Detta protokoll ger en tillförlitlig metod för att etablera humaniserade möss med både människans immunsystem och leverceller. Dubbla, rekonstituerade, immunotillräckliga möss som erhålls via intrasplenic injektion av humana hepatocyter och CD34+ hematopoietiska stamceller är mottagliga för humant immunbristvirus-1 infektion och recapitulate leverskada som observerats i HIV-infekterade patienter.
Trots den ökade livslängden hos patienter infekterade med humant immunbristvirus-1 (HIV-1), leversjukdom har uppstått som en vanlig orsak till deras sjuklighet. Levern immunopatologi orsakad av HIV-1 förblir svårfångade. Små xenograft djurmodeller med mänskliga hepatocyter och humant immunförsvar kan recapitulate människans biologi av sjukdomens patogenes. Häri, ett protokoll beskrivs för att etablera en dubbel humaniserad musmodell genom mänskliga hepatocyter och CD34+ hematopoietiska Stem/progenitorceller (HSPCs) transplantation, att studera leverimmunopatologi som OBSERVERATS hos hiv-infekterade patienter. För att uppnå dubbel beredning, manliga TK-NOG (NOD. CG-Prkdcscid Il2rgtm1Sug TG (ALB-tk) 7-2/shijic) möss injiceras intraperitonealt med ganciklovir (GCV) doser för att eliminera mus transgena leverceller, och med treosulfläkt för nonmyeloablativ konditionering, som båda underlätta humana hepatocyte (HEP) inympningoch människans immunsystem (hans) utveckling. Humant albumin (ALB) nivåer utvärderas för levertransplantation, och närvaron av mänskliga immunceller i blodet upptäcks av flödescytometri bekräftar inrättandet av människans immunsystem. Den modell som utvecklats med hjälp av det protokoll som beskrivs här liknar flera komponenter av leverskador från HIV-1-infektion. Dess etablering kan visa sig vara avgörande för studier av samtidig hepatitvirus-infektion och för utvärdering av antivirala och antiretrovirala läkemedel.
Sedan tillkomsten av antiretroviral terapi, det har skett en betydande minskning av dödsfall i samband med HIV-1-monoinfektion. Emellertid, leversjukdom har uppstått som en vanlig orsak till sjuklighet hos hiv-infekterade patienter1,2. Infektioner av hepatitvirus med hiv-1-infektion är vanligare, står för 10%-30% av hiv-infekterade personer i USA3,4,5.
Värd-specificiteten av HIV-1 och hepatitvirus begränsar nyttan av små djurmodeller för att studera människospecifika infektionssjukdomar eller för att undersöka flera aspekter av HIV-1-associerad leverpatogenes. Möss med nedsatt immunförsvar som möjliggör en inympelse av mänskliga celler och/eller vävnader (s.k. humaniserade musmodeller) är acceptabla djurmodeller för prekliniska studier6,7,8. Sedan introduktionen av humaniserade möss i början av 2000-talet, flera prekliniska studier av kolestatisk Human levertoxicitet, Human-specifika patogener, inklusive HIV-1 och HIV-associerade neurokognitiva störningar, Epstein Barr-virus, hepatit och andra infektionssjukdomar, har undersökts i dessa möss6,9,10,11. Flera musmodeller för CD34+ hspcs och/eller Human hepatocyte transplantation har länge utvecklats och har förbättrats med tiden för att studera sjukdomens patogenes av hepatit B-virus (HBV)-associerad leversjukdom12, 13 , 14. flera modeller för HSPC och Human hepatocyte (HEP) transplantation baseras på stammar, känd som nog (nod. CG-prkdcscid Il2rgtm1Sug/jictac)8,13, NSG (nod. CG-prkdcscid Il2rgtm1Wjl/szj)15, Balb/C-Rag2-/- c-/- (Rag2TM 1.1 FLV Il2rgTM 1.1 FLV/j)12, och Fah-/- nod rag1-/- il2rγnull mus16. Men varje modell har sina egna fördelar och begränsningar; till exempel AFC8 dubbla humaniserade möss för heps och mänskliga stamceller (hscs) på en Balb/C-Rag2-/- c-/- bakgrund möjliggör en lyckad inympelse av immunceller och hscs, men det finns en avsaknad av en antigen-specifik T-och B-cell svar i denna modell12. Den stora oro för beredning av dubbla humaniserade möss inkluderar suboptimala engrafations, en brist på lämpliga modeller för att stödja olika vävnader, avvikande villkor, immun avvisande, eller graft-Versus-host sjukdom (GVHD), och tekniska svårigheter, såsom riskfyllda manipulationer med nyfödda och hög dödlighet på grund av metabola avvikelser13.
Även humaniserade möss har använts för hiv-forskning för många år17,18,19, användning av humaniserade möss för att studera leverskador orsakade av hiv-1 har varit begränsad20. Vi rapporterade tidigare inrättandet av en dubbel humaniserad TK-NOG musmodell och dess tillämpning i HIV-associerad leversjukdom8. Denna modell visar robust inympelse av lever och immunceller och recapitulates HIV-infektion patogenes. Denna diskussion presenterar ett detaljerat protokoll, inklusive de mest kritiska stegen i transplantation av humana hepatocyter. En beskrivning av HSPCs krävs för en lyckad inympningen av HEPs och inrättandet av ett funktionellt immunförsvar i TK-NOG möss presenteras också. TK-nog manliga möss som transporterar en leverspecifik herpes simplex virus typ 1 tymidin Kinas (HSV-tk) transgenens används. Transplanterade humana leverceller upprätthålls stabilt inom musen levern utan EXOGEN droger21. Mössen är också konditionerade med nonmyeloablativa doser av treosulfläkt att skapa en nisch i musen benmärg för mänskliga celler8. Immunobristfällig TK-NOG möss är intrasplenically injiceras med HEPs och multipotenta HSPCs. Mössen övervakas sedan regelbundet för blod-och lever beredning genom immunophenotypning av blod och mätningar av Human-albuminnivåer i serum. Möss med en lyckad beredning av mer än 15% för både humana immunceller och HEPs är intraperitonealt injiceras med HIV-1. Effekten av HIV på levern kan bedömas så tidigt som 4-5 veckor efter infektion. Det är viktigt att notera att, eftersom HIV-1 används, alla nödvändiga försiktighetsåtgärder måste vidtas vid hantering av viruset och injicera det i möss.
Levern äventyras och skadas hos HIV-infekterade patienter24.
