Hier presenteren we een aantal eenvoudige methoden voor de evaluatie van de levensvatbaarheid en de dood in 3D kanker cel sferoïden, die de fysisch-chemische gradiënten van in vivo tumoren veel beter dan de 2D-cultuur na te bootsen. Het sferoïde model maakt het dus mogelijk om de effectiviteit van kanker medicijnen te evalueren met een verbeterde vertaling naar in vivo omstandigheden.
Driedimensionale sferoïden van kankercellen zijn belangrijke hulpmiddelen voor beide kanker geneesmiddelen schermen en voor het verkrijgen van mechanistische inzicht in kanker celbiologie. De kracht van deze voorbereiding ligt in zijn vermogen om te imiteren vele aspecten van de in vivo voorwaarden van tumoren terwijl ze snel, goedkoop, en veelzijdig genoeg om relatief high-throughput screening mogelijk te maken. De sferoïde cultuuromstandigheden kunnen recapituleren de fysisch-chemische gradiënten in een tumor, met inbegrip van de toenemende extracellulaire zuurgraad, verhoogde lactaat, en het verminderen van glucose en zuurstof beschikbaarheid, van de sferoïde periferie aan zijn kern. Ook de mechanische eigenschappen en cel-cel interacties van in vivo tumoren zijn deels nagebootst door dit model. De specifieke eigenschappen en bijgevolg de optimale groeiomstandigheden, van 3D sferoïden, verschillen sterk tussen verschillende soorten kankercellen. Voorts vereist de beoordeling van de levensvatbaarheid van de cellen en de dood in 3D sferoïden methodes die gedeeltelijk van die voor 2D culturen verschillen. Hier beschrijven we verschillende protocollen voor de voorbereiding van 3D sferoïden van kankercellen, en voor het gebruik van dergelijke culturen om de levensvatbaarheid van cellen en de dood te beoordelen in het kader van de evaluatie van de effectiviteit van antikanker
Het gebruik van multicellulaire sferoïde modellen in de Kankerbiologie is een aantal decennia oud1,2, maar heeft aanzienlijke dynamiek opgedaan in de afgelopen jaren. Voor een groot deel weerspiegelt dit meer besef hoe sterk het fenotype van kankercellen afhankelijk is van hun micromilieu en specifieke groeiomstandigheden. De micromilieu in vaste tumoren is fundamenteel verschillend van dat in overeenkomstige normale weefsels. Dit omvat fysisch-chemische voorwaarden zoals pH, zuurstof spanning, evenals interstitiële druk, concentratie gradiënten van oplosbare factoren zoals voedingsmiddelen, afvalproducten, en afgescheiden signalerende samenstellingen (groeifactoren, cytokines). Bovendien omvat het de organisatie van de extracellulaire matrix (ECM), cel-cel interacties en intercellulaire signalering, en andere aspecten van de bijzondere drie-dimensionale (3D) architectuur van de tumor3,4, 5,6. De specifieke microecologische omstandigheden waarin kankercellen bestaan, hebben een diepgaande invloed op hun genexpressie profiel en functionele eigenschappen, en het is duidelijk dat, vergeleken met die van cellen gekweekt in 2D, het fenotype van 3D sferoïden veel nauwer nabootst die van in vivo tumoren7, 8,9,10,11. 2D-modellen, zelfs als ze in dienst hypoxie, zure pH, en hoge lactaat concentraties om na te bootsen bekende aspecten van de tumor micromilieu, nog steeds niet aan de gradiënten van de fysisch-chemische parameters die ontstaan binnen tumoren, evenals hun 3D-tumor vast te leggen Architectuur. Aan de andere kant, dierlijke modellen zijn kostbaar, traag en ethisch problematisch, en in het algemeen, hebben ook tekortkomingen in hun vermogen om de menselijke tumor voorwaarden recapituleren. Bijgevolg zijn 3D sferoïden toegepast als een intermediair complexiteits model in studies van een breed scala van eigenschappen van de meeste vaste kankers9,11,12,13, 14,15,16,17.
Een veelgebruikt gebruik van 3D sferoïden is in screening assays van kankertherapie werkzaamheid9,18,19,20. De reacties van de behandeling zijn bijzonder gevoelig voor de tumor microenvironment, die zowel de invloed van de tortuosity, de beperkte verspreiding, de hoge interstitiële druk, als zure milieu pH op druglevering weerspiegelt, en de invloed van hypoxie en andere aspecten van het micromilieu op de reactie van de celdood9,17. Omdat het milieu binnen 3D sferoïden inherent al deze eigenschappen7,8,9,10,11ontwikkelt, kan het tewerkstellen van 3D cel culturen aanzienlijk verbeteren van de vertaling van de resultaten in vivo omstandigheden, maar toch een efficiënte en betaalbare high-throughput screening van de netto-groei mogelijk te maken. Echter, de grote meerderheid van de studies over de drug Response van kankercellen worden nog steeds uitgevoerd onder 2D-omstandigheden. Dit waarschijnlijk weerspiegelt dat, terwijl sommige analyses kunnen relatief gemakkelijk worden geïmplementeerd voor 3D-cel culturen, veel, zoals levensvatbaarheid assays, West-bevlekken, en immunofluorescentie analyse, zijn veel handiger gedaan in 2D dan in 3D.
Het doel van het huidige werk is het verstrekken van gemakkelijk vatbaar analyses en precieze protocollen voor analyses van het effect van de behandeling met anti-kanker medicijnen op kankercellen levensvatbaarheid en overleving in een 3D-tumor nabootsen instelling. Concreet, bieden en vergelijken we drie verschillende methoden voor sferoïde vorming, gevolgd door methoden voor kwalitatieve en kwantitatieve analyses van groei, levensvatbaarheid en drug Response.
Het gebruik van 3D kanker cel sferoïden heeft bewezen een waardevol en veelzijdig instrument niet alleen voor antikanker drug screening, maar ook voor het verkrijgen van mechanistische inzicht in de regulering van kankercellen dood en levensvatbaarheid onder omstandigheden nabootsen die in de tumor micro-omgeving. Dit is vooral van cruciaal belang als de toegankelijkheid, cellulaire opname, en intracellulaire effecten van chemotherapeutische drugs zijn diep beïnvloed door de fysisch-chemische omstandigheden in de tumor…
