O número de novos biomateriais, projetado para a reparação de lesões ósseas grande está expandindo continuamente com o objectivo de reforçar a consolidação óssea e superar as complicações associadas com o transplante de osso. Aqui, apresentamos uma estratégia multidisciplinar para testes de pré-clínica biocompatibilidade de biomateriais para reparo ósseo.
Fraturas de grandes não-união óssea são um desafio significativo em cirurgia ortopédica. Embora autoe enxertos ósseos alogênico são excelentes para a cura dessas lesões, existem complicações potenciais com a sua utilização. Assim, materiais cientistas estão desenvolvendo biomateriais sintéticos, biocompatíveis para superar esses problemas. Neste estudo, apresentamos uma plataforma multidisciplinar de avaliação de biomateriais para reparo ósseo. Nós combinamos conhecimentos de biologia óssea e Imunologia para desenvolver uma plataforma incluindo em vitro osteoclast (OC) e ensaios de osteoblastos (OB) e na vivo modelos de rato de reparação óssea, imunogenicidade e alergenicidade. Vamos demonstrar como realizar os experimentos, um resumo dos resultados e relatório sobre a biocompatibilidade de biomateriais. Em particular, nós testamos a viabilidade de OB, diferenciação, mineralização e viabilidade OC e diferenciação no contexto de discos de β-tricálcio fosfato (β-TCP). Nós também testamos uma β-TCP/colágeno (β-TCP/C) espuma, que é um material disponível comercialmente usado clinicamente para reparo ósseo em um modelo de mouse de defeito de tamanho crítico raspagem óssea para determinar os efeitos na fase inicial de consolidação óssea. Em experiências paralelas, avaliaram-se respostas imunes e alérgicas em ratos. Nossa abordagem gera um perfil de compatibilidade biológica de um biomaterial osso com uma gama de parâmetros necessários para prever a biocompatibilidade de biomateriais utilizados para consolidação óssea e reparação em pacientes.
Reparo ósseo é um processo complexo que começa com a formação de hematoma, inflamação, formação e remodelação em seguida1,2de calos. No entanto, o potencial de regeneração óssea é limitada ao tamanho do osso fratura1,3. Por exemplo, grandes fraturas causadas por trauma, câncer ou osteoporose não podem curar e são chamadas de não-união óssea de fraturas. Estas lesões ósseas, muitas vezes, exigem tratamento para promover a regeneração e reparação óssea fisiológica saudável. Atualmente, o auto-enxerto e aloenxerto transplante ósseo é a abordagem de escolha4 com procedimentos de substituição óssea 2,2 milhões anualmente5. Embora esses procedimentos têm uma elevada taxa de sucesso, pode haver complicações, por exemplo, a limitada disponibilidade de osso, infecção, morbidade do sítio doador e rejeição4. Novas alternativas para a engenharia de tecido ósseo estão a ser procuradas para enfrentar estes desafios.
O design dos biomateriais baseados em polímeros naturais ou sintéticos, biocerâmicas ou metais em combinação com células e moléculas bioativas é sobre a ascensão de6. Nossa compreensão atual do osso fisiológico cura e cura no contexto de biomateriais depende de vários fatores, tais como propriedades mecânicas e múltiplos fatores locais e sistêmicos, incluindo células da circulação e o local da fratura7 ,8,9. Biomateriais para regeneração óssea visam promover osteogenicity e osseointegração10 e são idealmente biocompatível, biodegradável e porosa (promovendo a migração celular, oxigênio e nutrientes). No entanto, se o biomaterial induz inflamação excessiva e respostas alérgicas, isso pode limitar ou inibir11,12de consolidação óssea. Assim, numa abordagem interdisciplinar é necessária avaliar biomateriais desenvolvidos para reparação óssea.
Neste estudo, apresentamos uma avaliação pré-clínica de materiais representativos, 1) Orthovita Vitoss espuma, que é o substituto de enxerto ósseo esponjoso comercialmente disponível, consistindo de fosfato tricálcico composta de nanômetros de tamanho puro β-tricálcio partículas de fosfato (β-TCP) e colágeno bovino tipo 1 (C) (β-TCP/C espuma) e 2) discos de β-TCP. Aqui, ilustramos teste de biocompatibilidade destes biomateriais usando primário osteoblastos (OB) e ensaios de osteoclasto (OC), um modelo in vivo de reparação óssea, avaliação imunológica, compreendendo em vitro proliferação dos linfócitos T e produção de citocinas e na vivo imunogenicidade e alergenicidade, como relatado anteriormente13.
Aqui, nós mostramos uma abordagem multidisciplinar para a avaliação pré-clínica de biocompatibilidade de biomateriais representativos desenvolvido para regeneração óssea e reparação.
