Hier stellen wir eine modifizierte Methode zur Isolierung und Kultur menschlicher Zahnfleischepithelzellen vor, indem wir den Rock-Inhibitor Y-27632 zur traditionellen Methode hinzufügen. Diese Methode ist einfacher, weniger zeitaufwendig, verbessert die Stammzelleigenschaften und produziert eine größere Anzahl von Epithelzellen mit hohem Potenzial sowohl für das Labor als auch für klinische Anwendungen.
Das Zahnfleischgewebe ist die erste Struktur, die parodontales Gewebe schützt und bei vielen oralen Funktionen eine bedeutende Rolle spielt. Das Zahnfleischepithel ist eine wichtige Struktur des Zahnfleischgewebes, insbesondere bei der Reparatur und Regeneration von Parodontalgewebe. Die Untersuchung der Funktionen von Zahnfleischepithelzellen hat einen entscheidenden wissenschaftlichen Wert, wie die Reparatur von mundlichen Defekten und die Erkennung der Kompatibilität von Biomaterialien. Da es sich bei menschlichen Zahnfleischepithelzellen um hochdifferenzierte keratinisierte Zellen handelt, ist ihre Lebensdauer kurz und sie sind schwer zu durchlassen. Bisher gibt es nur zwei Möglichkeiten, Zahnfleischepithelzellen zu isolieren und zu kulturieren, eine direkte Explantierungsmethode und eine enzymatische Methode. Die Zeit, die benötigt wird, um Epithelzellen mit der Direktentpflanzungsmethode zu erhalten, ist jedoch länger und die Zellüberlebensrate der enzymatischen Methode ist niedriger. Klinisch ist der Erwerb von Zahnfleischgewebe begrenzt, so dass ein stabiles, effizientes und einfaches In-vitro-Isolations- und Kultursystem erforderlich ist. Wir haben die traditionelle enzymatische Methode verbessert, indem wir Y-27632 hinzugefügt haben, einen Rho-assoziierten Kinase(ROCK)-Inhibitor, der selektiv das Wachstum von Epithelzellen fördern kann. Unsere modifizierte enzymatische Methode vereinfacht die Schritte der traditionellen enzymatischen Methode und erhöht die Effizienz der Kultivierung von Epithelzellen, was erhebliche Vorteile gegenüber der Direktentregungsmethode und der enzymatischen Methode hat.
Die menschliche Gingiva, die erste Verteidigungsstruktur, die Parodontalgewebe schützt, ist nicht nur eine physikalische und chemische Barriere1,sondern sezerniert auch verschiedene Klassen von Entzündungsmediatoren, die an Immunantworten beteiligt sind und eine Immunbarriere bilden2,3. Das Zahnfleischepithel spielt eine wichtige Rolle bei der Reparatur und Regeneration von Parodontalgewebe. Daher ist die Untersuchung der Abwehr und Immunität des Zahnfleischepithels von großer Bedeutung für das Verständnis des Auftretens, der Diagnose und der Behandlung von Parodontitis. Die Isolierung und Kultur von Zahnfleischepithelzellen aus menschlichem Zahnfleischgewebe ist der erste Schritt, der für die Untersuchung des Zahnfleischepithels erforderlich ist. Ein solches Verfahren erfordert grundlegende Operationen wie die Herstellung von Samenzellen für das Tissue Engineering, In-vitro-Modelle von parodontalen assoziierten Erkrankungen und Materialien zur Reparatur von Parodontaldefekten.
Primäre Zahnfleischepithelzellen zeichnen sich durch eine geringe Teilungsrate in vitro4aus, Forscher suchen seit Jahrzehnten nach einer optimalen Isolations- und Kultivierungsmethode. Bis heute werden zwei verschiedene Techniken, eine Direktexplantenmethode und eine enzymatische Methode, üblicherweise in Laboratorien verwendet, um primäre Zahnfleischepithelzellen in vitrozu erhalten4,5. Die Direktexplant-Methode hat Vorteile wie die Anforderung einer geringeren Menge an Gewebeproben und ein einfaches Isolationsverfahren, hat jedoch die Nachteile einer längeren Kulturzeit und Kontaminationsanfälligkeit5. Obwohl die enzymatische Methode die benötigte Kulturzeit verkürzt, ist die Effizienz relativ gering und variiert je nach verwendeten Enzymen und Medium. Kedjarune et al.6 zeigten, dass die Direktentpflanzungsmethode, die mehr Zeit vor der Subkultur (2 Wochen) benötigt, für die Kultivierung von Zahnfleischepithelzellen erfolgreicher zu sein schien als die enzymatische Methode. Beim Vergleich dieser beiden Methoden fanden Klingbeil et al.7 jedoch heraus, dass die enzymatische Methode die besten Ergebnisse für Primärkulturen oraler Epithelzellen erzielte und es möglich war, die optimale Zellausbeute innerhalb kürzester Zeit zu erzielen (11,9 Tage gegenüber 14,2 Tagen).
Daher war es wichtig, eine bequemere und effektivere Methode zur Isolierung und Kultur oraler Epithelzellen zu entwickeln4. Wir haben bereits berichtet, dass die Zugabe von Y-27632, einem Inhibitor der Rho-assoziierten Proteinkinase (ROCK), das Isolationsverfahren von menschlichen primären epidermalen Zellen und Keratinozyten aus adultem Hautgewebe vereinfacht8,9,10. Wir entwickelten G-Medium, ein neues konditioniertes Impfmedium, das spontan epidermale von dermalen Zellen trennt und das Wachstum und dieAusbeuteprimärer epidermaler Zellen unterstützt 8,9,10. In der vorliegenden Studie haben wir eine neue serumfreie Isolations- und Kulturtechnik für Zahnfleischepithelzellen entwickelt, indem wir G-Medium mit Y-27632 kombiniert haben. Im Wesentlichen basiert unsere Methode auf einer Vereinfachung der traditionellen zweistufigen enzymatischen Methode, daher haben wir unsere neue Methode mit der Direktentstechtungsmethode verglichen. Diese modifizierte enzymatische Methode verkürzt signifikant die Zeit, die benötigt wird, um Zahnfleischepithelzellen vom Zahnfleischgewebe zu trennen, und erhöht die Effizienz der Kultivierung von Zahnfleischepithelzellen.
