Nevrale kretser er topografisk organisert i funksjonelle rom med spesifikke molekylære profiler. Her gir vi de praktiske og tekniske tiltak for å avsløre globale hjerne topografi med en allsidig wholemount immunhistokjemisk farging tilnærming. Vi viser nytten av metoden ved hjelp av godt forstått cytoarchitecture og krets av lillehjernen.
Den gjentatte og godt forstått mobilnettet arkitektur av lillehjernen gjør det til et ideelt modellsystem for å utforske hjernen topografi. Underliggende sin relativt ensartet cytoarchitecture er en kompleks rekke parasagittal domener av gen og protein uttrykk. Den molekylære compartmentalization av lillehjernen speiles av den anatomiske og funksjonelle organiseringen av afferente fibre. Å få fullt utbytte av kompleksiteten i lillehjernen organisasjon vi tidligere videreutviklet en wholemount farging tilnærming for høy gjennomstrømning analyse av mønster defekter i musen lillehjernen. Denne protokollen beskriver i detalj de reagenser, verktøy og praktiske tiltak som er nyttige for å kunne avdekke protein uttrykk mønstre i den voksne musen lillehjernen ved hjelp wholemount farging. Trinnene uthevet her demonstrere nytten av denne metoden å bruke uttrykket av zebrinII / aldolaseC som et eksempel på hvordan den fine topografien i hjernen kan bli avslørt i sininnfødt tredimensjonale konformasjon. Også beskrevet er tilpasninger til protokollen som tillater visualisering av protein uttrykk i afferente anslag og stor cerebella for komparative studier av molekylære topografi. For å illustrere disse programmene, er data fra afferent farging av rotte lillehjernen inkludert.
Vi har beskrevet de tekniske detaljene som kreves for vellykket wholemount farging med en allsidig immunhistokjemisk tilnærming for avslørende protein uttrykk i utviklingsland og voksne hjernen. Ved å bruke denne tilnærmingen, kan kompliserte molekylære uttrykk mønstre bli analysert og hjerne topografi verdsatt uten behov for arbeidskrevende og tidkrevende vev utsnitt prosedyrer.
