Die Anzahl der neue Biomaterialien für die Reparatur von großen Knochenläsionen entwickelt wird laufend erweitert, mit dem Ziel zu verbessern, Knochenheilung und Komplikationen im Zusammenhang mit Knochentransplantation überwinden. Hier präsentieren wir Ihnen eine multidisziplinäre Strategie zu Testzwecken vorklinischen Biokompatibilität von Biomaterialien für die Knochen-Reparatur.
Großen Pseudarthrose Knochenbrüche sind eine große Herausforderung in der orthopädischen Chirurgie. Obwohl Auto und allogene Knochentransplantate ausgezeichnet zur Heilung solcher Läsionen sind, gibt es mögliche Komplikationen mit deren Einsatz. So entwickeln die Materialwissenschaftler synthetische, biokompatible Biomaterialien um diese Probleme zu überwinden. In dieser Studie stellen wir eine multidisziplinäre Plattform für die Bewertung der Biomaterialien für Knochen-Reparatur. Wir kombinieren Fachwissen aus Knochenbiologie und Immunologie, einschließlich in-vitro- Osteoklasten (OC) und Osteoblasten (OB) Assays und in Vivo Mausmodelle der Knochen-Reparatur, Immunogenität und Allergenität Plattform zu entwickeln. Wir zeigen, wie die Experimente durchführen, Zusammenfassung der Ergebnisse und Bericht über Biomaterial Biokompatibilität. Insbesondere haben wir OB Lebensfähigkeit, Differenzierung, und Mineralisierung und OC Lebensfähigkeit und Differenzierung im Rahmen der β-Tricalcium Phosphat (β-TCP) Festplatten getestet. Wir testeten ebenfalls einen β-TCP/Kollagen (β-TCP/C)-Schaum ist ein handelsübliches Material klinisch für Knochen-Reparatur in eine kritische Größe calvarial Knochen-defekt-Maus-Modell verwendet, um festzustellen, die Auswirkungen auf die frühe Phase der Knochenheilung. In parallelen Experimenten haben wir immun und allergische Reaktionen bei Mäusen ausgewertet. Knochen-Reparatur ist ein komplexer Prozess, der mit Hämatombildung, Entzündung beginnt, Kallus, Bildung und dann Umbau1,2. Knochenregeneration Potenzial ist jedoch beschränkt auf die Größe der Knochenbruch1,3. Zum Beispiel große Knochenbrüche verursacht durch ein Trauma, Krebs oder Osteoporose können nicht heilen und Pseudarthrose Knochenbrüche bezeichnet werden. Diese Knochenveränderungen erfordern oft Behandlung gesunde physiologische Knochen Reparatur und Regeneration zu fördern. Derzeit Autotransplantation und Allograft Knochen-Transplantation ist der Ansatz der Wahl4 mit 2,2 Millionen Knochen Ersatzverfahren jährlich5. Obwohl diese Verfahren eine hohe Erfolgsquote haben, möglicherweise gibt es Komplikationen, z. B. begrenzte Verfügbarkeit von Knochen, Infektion, Spender Website Morbidität und Ablehnung4. Neue Alternativen für das Bone Tissue Engineering werden gesucht, um diese Herausforderungen zu bewältigen.
Das Design von Biomaterialien, basierend auf natürlichen oder synthetischen Polymeren, Biokeramik oder Metalle in Verbindung mit Zellen und bioaktiven Moleküle ist auf dem Aufstieg6. Unser gegenwärtige Verständnis der physiologischen Knochen heilen und Heilung im Zusammenhang mit Biomaterialien hängt von mehrere Faktoren wie mechanische Eigenschaften und mehrere lokale und systemische Faktoren, einschließlich der Zellen aus dem Verkehr und Frakturstelle7 ,8,9. Biomaterialien für Knochenregeneration Osteogenicity und Osseointegration10 fördern und sind im Idealfall biokompatibel, biologisch abbaubar und poröse (Förderung der Zellwanderung, Sauerstoff und Nährstoffe). Sie müssen auch stark genug zur Unterstützung der Frakturstelle, Schmerzen zu lindern. Darüber hinaus sind entzündliche Faktoren erforderlich, um den Heilungsprozess einzuleiten. Jedoch wenn das Biomaterial übermäßige Entzündungen und allergische Reaktionen induziert, könnte dies zu begrenzen oder hemmen Knochenheilung11,12. So ist ein interdisziplinärer Ansatz notwendig, Biomaterialien entwickelt für Knochen-Reparatur zu bewerten.
In dieser Studie stellen wir eine prä-klinischen Bewertung der repräsentativen Materialien (1) Orthovita Vitoss, den Schaum, der im Handel erhältlichen Spongiosa Graft Ersatz bestehend aus Tricalcium Phosphat ist bestehend aus nanometergroßen reines β-tricalcium Phosphat (β-TCP) Teilchen und bovinen Kollagen Typ1 (C) (β-TCP/C-Schaum) und 2) β-TCP-Festplatten. Hier wir illustrieren Biokompatibilität testen diese Biomaterialien mit primären Osteoblasten (OB) und Osteoklasten (OC) assays, eine in Vivo Modell von Knochen zu reparieren, eine immunologische Bewertung bestehend aus in-vitro- Vermehrung der T-Lymphozyten und Produktion von Zytokinen und in Vivo Immunogenität und Allergenität, wie bereits berichtet13.
