Manipulationen wurden bei Untersuchungen der Hippokampus und kortikalen Schwingungen in-vitro-34,35,36,37,38 und angewendet in-vivo30,39,40,41,42,43,44,45, einschließlich funktioneller Untersuchungen von Gamma5,12,36,46,47,48,49,50, 51,52 und welligkeit Schwingungen40,53,54 und Schlaf Spindeln55,56. Vor kurzem haben wir einen Cre-abhängige ChR2-Virus in den Mitgliedstaaten eine Schlüsselregion für die Generation des Hippokampus Theta-Rhythmus von PV-Cre-Mäusen. Mit Hilfe dieses Präparat, waren optogenetische Stimulation der hemmende Projektionen der MS in der Hippocampus-11Funktionen der hippocampalen Theta Schwingungen (Häufigkeit und zeitliche Stabilität) gesteuert. Darüber hinaus evoziert Theta-Frequenz optogenetische Stimulation der hemmenden Septo-hippocampal Projektionen Theta-Rhythmus während wach Immobilität. Die Optogenetically mitgerissen Theta-Rhythmus angezeigt Eigenschaften des spontanen Theta Schwingungen in der Maus bei LFP und neuronale Aktivität.
Wichtige Merkmale dieses Protokolls sind: (1) Nutzung von einem hemmenden Weg, das physiologisch wichtig für spontane Theta Schwingungen ist unter Vermeidung unspezifische Effekte auf hippocampal Erregbarkeit; (2) axonalen, d. h., Projektion-spezifische Stimulation, einen direkten Einfluss auf nicht-hippocampal MS Efferents; zu minimieren (3) lokale Theta-rhythmische Lichtstimulation, gewährleistet einen minimalen direkten Eingriff in Theta-rhythmische Septo-hippocampal Dynamik und einer globalen bilateralen Entrainment Theta Schwingungen; (4) parametrische Kontrolle der Theta-Schwingungen-Häufigkeit und Regelmäßigkeit; und (5) Quantifizierung der Entrainment Treue mit hoher zeitlicher Auflösung mit LFP um quantitative Kausalität Analyse im Verhalten der Tiere zu ermöglichen. Da dieses Präparat im Wesentlichen auf eine bekannte Rolle der Septo-hippocampal Enthemmung in Theta Generation25,30nutzt, ermöglicht es robuste Kontrolle über verschiedene Parameter der Theta-Schwingungen im Verhalten Mäuse. Wo andere weniger untersuchten Signalwege und Zelltypen der Septo-hippocampal Schaltung wurden Studien manipuliert38,39,47,49,50,51 , 52 , 53 , 54 , 55 , 56 , 57 , 58 offenbaren weitere Mechanismen des Theta-Rhythmus.
Hier präsentierten wir eine allgemein zugängliche Methode zur mitzureißen und hippocampal Theta Schwingungen im Verhalten Tier zu entlocken. Diese Methode ist nützlich für die Studien der Theta-Rhythmus Funktionen in der Informationsverarbeitung und Verhalten.
