Fluoreszierend<em> In situ</em> Hybridisierung (FISH) auf mRNA-Transkripte in den einzelnen Zellen zu identifizieren ermöglicht die Analyse von polygenen Tätigkeit wie die gleichzeitige Transkription von mehr als einem Mitglied der<em> Var</em> Multigenfamilie in<em> Plasmodium falciparum</em> Infizierten Erythrozyten<sup> 1</sup>. Die Technik ist anpassbar und kann bei verschiedenen Arten von Genen, Zellen und Organismen verwendet werden.
Adhäsion von Plasmodium falciparum infizierten Erythrozyten (IE) für die menschliche endotheliale Rezeptoren bei Malaria-Infektionen durch Expression PfEMP1 Protein-Varianten durch die Gene kodiert var vermittelt.
Die haploiden P. falciparum Genoms beherbergt etwa 60 verschiedene Gene var von denen nur eine geglaubt hat, die pro Zelle zu einem Zeitpunkt während des Blut Stadium der Infektion transkribiert. Wie solche gegenseitig ausschließenden Regulierung der Gentranskription var erreicht ist unklar, da die Identifizierung einzelner Gene oder var Untergruppen von var Gene mit verschiedenen Rezeptoren und die Folge der unterschiedlichen Bindung auf das klinische Ergebnis von P. zugeordneten falciparum-Infektionen. Vor kurzem hat die gegenseitig ausschließende Transkription Paradigma in Zweifel durch Transkription Assays auf Basis einzelner P. aufgerufen falciparum transcript Identifikation in einzelnen infektiert erythrozytären Zellen unter Verwendung von RNA Fluoreszenz in situ Hybridisierung (FISH) von var Gentranskription durch den Parasiten im einzelnen Kerne von P. falciparum IE 1.
Wir beschreiben hier ein ausführliches Protokoll zur Durchführung des RNA-FISH-Methodik zur Analyse von var Gentranskription in Einzel-Kerne von P. falciparum infiziert menschliche Erythrozyten. Das Verfahren beruht auf der Verwendung von Digoxigenin-Basis und Biotin-markierte Antisense-RNA-Sonden unter Verwendung der Fluoreszenz-Plus-TSA Palette System 2 (Perkin Elmer), mikroskopische Analysen und frisch ausgewählt P. falciparum IE. Die in situ-Hybridisierungsverfahren verwendet werden, um Transkription und Regulation einer Vielzahl von Genen in den verschiedenen Phasen des P. ausgedrückt überwachen falciparum Lebenszyklus und ist anpassungsfähig an andere Malaria-Parasiten Arten und anderen Organismen und Zelltypen.
FISH-Analyse RNA im Gegensatz zu Verfahren, wie Northern Blot und RT-PCR, erlaubt die Diskriminierung von spezifischen mRNA-Transkripte in der einzelnen Zelle. Dies macht es möglich, zwischen transkriptionell aktiven und inaktiven Zellen zu unterscheiden, in diesem Beispiel, P. parasitischen Protozoen falciparum in menschlichen roten Blutzellen. Solche whole-cell Beobachtungen sind oft notwendig und kann zu entwirren wichtige und neuartige transkriptionellen Mustern ein.
