Hier presenteren we een aangepaste methode voor de isolatie en kweek van menselijke gingivale epitheelcellen door de Rock-remmer, Y-27632, toe te voegen aan de traditionele methode. Deze methode is eenvoudiger, minder tijdrovend, verbetert de eigenschappen van stamcellen en produceert grotere aantallen epitheelcellen met een hoog potentieel, zowel voor het laboratorium als voor klinische toepassingen.
Het gingivale weefsel is de eerste structuur die parodontale weefsels beschermt en een betekenisvolle rol speelt in veel orale functies. Het gingivale epitheel is een belangrijke structuur van gingivaal weefsel, vooral bij het herstel en de regeneratie van parodontaal weefsel. Het bestuderen van de functies van gingivale epitheelcellen heeft cruciale wetenschappelijke waarde, zoals het repareren van orale defecten en het detecteren van de compatibiliteit van biomaterialen. Omdat menselijke gingivale epitheelcellen sterk gedifferentieerde verhoornde cellen zijn, is hun levensduur kort en zijn ze moeilijk te passeren. Tot nu toe zijn er slechts twee manieren om gingivale epitheelcellen te isoleren en te kweken, een directe explantmethode en een enzymatische methode. De tijd die nodig is om epitheelcellen te verkrijgen met behulp van de directe explantmethode is echter langer en de celoverleving van de enzymatische methode is lager. Klinisch gezien is de verwerving van tandvleesweefsel beperkt, dus een stabiel, efficiënt en eenvoudig in vitro isolatie- en kweeksysteem is nodig. We verbeterden de traditionele enzymatische methode door Y-27632 toe te voegen, een Rho-geassocieerde kinase (ROCK) -remmer, die selectief de groei van epitheelcellen kan bevorderen. Onze aangepaste enzymatische methode vereenvoudigt de stappen van de traditionele enzymatische methode en verhoogt de efficiëntie van het kweken van epitheelcellen, wat aanzienlijke voordelen heeft ten opzichte van de directe explantmethode en de enzymatische methode.
De menselijke gingiva, de eerstelijns verdedigingsstructuur die parodontaal weefsel beschermt, is niet alleen een fysieke en chemische barrière1, maar scheidt ook verschillende klassen van ontstekingsmediatoren af die deelnemen aan immuunresponsen en een immuunbarrière vormen2,3. Het gingivale epitheel speelt een belangrijke rol bij het herstel en de regeneratie van parodontaal weefsel. Daarom is het bestuderen van de verdediging en immuniteit van het gingivale epitheel van groot belang voor het begrijpen van het optreden, de diagnose en de behandeling van parodontitis. De isolatie en kweek van gingivale epitheelcellen uit menselijk gingivaal weefsel is de eerste stap die nodig is voor het bestuderen van het gingivale epitheel. Een dergelijke procedure vereist basisoperaties zoals het produceren van zaadcellen voor weefselmanipulatie, in vitro modellen van parodontale geassocieerde ziekten en materialen voor het repareren van parodontale defecten.
Primaire gingivale epitheelcellen worden gekenmerkt door een lage delingsgraad in vitro4, onderzoekers zijn al tientallen jaren op zoek naar een optimale isolatie- en teeltmethode. Tot op heden worden twee verschillende technieken, een directe explantmethode en een enzymatische methode, vaak gebruikt in laboratoria om primaire gingivale epitheelcellen in vitrote verkrijgen4,5. De directe explantmethode heeft voordelen zoals de vereiste van een lagere hoeveelheid weefselmonsters en een eenvoudige isolatieprocedure, maar heeft de nadelen van een langere kweektijd en gevoeligheid voor besmetting5. Hoewel de enzymatische methode de benodigde kweektijd verkort, is de efficiëntie relatief laag en varieert afhankelijk van de gebruikte enzymen en het gebruikte medium. Kedjarune et al.6 toonden aan dat de directe explantmethode, die meer tijd nodig heeft voor subcultuur (2 weken), succesvoller bleek te zijn voor het kweken van gingivale epitheelcellen in vergelijking met de enzymatische methode. Bij het vergelijken van deze twee methoden vonden Klingbeil et al.7 echter dat de enzymatische methode de beste resultaten had voor primaire culturen van orale epitheelcellen en dat het mogelijk was om de optimale celopbrengst te verkrijgen binnen de kortste tijdsperiode (11,9 dagen versus 14,2 dagen).
Daarom was het belangrijk om een handigere en effectievere methode te ontwikkelen voor de isolatie en kweek van orale epitheelcellen4. We hebben eerder gemeld dat het toevoegen van Y-27632, een remmer van Rho-geassocieerd eiwitkinase (ROCK), de isolatieprocedure van menselijke primaire epidermale cellen en keratinocyten uit volwassen huidweefselsvereenvoudigt 8,9,10. We ontwikkelden G-medium, een nieuw geconditioneerd inentingsmedium dat spontaan epidermale van dermale cellen scheidt en de groei en opbrengst van primaire epidermale cellenondersteunt 8,9,10. In de huidige studie ontwikkelden we een nieuwe serumvrije isolatie- en kweektechniek voor gingivale epitheelcellen door G-medium te combineren met Y-27632. In essentie is onze methode gebaseerd op een vereenvoudiging van de traditionele tweestaps enzymatische methode, dus hebben we onze nieuwe methode vergeleken met de directe explantmethode. Deze gemodificeerde enzymatische methode verkort aanzienlijk de tijd die nodig is om gingivale epitheelcellen te scheiden van gingivaal weefsel en verhoogt de efficiëntie van het kweken van gingivale epitheelcellen.
Het gingivale weefsel is een belangrijke structuur die parodontale integriteit en gezondheid handhaaft.
