Føtal alkoholspektrumforstyrrelser (FASD) beskriver et svært variabelt kontinuum av etanolinduserte utviklingsdefekter, inkludert ansiktsdysmorfologier og nevrologiske funksjonsnedsettelser. Med en kompleks patologi påvirker FASD ca. 1 av 100 barn født i USA hvert år. På grunn av FASDs svært variable natur har dyremodeller vist seg å være kritiske i vår nåværende mekanistiske forståelse av etanolinduserte utviklingsdefekter. Et økende antall laboratorier har fokusert på å bruke sebrafisk for å undersøke etanolinduserte utviklingsdefekter. Sebrafisk produserer et stort antall eksternt befruktede, genetisk tractable, gjennomsiktige embryoer. Dette gjør det mulig for forskere å nøyaktig kontrollere timing og dosering av etanoleksponering i flere genetiske sammenhenger og kvantifisere virkningen av embryonisk etanoleksponering gjennom levende bildeteknikker. Dette, kombinert med den høye graden av bevaring av både genetikk og utvikling med mennesker, har vist sebrafisk å være en kraftig modell for å studere mekanistisk grunnlag av etanol teratogenitet. Etanoleksponeringsregimer har imidlertid variert mellom ulike sebrafiskstudier, som har forvirret tolkningen av sebrafiskdata på tvers av disse studiene. Føtal alkoholspektrumforstyrrelser (FASD) beskriver et bredt spekter av nevrologiske funksjonsnedsettelser og kraniofaciale dysmorfologier forbundet med embryonisk etanoleksponering1. Flere faktorer, inkludert timing og dosering av etanoleksponering og genetisk bakgrunn, bidrar til variasjonen av FASD2,3. Hos mennesker gjør det komplekse forholdet mellom disse variablene å studere og forstå etiologien til FASD utfordrende. Dyremodeller har vist seg avgjørende for å utvikle vår forståelse av det mekanistiske grunnlaget for etanolteratogenisitet. Et bredt utvalg av dyremodellsystemer har blitt brukt til å studere flere aspekter av FASD, og resultatene har vært bemerkelsesverdig konsistente med det som finnes i eksponering hos mennesker4. Gnagermodellsystemer brukes til å undersøke mange aspekter av FASD, med mus som den vanligste5,6,7. Mesteparten av dette arbeidet har fokusert på utviklingsdefekter til tidlig etanoleksponering8, men senere eksponering for etanol har vist seg å forårsake utviklingsavvik også9. Videre har de genetiske egenskapene til mus i stor grad hjulpet i vår evne til å undersøke de genetiske fundamentene til FASD10,11. Disse studiene hos mus tyder sterkt på at det er gen-etanol interaksjoner med sonic pinnsvin banen, retinsyre signalering, Superoxide dismutase, Nitrogenoksid synthase I, Aldh2 og Fancd28,10,11,12,13,14,15,16,17,18, 19,20,21. Disse studiene viser at dyremodeller er avgjørende for å fremme vår forståelse av FASD og dens underliggende mekanismer.
Sebrafisken har dukket opp som et kraftig modellsystem for å undersøke mange aspekter av etanol teratogenese22,23. På grunn av deres eksterne befruktning, høy fecundity, genetisk tractability, og levende bildebehandling evner, sebrafisk er ideelt egnet til å studere faktorer som timing, dosering, og genetikk av etanol teratogenese. Etanol kan administreres til nøyaktig iscenesatte embryoer, og embryoene kan deretter avbildet for å undersøke den direkte virkningen av etanol under utviklingsprosesser. Dette arbeidet kan være relatert direkte til mennesker, fordi de genetiske utviklingsprogrammene er høyt bevart mellom sebrafisk og mennesker og kan derfor bidra til å veilede FASD menneskelige studier24. Mens sebrafisk har blitt brukt til å undersøke etanol teratogenese, gjør mangel på konsensus i rapportering av embryonale etanolkonsentrasjoner sammenligning med mennesker vanskelig25. I pattedyrsystemer korrelerer blod-alkoholnivåer direkte med vevsetanolnivåer26. Mange av sebrafiskstudiene behandler embryoer før fullstendig dannelse av sirkulasjonssystemet. Uten maternal prøve å undersøke, er det nødvendig med en prosess for å vurdere etanolkonsentrasjoner for å kvantifisere etanolnivåer i embryoet. Som et utviklingsmodellsystem er sebrafisk ideelt egnet til å studere virkningen av miljøfaktorer på utvikling. De produserer et stort antall eksternt befruktede embryoer, noe som gir presis timing og doseringparadigmer i etanolstudier. Dette, kombinert med live imaging evner og genetisk og utviklingsmessig bevaring med mennesker, gjør sebrafisk et kraftig modellsystem for teratologi studier.
