Ex vivo ERG can be used to record electrical activity of retinal cells directly from isolated intact retinas of animals or humans. We demonstrate here how common in vivo ERG systems can be adapted for ex vivo ERG recordings in order to dissect the electrical activity of retinal cells.
An In vivo electroretinogram (ERG) signal is composed of several overlapping components originating from different retinal cell types, as well as noise from extra-retinal sources. Ex vivo ERG provides an efficient method to dissect the function of retinal cells directly from an intact isolated retina of animals or donor eyes. In addition, ex vivo ERG can be used to test the efficacy and safety of potential therapeutic agents on retina tissue from animals or humans. We show here how commercially available in vivo ERG systems can be used to conduct ex vivo ERG recordings from isolated mouse retinas. We combine the light stimulation, electronic and heating units of a standard in vivo system with custom-designed specimen holder, gravity-controlled perfusion system and electromagnetic noise shielding to record low-noise ex vivo ERG signals simultaneously from two retinas with the acquisition software included in commercial in vivo systems. Further, we demonstrate how to use this method in combination with pharmacological treatments that remove specific ERG components in order to dissect the function of certain retinal cell types.
Elektroretinogrammet (ERG) er en veletablert teknikk som kan brukes til å registrere den elektriske aktiviteten i retina utløst av lys. ERG signal genereres i hovedsak av spenningsendringer forårsaket av radiale strømmer (langs aksen av fotoreseptorene og bipolare celler) som strømmer i den resistive ekstracellulære rom av netthinnen. Den første ERG signal ble registrert i 1865 av Holmgren fra overflaten av en fisk øye 1. Einthoven og Jolly 1908 2 delt ERG respons på angrep av lys i tre forskjellige bølger, kalt A-, B- og C-bølgene, som nå er kjent for å reflektere hovedsakelig aktiviteten av fotoreseptorer, ON bipolare celler, og pigment epitelet -celler, henholdsvis 3-8. ERG kan tas opp fra øynene til bedøvede dyr eller mennesker (in vivo), fra isolerte øye forberedelse 9, over isolert intakt retina (ex vivo) 3,10-15 eller i bestemte netthinnen lag med mikroelektroder (lokalERG) 4,16. Av disse er in vivo ERG i dag den mest brukte metoden for å vurdere retinal funksjon. Det er en ikke-invasiv teknikk som kan brukes for diagnostiske formål eller for å følge progresjonen av retinale sykdommer i dyr eller pasienter. Men in vivo ERG opptak produsere et komplisert signal med flere overlappende komponenter, ofte forurenset av extraocular fysiologiske støy (f.eks, pust og hjerteaktivitet).
Lokal ERG kan brukes til å registrere signalet i bestemte lag av netthinnen, men det er mest invasiv og har den laveste signal-til-støy-forhold (SNR) i forhold til de andre ERG opptaks konfigurasjoner. Lokal ERG er også teknisk krevende og krever kostbart utstyr (f.eks mikroskop og micromanipulators). Transretinal ERG fra intakt, isolert retina (ex vivo ERG) tilbyr et kompromiss mellom in vivo og lokale ERG metoder slik at stabil og HiGh SNR opptak fra intakte netthinne av dyr eller mennesker 17. Nylig har denne metoden vært brukt med hell for å studere stang og kjegle fotoreseptor funksjon i pattedyr, primater og mennesker netthinne 18-20. I tillegg, på grunn av fravær av pigment epitelet i ex vivo hinnen, blir den positive C-bølgekomponent av ERG signal fjernet og en fremstående negativ langsom PIII komponent er åpenbart i den ex vivo-opptak. Den langsomme PIII komponenten har vist seg å stamme fra aktiviteten av Muller gliaceller i retina 21-23. Dermed kan ex vivo ERG metoden også brukes til å studere Muller celler i intakt hinnen. Flere studier har også vist at ex vivo ERG opptakene kunne brukes til å måle konsentrasjonen av farmakologiske midler rundt retina 24 og teste sikkerhet og effekt av legemidler 25-27.
Multiple kommersiell in vivo systemer er tilgjengelig ogbrukes i mange laboratorier som ikke nødvendigvis har omfattende elektrobakgrunn. I kontrast, har ex vivo-enheter ikke vært tilgjengelig inntil nylig 17 og som et resultat bare svært få laboratorier drar nytte av dette kraftige teknikken. Det ville være gunstig å foreta ex vivo ERG opptak tilgjengelig for flere laboratorier for å fremme vår kunnskap om retinal fysiologi og patologi, og for å utvikle nye behandlingsformer for blendende sykdommer. Vi viser her en enkel og rimelig ex vivo ERG-enheten 17, og viser hvordan det kan brukes i kombinasjon med flere kommersielt tilgjengelige in vivo ERG-systemer for å registrere og stempelstang kjegle-mediert signalisering (A- og B-bølger), og funksjonen av Müller celler (bremse PIII) fra intakte villtype mus netthinne.
Vi viser her de viktige skritt for å oppnå høy kvalitet ex vivo ERG opptak samtidig fra to isolerte mus netthinne ved hjelp in vivo ERG systemkomponenter sammen med en ex vivo ERG adapter. I denne studien perfusert vi både netthinner fra dyret med den samme oppløsning (enten Ames ', Lockes eller Ringers), men det er også mulig å perfuse hver netthinnen med en annen løsning for eksempel for medikamenttestformål.
