Le nombre de nouveaux biomatériaux conçus pour réparer les lésions osseuses importantes est en expansion continue dans le but d’améliorer la guérison osseuse et surmonter les complications associées à la transplantation osseuse. Nous présentons ici une stratégie multidisciplinaire pour les tests de biocompatibilité préclinique de biomatériaux pour la réparation osseuse.
Fractures osseuses importantes de non syndiqués sont un défi en chirurgie orthopédique. Bien qu’auto – et greffe osseuse allogénique est excellente pour la guérison des lésions, il y a des complications potentielles à leur utilisation. Ainsi, matériels scientifiques mettent au point des biomatériaux synthétiques, biocompatibles pour résoudre ces problèmes. Dans cette étude, nous présentons une plateforme multidisciplinaire pour l’évaluation de biomatériaux pour la réparation osseuse. Nous avons combiné expertise biologie osseuse et d’immunologie de développer une plate-forme y compris in vitro ostéoclastes (OC) et dosages des ostéoblastes (OB) et in vivo de modèles murins de la réparation osseuse, immunogénicité et d’allergénicité. Nous montrent comment réaliser les expériences, résument les résultats et rapport sur la biocompatibilité de biomatériau. En particulier, nous avons testé la viabilité de l’OB, la différenciation et minéralisation et viabilité OC et différenciation dans le contexte des disques de β-tricalcium phosphate (β-TCP). Nous avons aussi testé une mousse (β-TCP/C) de β-TCP/collagène qui est un matériau disponible dans le commerce, utilisé en clinique pour la réparation osseuse dans un modèle murin de taille critique de substance osseuse défaut pour déterminer les effets sur la première phase de cicatrisation osseuse. Dans des expériences parallèles, nous avons évalué les réponses immunitaires et allergiques chez les souris. Notre approche génère un profil de compatibilité biologique d’un biomatériau osseux avec une gamme de paramètres nécessaires pour prédire la biocompatibilité des biomatériaux utilisés pour la guérison osseuse et la réparation chez les patients.
La réparation osseuse est un processus complexe qui commence par la formation d’un hématome, une inflammation, Cal formation et remodelage puis1,2. Toutefois, le potentiel de régénération osseuse est limité à la taille de l’OS fracture1,3. Par exemple, les fractures des os grand causées par un traumatisme, le cancer ou l’ostéoporose ne peuvent pas guérir et sont appelées fractures osseuses non syndiqués. Ces lésions osseuses nécessitent souvent des traitements pour promouvoir la régénération et la réparation osseuse physiologique sain. Actuellement, autogreffe et allogreffe osseuse transplantation est l’approche de choix4 avec les procédures de remplacement des OS 2,2 millions par an5. Bien que ces procédures ont un taux de réussite élevé, il peut y avoir des complications, par exemple, la disponibilité limitée des os, infection, morbidité du site donneur et rejet4. De nouvelles alternatives pour l’ingénierie tissulaire osseuse sont recherchés pour relever ces défis.
La conception des biomatériaux à base de polymères naturels ou synthétiques, des biocéramiques ou des métaux en combinaison avec les cellules et molécules bioactives est sur l’ élévation6. Notre compréhension actuelle des os physiologiques de guérison et de guérison dans le contexte des biomatériaux dépend de divers facteurs tels que les propriétés mécaniques et de multiples facteurs les et systémiques, y compris les cellules de la circulation et le site de fracture7 ,8,9. Biomatériaux pour la régénération osseuse vise à promouvoir les osteogenicity et ostéointégration10 et est idéalement biocompatible et biodégradable poreux (promotion de nutriments, l’oxygène et la migration cellulaire). Dans cette étude, nous présentons une évaluation préclinique de matières représentatives, 1) Orthovita Vitoss mousse qui est un substitut de greffe disponible dans le commerce d’os spongieux consistant en phosphate tricalcique composée de taille nanométrique pur β tricalcique les particules de phosphate (β-TCP) et collagène bovin de Type 1 (C) (β-TCP/C mousse) et 2) des disques de β-TCP. Ici, nous illustrons test de biocompatibilité de ces biomatériaux à l’aide des ostéoblastes primaires (OB) et dosages des ostéoclastes (OC), un modèle in vivo de la réparation osseuse, une évaluation immunologique comprenant in vitro la prolifération des lymphocytes T et la production de cytokines et in vivo de l’immunogénicité et l’allergénicité, comme indiqué précédemment13.
Ici, nous montrons une approche multidisciplinaire pour l’évaluation préclinique de la biocompatibilité des biomatériaux représentatif mis au point pour la réparation et la régénération osseuse.
