Nous décrivons ici une méthode pour purifier les chloroplastes intacts des feuilles de l’Arabidopsis et leurs trois compartiments secondaires principaux (enveloppe, stroma et les thylakoïdes), en utilisant une combinaison de centrifugations différentielles, gradient de Percoll continue, et gradients de sucrose discontinu. La méthode est importante par subplastidial et localisation sous-cellulaire des protéines par immunotransfert et protéomique.
