Pair recordings are simultaneous whole cell patch clamp recordings from two synaptically connected neurons, enabling precise electrophysiological and pharmacological characterization of the synapses between individual neurons. Here we describe the detailed methodology and requirements for establishing this technique in organotypic hippocampal slice cultures in any laboratory equipped for electrophysiology.
Pair recordings involve simultaneous whole cell patch clamp recordings from two synaptically connected neurons, enabling not only direct electrophysiological characterization of the synaptic connections between individual neurons, but also pharmacological manipulation of either the presynaptic or the postsynaptic neuron. When carried out in organotypic hippocampal slice cultures, the probability that two neurons are synaptically connected is significantly increased. This preparation readily enables identification of cell types, and the neurons maintain their morphology and properties of synaptic function similar to that in native brain tissue. A major advantage of paired whole cell recordings is the highly precise information it can provide on the properties of synaptic transmission and plasticity that are not possible with other more crude techniques utilizing extracellular axonal stimulation. Paired whole cell recordings are often perceived as too challenging to perform. While there are challenging aspects to this technique, paired recordings can be performed by anyone trained in whole cell patch clamping provided specific hardware and methodological criteria are followed. The probability of attaining synaptically connected paired recordings significantly increases with healthy organotypic slices and stable micromanipulation allowing independent attainment of pre- and postsynaptic whole cell recordings. While CA3-CA3 pyramidal cell pairs are most widely used in the organotypic slice hippocampal preparation, this technique has also been successful in CA3-CA1 pairs and can be adapted to any neurons that are synaptically connected in the same slice preparation. In this manuscript we provide the detailed methodology and requirements for establishing this technique in any laboratory equipped for electrophysiology.
Glutamaat receptoren bemiddelen de meerderheid van prikkelende synaptische transmissie in het centrale zenuwstelsel synapsen. De twee belangrijkste subtypes van ionotrope glutamaatreceptoren gelokaliseerd aan het hoofd van de wervelkolom postsynaptische membraan N-methyl-D-aspartaat (NMDA) en α-amino-3-hydroxy-5-methylisoxazool-4-proprioniczuur (AMPA) receptoren. Bij rust membraanpotentialen, AMPA receptoren dragen meeste postsynaptische stroom tijdens synaptische transmissie. In de hippocampus, het NMDA receptor speelt een belangrijke rol bij het ontstaan van veranderingen in het aantal AMPA-receptoren in het postsynaptische membraan: door als "koincidentiedetektor" 1 tot veranderingen in synaptische sterkte 1 initiëren de NMDA receptor deelneemt aan de synaptische mechanismen die worden verondersteld om te leren en het geheugen ondersteunen op een subcellulaire niveau. In reactie op depolarisatie van het postsynaptische neuron parallel aan presynaptische afgifte van transmitter, calcium komt via de NMDAreceptor AMPA receptor inbrengen of verwijderen 2 initiëren. Deze receptor dynamiek ten grondslag liggen synaps plasticiteit: een toename van de synaptische kracht is op lange termijn potentiëring 2,3 (LTP), terwijl een afname in synaptische kracht is op lange termijn depressie 4 (LTD). Daarom AMPA receptor beweging wordt verantwoordelijk geacht voor synaptische plasticiteit expressie, terwijl NMDA-receptoren wordt gedacht dat de inductie controleren.
Het bepalen van de precieze mechanismen die ten grondslag liggen aan synaptische transmissie en plasticiteit vereist het bestuderen van kleine populaties van synapsen, idealiter enkele synapsen. Terwijl sommige synapsen zeer geschikt voor onderzoek op dit niveau, bijvoorbeeld de calyx van Held 5 voor de meeste synaptische populaties is uiterst moeilijk door de kleine en diffuse aard van de synaptische verbindingen. Twee belangrijke elektrofysiologie technieken ontwikkeld enkelvoudige synaptische verbindingen onderzocht: het eerste is minimaal stimulatie, where een presynaptische vezel wordt verondersteld gestimuleerd extracellulair. De tweede techniek is gekoppeld opnamen, waarbij twee simultane gehele cellen opnames van synaptisch verbonden neuronen wordt uitgevoerd. Een groot voordeel van minimale stimulatie is dat het snel en relatief eenvoudig uit te voeren, waarbij de plaatsing van een extracellulair stimulerende elektrode in de axonale kanaal tegelijkertijd opnemen van een postsynaptische neuron. De primaire zorg bij het gebruik van deze techniek is dat betrouwbare stimulatie van een enkele cel zelden worden gegarandeerd proef na proef.
In de afgelopen vijftien jaar hebben we routinematig gebruikt gepaarde whole-cell opnames van twee synaptically verbonden piramidale neuronen 6-17. Het grote voordeel van deze techniek is dat slechts een presynaptische neuron consistent en betrouwbaar wordt gestimuleerd. Ook kan niet alleen elektrofysiologische karakterisering maar ook farmacologische manipulatie van het presynaptische neuron 6,18 </ Sup>. De waarschijnlijkheid van synaptische verbindingen tussen neuronen is laag, waardoor verbonden paren moeilijk te verkrijgen 19. Het gebruik van organotypische brain slice cultures omzeilt het obstakel als synaptische connectiviteit kan herstellen in vitro en bovendien de aard van de resulterende verbinding is vergelijkbaar met die van natief hersenweefsel 20. Bovendien organotypische kweken drukken LTP, LTD 7-10,12-15,21 en andere vormen van kortdurende synaptische plasticiteit inbegrip gepaarde puls facilitatie (PPF) en depressie (PPD) 6,22,23, waardoor plasticiteit mechanismen bestudeerd in paren neuronen. Hier beschrijven we de gedetailleerde methodologie betrokken succes bereiken gekoppeld opnamen in dit in vitro systeem. Deze informatie kan gemakkelijk worden aangepast aan andere experimentele systemen, waaronder acute segmenten en andere hersengebieden.
Hier hebben we beschreven de vereisten voor de oprichting van een succesvolle gepaarde whole cell opnames in organotypische hippocampus slice culturen. Gepaarde opnames kunnen ook worden uitgevoerd in verschillende bereidingen, waaronder acute plakjes en gedissocieerde kweeksystemen 26,27.
