Environmental enrichment provides a potential protective effect against neurodegenerative disorders. Currently, however, there is no easy way of determining the efficacy of enrichment procedures. This protocol describes a simple “Puzzle Box” method for assessing an animal’s cognitive function, in order to reveal the effectiveness of environmental enrichment.
Environmental enrichment can dramatically influence the development and function of neural circuits. Further, enrichment has been shown to successfully delay the onset of symptoms in models of Huntington’s disease 1-4, suggesting environmental factors can evoke a neuroprotective effect against the progressive, cellular level damage observed in neurodegenerative disorders. The ways in which an animal can be environmentally enriched, however, can vary considerably. Further, there is no straightforward manner in which the effects of environmental enrichment can be assessed: most methods require either fairly complicated behavioral paradigms and/or postmortem anatomical/physiological analyses. This protocol describes the use of a simple and inexpensive behavioral assay, the Puzzle Box 5-7 as a robust means of determining the efficacy of increased social, sensory and motor stimulation on mice compared to cohorts raised in standard laboratory conditions. This simple problem solving task takes advantage of a rodent’s innate desire to avoid open enclosures by seeking shelter. Cognitive ability is assessed by adding increasingly complex impediments to the shelter’s entrance. The time a given subject takes to successfully remove the obstructions and enter the shelter serves as the primary metric for task performance. This method could provide a reliable means of rapidly assessing the efficacy of different enrichment protocols on cognitive function, thus paving the way for systematically determining the role specific environmental factors play in delaying the onset of neurodevelopmental and neurodegenerative disease.
El enriquecimiento ambiental (EE) se puede definir como un entorno que proporcionan los animales con mayor oportunidad para la interacción social, la actividad motora, y una mayor estimulación sensorial que por lo general con experiencia en un environment.EE estándar de laboratorio se ha demostrado que afecta sistemáticamente el comportamiento de los animales, provocando cambios tales como la reducción del estrés y la actividad relacionada con la ansiedad-8-10, la mejora de rendimiento en las tareas de aprendizaje y memoria 8,11, inicio temprano de la coordinación motora y la actividad exploratoria 11, los cambios en la atención materna 8 así como la resistencia a las sustancias adictivas 12- 15. Además, EE ha sido revelado para mejorar los efectos de los trastornos neurodegenerativos, retrasar la aparición y la disminución de la severidad de los síntomas en los modelos animales de la enfermedad de Huntington 1-4,16, 17 de Parkinson y la enfermedad de 18 Alzheimer.
Estos cambios correlacionarse con las alteraciones anatómicas y moleculares EE se sabe que induce en todo el cerebro. Actualmente, no hay ninguna tarea de comportamiento consistente y simple que puede ser usado para medir la efectividad de varios paradigmas de EE en diferentes cepas y specs de los animales.
La tarea Puzzle Box fue diseñado como una prueba sencilla para determinar problema nativo de un animal habilidad 7 resolver. Se requieren animales colocados en el área abierta para eliminar los materiales situados en una pequeña abertura para acceder a una región / refugio cubierto obstruir. Cada sujeto se administra tres ensayos con la misma obstrucción a fin de evaluar tres atributos cognitivos diferentes. El primer ensayo se obtiene un indicador de línea de base de la capacidad de resolución de problemas inherentes o nativo. El segundo ensayo, se ejecutan en el mismo día, le da una idea de la capacidad del animal para mejorar y así reforzar las estrategias para eliminar la obstrucción específico. El tercer ensayo, llevado a cabo en el día siguiente, da una idea de la capacidad del sujeto para retener y recordar la solución aprendido a la tarea.
La motivación de la resolución de estos rompecabezas "obstrucción" de los animales puede ser variado, potencialmenteevocando un deseo innato para evitar los campos abiertos y buscar refugio, así como un impulso inherente a explorar su entorno 6,7. La multitud de potenciales conductores de comportamiento que subyacen al deseo de resolver el Puzzle Box sugiere que varias áreas del cerebro están involucradas en la mediación de la ejecución de tareas.
