Protokollen er beskrevet i dette dokumentet tar sikte på å forklare og forkorte de mange hindringer i veien for den intrikate rute som fører til endrede nukleosidtrifosfater. På den annen side, er modifiserte nukleosidtrifosfater aktivert byggeklosser som er blitt anvendt for den milde innføring av mange funksjonelle grupper i nukleinsyrer, en strategi som legger til rette for bruk av modifiserte nukleinsyrer i en bred palett av praktiske anvendelser som funksjonell tagging og generering av ribozymes og DNAzymes. En av de store utfordringer ligger i det detaljerte metode som leder til isolering og karakterisering av disse nukleosidanaloger.
I denne videoen artikkelen presenterer vi en detaljert protokoll for syntese av these modifiserte analoger ved hjelp av fosfor (III)-baserte reagenser. I tillegg er fremgangsmåten for deres biokjemiske karakterisering røpet, med spesiell vekt på primer forlengelsesreaksjoner og TdT tailing polymerisasjon. Denne detaljerte protokollen vil være til nytte for laging av modifiserte dNTPs og deres videre bruk i kjemisk biologi.
5'-nukleosidtrifosfater ((d) NTPS) representerer en klasse av viktige biomolekyler som er involvert i mange prosesser og funksjoner som varierer fra å være den universelle valutaen for energi til regulatorer av cellemetabolismen.
