Beskrivs här är en effektiv metod för hur man producerar hög viral titer lager av hepatit E-virus (HEV) för att effektivt infektera hepatoma celler. Med den presenterade metoden kan både icke-hölje, samt höljes virala partiklar skördas och användas för att inokukulera olika cellinjer.
Hepatit E-virus är den främsta orsaken till levercirros och leversvikt med ökande prevalens över hela världen. Det ensträngade RNA-viruset överförs huvudsakligen genom blodtransfusioner, otillräckliga sanitära förhållanden och förorenade livsmedelsprodukter. Hittills off-label drogen ribavirin (RBV) är behandling av val för många patienter. En specifik HEV-behandling återstår dock att identifiera. Hittills har kunskapen om HEV livscykel och patogenes allvarligt hämmats på grund av bristen på en effektiv HEV cell kultursystem. Ett robust cellodlingssystem är viktigt för studiet av den virala livscykeln som även omfattar viruspatogenesen. Med den metod som beskrivs här kan man producera virala titrar på upp till 3 x 106 fokusbildande enhet/ml (FFU/mL) av icke-höljeförsörda HEV och upp till 5 x 104 FFU/ml omsluten HEV. Med hjälp av dessa partiklar är det möjligt att infektera en mängd olika celler av olika ursprung, inklusive primära celler och mänskliga, samt djurcellslinjer. Produktionen av infektiösa HEV-partiklar från plasmider utgör en oändlig källa, vilket gör detta protokoll ytterst effektivt.
Hepatit E är en ganska underskattad sjukdom med ökande prevalens över hela världen. Omkring 20 miljoner infektioner resulterar i mer än 70.000 dödsfall per år1. Det underliggande medlet, hepatit E-virus (HEV), omplacerades nyligen och klassificeras nu inom familjen Hepeviridae inklusive släktena Orthohepevirus och Piscihepevirus. HEV av olika ursprung klassificeras inom arten Orthohepevirus A-D inklusive isolat från människor, svin, kaniner, råttor, fåglar och andra däggdjur2. För närvarande har åtta olika genotyper (GT) av det positiva, enkelsträngade RNA-viruset identifierats2. Även om de skiljer sig åt i sin sekvensidentitet, överföringsvägar och geografisk fördelning, är deras genomiska struktur mycket bevarad. Mer specifikt är 7,2 kbp HEV-genomet indelat i 3 stora öppna läsramar (ORF1-3). Medan ORF1 kodar alla enzymer som behövs för en lyckad replikering i värdcellen kodar ORF2 capsidproteinet och ORF3-proteinet fungerar som en funktionell jonkanal som krävs för montering och frisättning av infektiösa partiklar3. När de släpps ut i den basala eller apikala lumen HEV finns i båda, quasienveloped och icke-höljeförsedd / naken art beroende på om viruset kommer från blod eller avföring,respektive 4,5.
Medan GT1 och GT2 huvudsakligen finns i utvecklingsländer som enbart infekterar människor6 via fekal-oral väg, GT3, GT4 och GT7 förekommer främst i utvecklade länder1,7 med en mängd olika arter som fungerar som reservoarer, t.ex., svin8,råtta9,kyckling10,,11, rådjur12, mongoose13,fladdermus14,kanin15,,16, vildsvin17 och många fler7,,18,19, som visar zoonos7,,20,,21,22. Förutom otillräckliga sanitära förhållanden23 och förorenade livsmedelsprodukter12,,24,,25,26,överföring via blodtransfusion och organtransplantationer är också möjligt27,28. HEV är en vanlig orsak till levercirros och leversvikt29 särskilt hos patienter med redan existerande leversjukdom, immunkomprometterade individer (genotyp 3, 4 och 7) och gravida kvinnor (genotyp 1). Observera att det också finns extrahepatiska manifestationer som hematopoietisk sjukdom30,,31,32, neurologiska sjukdomar33 och njurskada34.
Hittills är off-label drogen ribavirin (RBV) behandling av val för många infekterade patienter35,36. Fall av behandlingssvikt och dåliga kliniska långsiktiga resultat har dock rapporterats. Behandlingssvikt har kopplats till viral mutagenesis och ökad viral heterogenitet hos kroniskt infekterade patienter37,38,39. Tvärtom kunde en nyligen genomförd europeisk retrospektiv multicenterstudie inte korrelera polymerasmutationer med RBV-behandlingssvikt40. I kliniska observationer och in vitro-experiment, interferon41,42,43, sofosbuvir44,45, zinksalter46 och silvestrol47,48 har också visat antivirala effekter. Icke desto mindre återstår en specifik HEV behandling att finna, hämmas av bristen på kunskap om HEV livscykel och dess patogenes. Därför behövs ett robust cellodlingssystem för virologiska studier och utveckling av nya antivirala läkemedel49.
Tyvärr, liksom andra hepatit virus, HEV är svårt att sprida sig i konventionella cellinjer och oftast fortskrider mycket långsamt leder till låga virusbelastningar. Ändå, vissa grupper kunde öka virusbelastningar av generering av cellinje subkloner50 eller justering av media kosttillskott51. Nyligen generering av cDNA kloner52 och anpassning av primära patienten isolat genom att passera53,54 ytterligare förbättrad HEV förökning i cellkultur55. I detta protokoll använde vi arvsmassan hos en cellkultur anpassad Kernow-C1 stam (kallad p6_WT)54 och en mutant stam som hyser en replikeringshöjande mutation (kallad p6_G1634R)37. Kernow-C1 är den mest använda stammen i HEV cellkultur och kan producera höga virusbelastningar. Genom att bedöma virala RNA-kopieringsnummer kan HEV-replikation övervakas in vitro. Dessa tekniker tillåter dock inte bedömning av antalet smittsamma partiklar som produceras. Därför har vi etablerat en immunofluorescensfärgning för att bestämma Fokusbildande enheter (FFU/mL).
Den här beskrivna metoden56 kan användas för att producera full längd infektiösa viruspartiklar som kan infektera en mängd olika celltyper från olika ursprung, inklusive primära celler och däggdjurs cellinjer. Detta är en grundläggande förutsättning för att dechiffrera viktiga aspekter av HEV-infektion och tropism. Det finns inget behov av inokulering med vanligtvis begränsade patientisolat. Produktionen av infektiösa HEV-partiklar från plasmider utgör en oändlig källa, vilket gör detta protokoll jämförbart effektivt. Dessutom kan detta system användas för omvänd genetik som möjliggör studier av in vivo-identifierade genomförändringar och deras inverkan på HEV-replikation och kondition. Denna teknik övervinner många begränsningar och, kan väg vägen för läkemedelsutveckling, mutagenesis studier och utvärdering av virus-värd interaktioner såsom begränsning eller faktorer inträde.
Från och med plasmidpreparatet bör DNA-avkastningen överstiga 150 ng/μL för att kunna utföra flera linjäriseringar från samma plasmidlager, vilket minimerar risken för bakterieinducerad mutagenes av avgörande genomsekvenser. Dessutom är det viktigt att kontrollera begränsningen sammandrag för fullständig plasmid linjärisering av gel elektrofores (figur 8b).
