Time-lapse imaging in the living animal provides valuable information on structural reorganization in the intact brain. Here, we introduce a thinned-skull preparation that allows transcranial imaging of fluorescently labeled synaptic structures in the living mouse cortex by two-photon microscopy.
In de zoogdieren cortex neuronen vormen extreem gecompliceerde netwerken en informatie uitwisselen bij synapsen. Veranderingen in synaptische sterkte, evenals toevoeging / verwijdering van synapsen, plaatsvinden in een ervaringsafhankelijke wijze die de structurele basis van neuronale plasticiteit. Als postsynaptische onderdelen van de exciterende synapsen in de cortex, worden dendritische beschouwd als een goede benadering van synapsen zijn. Het nemen van de voordelen van de muis genetica en fluorescentielabelling technieken, individuele neuronen en hun synaptische structuren kunnen in de intacte hersenen worden geëtiketteerd. Hier introduceren we een transcraniële imaging protocol met behulp van twee-foton laser scanning microscopie om fluorescent gelabelde postsynaptische vertakte stekels in de tijd in vivo te volgen. Dit protocol maakt gebruik van een uitgedund-schedel voorbereiding, waarbij de schedel intact houdt en voorkomt inflammatoire effecten veroorzaakt door blootstelling van de hersenvliezen en de cortex. Daarom beelden kunnen direct na su verworvenrgery wordt uitgevoerd. De experimentele procedure kan herhaaldelijk worden uitgevoerd op verschillende tijdstippen, variërend van enkele uren tot jaren. De toepassing van dit preparaat kan ook worden uitgebreid tot verschillende corticale gebieden en lagen, alsmede andere celtypes, onder fysiologische en pathologische omstandigheden.
De zoogdieren cortex participeert in vele hersenfuncties, van zintuiglijke waarneming en controle voor het abstracte informatieverwerking en cognitie. Verschillende corticale functies bouwen op verschillende neurale circuits, die zijn opgebouwd uit verschillende soorten neuronen communiceren en uitwisselen van informatie op individueel synapsen. De structuur en functie van synapsen consequent worden gewijzigd ingevolge ervaringen en pathologieën. In de volwassen hersenen, synaptische plasticiteit in de vorm van zowel kracht veranderingen en toevoeging / verwijdering van synapsen, spelen een belangrijke rol bij de vorming en instandhouding van een functionele neurale circuits. Dendritische stekels zijn de postsynaptische onderdelen van de meerderheid van exciterende synapsen in de hersenen van zoogdieren. De constante omzet en morfologische veranderingen van de stekels worden verondersteld om te dienen als een goede indicator van veranderingen in synaptische verbindingen 1-7.
Twee-foton laser scanning microscopie biedt diepe penetratie door dikke, ondoorzichtige voorbereidingen en lage fototoxiciteit, waardoor het geschikt is voor live-imaging in de intacte hersenen 8 maakt. In combinatie met fluorescentielabelling, twee-foton beeldvorming biedt een krachtig instrument om een kijkje in het levende brein en volg structurele reorganisatie bij individuele synapsen met een hoge ruimtelijke en temporele resolutie. Verschillende methoden zijn gebruikt om muizen te bereiden voor levende beeldvorming 9-13. Hier beschrijven we een uitgedunde schedel voorbereiding van in vivo twee-foton beeldvorming om de structurele plasticiteit van postsynaptische vertakte stekels in de muis cortex onderzoeken. Met deze benadering hebben onze recente studies een dynamisch beeld van dendritische spine veranderingen weergegeven in reactie op motorische vaardigheden leren Met de toenemende beschikbaarheid van met fluorescent gelabelde neuronale subsets en snelle ontwikkeling van in vivo labeling technieken transgene dieren, kan dit hier beschreven ook worden toegepast te onderzoekente andere celtypes en corticale gebieden, gecombineerd met andere manipulaties, alsook in ziektemodellen 16-23.
Om een succesvolle uitgedund-schedel voorbereiding te verkrijgen, verschillende stappen in dit protocol zijn cruciaal. 1) De dikte van de schedel.
