Een protocol voor het voorbereiden van 13C,15N-geëtiketteerden schimmel en plant monsters voor multidimensionale Solid state NMR spectroscopie en dynamische nucleaire polarisatie (DNP) onderzoeken wordt gepresenteerd.
Dit protocol laat zien hoe uniform 13C, 15N-geëtiketteerden schimmel materialen kunnen worden geproduceerd en hoe deze zachte materialen moeten worden voorafgegaan voor Solid state NMR en gevoeligheid-enhanced DNP experimenten. De monster veredeling van plantaardige biomassa is ook gedetailleerd. Met deze methode kan de meting van een reeks van 1D en 2D 13C –13C /15N correlaties spectra, waarmee de hoge resolutie structurele opheldering van complexe biomaterialen in hun oorspronkelijke staat, met minimale verstoring. De isotoop-labeling kan door het kwantificeren van de intensiteit in 1D spectra en het spuitrendement polarisatie in 2D correlatie spectra worden onderzocht. Het succes van de bereiding van de monsters van de dynamische nucleaire polarisatie (DNP) kan worden geëvalueerd door de factor van de verhoging van de gevoeligheid. Verdere experimenten onderzoeken de structurele aspecten van de polysacchariden en eiwitten zal leiden tot een model van de driedimensionale architectuur. Deze methoden kunnen worden gewijzigd en aangepast om te onderzoeken van een breed scala van koolhydraat-rijke materialen, met inbegrip van de natuurlijke celwanden van planten, schimmels, algen en bacteriën, evenals gesynthetiseerd of ontworpen koolhydraten polymeren en hun complex met andere moleculen.
Koolhydraten spelen een centrale rol in verschillende biologische processen zoals energieopslag, structurele gebouw, en cellulaire erkenning en hechting. Ze zijn verrijkt in de celwand, die is een fundamentele component in planten, schimmels, algen en bacteriën1,2,3. De celwand fungeert als een centrale bron voor de productie van biobrandstof en biomaterialen, evenals een veelbelovende doelgroep voor antimicrobiële therapie4,5,6,7,8 , 9.
Het moderne begrip van deze complexe materialen heeft aanzienlijk door decennia van inspanningen die werden gewijd aan de structurele karakterisering met behulp van vier belangrijke biochemische of genetische methoden naar voren geschoven. De eerste belangrijke methode berust op opeenvolgende behandelingen met behulp van agressieve chemicaliën of enzymen te breken de celwanden in verschillende delen, die wordt gevolgd door de samenstelling en analyse van de koppeling van suikers in elke fractie10. Deze methode werpt licht op het domein distributie van polymeren kan, maar de interpretatie misleidend zijn als gevolg van de chemische en fysische eigenschappen van biomoleculen. Bijvoorbeeld, is het moeilijk om te bepalen of de alkali-extraheerbare fractie afkomstig uit één domein van minder strak gestructureerde moleculen of ruimtelijk gescheiden moleculen met vergelijkbare oplosbaarheid is. Ten tweede, de uitgepakte gedeelten of hele celwanden kan ook worden gemeten met behulp van oplossing NMR om te bepalen van de covalente verbanden, ook aangeduid als crosslinking, tussen verschillende moleculen11,12,13, 14,15. Op deze manier de gedetailleerde structuur van covalente ankers gesondeerd kon worden, maar beperkingen kunnen bestaan als gevolg van de lage oplosbaarheid van polysacchariden, het relatief kleine aantal crosslinking sites en de onwetendheid van de niet-covalente effecten die stabiliseert polysaccharide verpakking, met inbegrip van de waterstof-bonding, van der Waals force, elektrostatische interactie en polymeer entanglement. Ten derde, de bindende affiniteit is vastbesloten in vitro met behulp van geïsoleerde polysacchariden16,17,18,19, maar de reiniging procedures kunnen ingrijpend wordt gewijzigd de structuur en eigenschappen van deze biomoleculen. Deze methode mislukt ook de geavanceerde afzetting en assemblage van macromoleculen na biosynthese wilt repliceren. Ten slotte het fenotype, cel morfologie en mechanische eigenschappen van genetische mutanten met verzwakte productie van bepaalde celwand component werpen licht op de structurele functies van polysacchariden, maar meer moleculaire bewijs is nodig om deze brug macroscopische waarnemingen met de gemodificeerde functie van eiwit machineries20.
Recente vooruitgang in de ontwikkeling en toepassing van de multidimensionale Solid state NMR spectroscopie hebben ingevoerd een unieke gelegenheid om deze structurele puzzels op te lossen. 2D/3D solid state NMR experimenten inschakelen met een hoge resolutie onderzoek naar de samenstelling en architectuur van koolhydraat-rijke materialen in de geboortestaat zonder grote perturbation. Structurele studies zijn met succes uitgevoerd op zowel de primaire als de secundaire celwanden van planten, de catalytically behandelde biomassa, bacteriële biofilm, het pigment geesten in schimmels en, onlangs door de auteurs, de intact celwanden in een pathogene schimmel Aspergillus fumigatus 21 , 22 , 23 , 24 , 25 , 26 , 27 , 28 , 29 , 30 , 31. de ontwikkeling van dynamische nucleaire polarisatie (DNP)32,33,34,35,,36,,37,38 , 39 , 40 , 41 , NMR structurele opheldering vergemakkelijkt 42 aanzienlijk als de verhoging van de gevoeligheid door DNP aanzienlijk de experimentele tijd op deze complexe biomaterialen verkort. Het protocol hier beschreven details over de procedures voor de schimmel A. fumigatus isotoop-labeling en voorbereiden van schimmel en plant monsters voor Solid state NMR en DNP karakterisering. Soortgelijke labeling procedures gelden andere schimmels met gewijzigde medium, en de monsterbereidingsprocedures gelden over het algemeen andere koolhydraatrijke biomaterialen.
Vergeleken met de biochemische methoden, heeft Solid state NMR voordelen als een niet-destructieve en hoge resolutie techniek. NMR is ook kwantitatieve samenstelling analyse, en in tegenstelling tot de meeste andere analytische methoden, doet niet hebben de onzekerheden ingevoerd door de beperkte oplosbaarheid van biopolymeren. Oprichting van het huidige protocol vergemakkelijkt toekomstige studies op koolhydraat-rijke biomaterialen en matiemaatschappij polymeren. Echter moet worden opgemerkt dat de resonantie-toewijzing…
