En protokol for at forberede 13C,15N-mærket svampe og vegetabilske prøver til multidimensional solid-state NMR spektroskopi og dynamisk nuklear polarisering (DNP) undersøgelser er præsenteret.
Denne protokol viser hvordan ensartet 13C, 15N-mærket svampe materialer kan være produceret og hvordan disse bløde materialer bør være gået for solid-state NMR og følsomhed-forstærket DNP eksperimenter. Prøven forædling af plantebiomasse er også detaljeret. Denne metode giver mulighed for måling af en serie af 1D og 2D 13C –13C /15N korrelationer spektre, som giver høj opløsning strukturelle udredning af komplekse biomaterialer i deres oprindelige tilstand, med minimal undertrykkelse af netbårne. Isotop-mærkning kan blive undersøgt af kvantificere intensitet i 1D spectra og polarisering overførsel effektivitet i 2D korrelation spektre. Succes dynamisk nuklear polarisering (DNP) til forberedelse af prøven kan vurderes af følsomhed enhancement faktor. Yderligere eksperimenter undersøger de strukturelle aspekter af polysaccharider og proteiner vil føre til en model af de tre-dimensionelle arkitektur. Disse metoder kan ændres og tilpasses for at undersøge en lang række kulhydratrige materialer, herunder de naturlige cellevægge af planter, svampe, alger og bakterier, såvel som syntetiseres eller designet kulhydratpolymerer og deres komplekse med andre molekyler.
Kulhydrater spiller en central rolle i forskellige biologiske processer såsom energilagring, strukturelle opbygning og cellular anerkendelse og vedhæftning. De er beriget i cellevæggen, hvilket er en grundlæggende komponent i planter, svampe, alger og bakterier1,2,3. Cellens væg fungerer som en central kilde til produktion af biobrændstoffer og biomaterialer samt et lovende mål for antimikrobielle behandlingsformer4,5,6,7,8 , 9.
Den moderne forståelse af disse komplekse materialer har været betydeligt avancerede af årtiers bestræbelser, der var afsat til strukturel karakterisering ved hjælp af fire større biokemiske eller genetiske metoder. Den første store metode bygger på sekventiel behandlinger ved hjælp af barske kemikalier eller enzymer til at nedbryde cellevægge i forskellige dele, der efterfølges af kompositoriske og kobling analyse af sukker i hver fraktion10. Denne metode belyser domæne fordelingen af polymerer, men fortolkningen kan være misvisende på grund af de kemiske og fysiske egenskaber af biomolekyler. For eksempel, er det vanskeligt at afgøre, om den alkali-ekstraherbare brøkdel stammer fra et enkelt domæne af mindre struktureret molekyler eller rumligt separerede molekyler med sammenlignelige opløselighed. For det andet den udpakkede dele eller hele cellevægge kan også måles ved hjælp af løsning NMR til at bestemme de kovalente forbindelser, også betegnes som crosslinking, mellem forskellige molekyler11,12,13, 14,15. På denne måde, kovalente ankre detaljerede struktur kunne blive aftestede, men begrænsninger kan findes på grund af den lave Opløselighed af polysaccharider, det relativt lille antal crosslinking websteder og uvidenhed om non-kovalente effekter, der stabiliserer polysaccharid pakning, herunder hydrogenbindende, van der Waals kraft, elektrostatiske interaktion og polymer entanglement. For det tredje har bindende affinitet været beslutsom in vitro- ved hjælp af isolerede polysaccharider16,17,18,19, men rensning procedurer kan væsentligt ændre struktur og egenskaber af disse biomolekyler. Denne metode også undlader at replikere sofistikerede deposition og samling af makromolekyler efter biosyntesen. Endelig, fænotype, celle morfologi og mekaniske egenskaber af genetiske mutanter med svækkede produktionen af visse cellevæg komponent kaste lys på de strukturelle funktioner af polysaccharider, men mere molekylære beviser er nødvendige for at slå bro over disse makroskopiske observationer med funktionen manipuleret af protein machineries20.
Seneste fremskridt inden for udvikling og anvendelse af multidimensional solid-state NMR spektroskopi har indført en enestående lejlighed til at løse disse strukturelle opgaver. 2D/3D solid-state NMR eksperimenter aktiverer høj opløsning undersøgelse af sammensætningen og arkitektur af kulhydrat-rige materialer i den native tilstand uden større undertrykkelse af netbårne. Strukturelle undersøgelser er gennemført med succes på både primær og sekundær cellevægge af planter, katalytisk behandlede biomassen, bakterielle biofilm, pigment spøgelser i svampe og for nylig af forfattere, intakt cellevægge i en patogen svamp Aspergillus fumigatus 21 , 22 , 23 , 24 , 25 , 26 , 27 , 28 , 29 , 30 , 31. udvikling af dynamiske nukleare polarisering (DNP)32,33,34,35,36,37,38 , 39 , 40 , 41 , 42 letter væsentligt NMR strukturelle udredning som følsomhed styrkelse af DNP markant forkorter den eksperimentelle tid på disse komplekse biomaterialer. Protokollen beskrevet her detaljer procedurerne for isotop-mærkning svamp A. fumigatus og forberede svampe og vegetabilske prøver for solid-state NMR og DNP karakterisering. Lignende mærkning procedurer bør finde anvendelse på andre svampe med ændrede medium, og prøven forberedelse procedurer bør gælde generelt for andre kulhydratrige biomaterialer.
Sammenlignet med de biokemiske metoder, har solid-state NMR fordele som en ikke-destruktiv og høj opløsning teknik. NMR er også kvantitative sammensætning analyse, og i modsætning til de fleste andre analysemetoder, gør ikke har usikkerheden indført af begrænset opløseligheden af Biopolymerer.
