To obtain basic information on the sorption and recycling of gold from aqueous systems the interaction of Au(III) and Au(0) nanoparticles on S-layer proteins were investigated. The sorption of protein polymers was investigated by ICP-MS and that of proteinaceous monolayers by QCM-D. Subsequent AFM enables the imaging of the nanostructures.
In this publication the gold sorption behavior of surface layer (S-layer) proteins (Slp1) of Lysinibacillus sphaericus JG-B53 is described. These biomolecules arrange in paracrystalline two-dimensional arrays on surfaces, bind metals, and are thus interesting for several biotechnical applications, such as biosorptive materials for the removal or recovery of different elements from the environment and industrial processes. The deposition of Au(0) nanoparticles on S-layers, either by S-layer directed synthesis 1 or adsorption of nanoparticles, opens new possibilities for diverse sensory applications. Although numerous studies have described the biosorptive properties of S-layers 2-5, a deeper understanding of protein-protein and protein-metal interaction still remains challenging. In the following study, inductively coupled mass spectrometry (ICP-MS) was used for the detection of metal sorption by suspended S-layers. This was correlated to measurements of quartz crystal microbalance with dissipation monitoring (QCM-D), which allows the online detection of proteinaceous monolayer formation and metal deposition, and thus, a more detailed understanding on metal binding.
The ICP-MS results indicated that the binding of Au(III) to the suspended S-layer polymers is pH dependent. The maximum binding of Au(III) was obtained at pH 4.0. The QCM-D investigations enabled the detection of Au(III) sorption as well as the deposition of Au(0)-NPs in real-time during the in situ experiments. Further, this method allowed studying the influence of metal binding on the protein lattice stability of Slp1. Structural properties and protein layer stability could be visualized directly after QCM-D experiment using atomic force microscopy (AFM). In conclusion, the combination of these different methods provides a deeper understanding of metal binding by bacterial S-layer proteins in suspension or as monolayers on either bacterial cells or recrystallized surfaces.
På grunn av den økende bruken av gull i flere applikasjoner som elektronikk, katalysatorer, biosensorer, eller medisinske instrumenter, har etterspørselen av dette edelt metall vokst de siste par års tid 6-9. Gull samt mange andre edle og tungmetaller slippes ut i miljøet via industrielle utslipp i fortynnede konsentrasjoner, gjennom gruvevirksomhet, og avfallshåndtering 7,8,10, selv om de fleste miljøforurensning av tunge eller edle metaller er en pågående prosess hovedsakelig forårsaket av teknologiske aktiviteter. Dette fører til en betydelig innblanding av naturlige økosystemer og potensielt kan true menneskers helse 9. Kjenner disse negative utfall fremmer jakten på nye teknikker for å fjerne metaller fra forurensede økosystemer og forbedringer i resirkulering metaller fra industrielt avløpsvann. Veletablerte fysikalsk-kjemiske metoder som utfelling eller ionebytting ikke er så effektiv, særlig i høyly utvannet løsninger 7,8,11. Biosorption, enten med levende eller død biomasse, er et attraktivt alternativ for rensing av avløpsvann 10,12. Bruken av slike biologiske materialer kan redusere forbruket av giftige kjemikalier. Mange mikroorganismer er blitt beskrevet til å akkumulere eller immobilisere metaller. For eksempel kan celler av Lysinibacillus sphaericus (L. sphaericus) JG-A12 er vist en høy bindingskapasitet for edle metaller, for eksempel, Pd (II), Pt (II), Au (III), og andre giftige metaller som Pb (II) eller U (VI) 4,13, celler av Bacillus megaterium for Cr (VI) 14, celler av Saccharomyces cerevisiae på Pt (II) og Pd (II) 15, og Chlorella vulgær for Au (III) og U (VI) 16 , 17. Bindingen av foregående metaller som Au (III), Pd (II), og Pt (II) har også blitt rapportert for Desulfovibrio desulfuricans 18 og for L. sphaericus JG-B53 19,20. Likevel, ikke all mikrober binde store mengder metaller og deres søknad som sorptive materialet er begrenset 12,21. Videre metall bindingsevne avhenger av forskjellige parametre, f.eks celle sammensetning, den brukes bio-komponent, eller miljømessige og eksperimentelle forhold (pH, ionestyrke, temperatur etc.). Studiet av isolerte celleveggfragmenter 22,23, som membranlipider, peptidoglykan, proteiner eller andre komponenter, hjelper til å forstå den metallbindende prosessene komplekse konstruerte hele celler 8,21.
Cellekomponenter fokusert på i denne studien er S-lags proteiner. S-lag protein er en del av den ytre celleveggen av mange bakterier og archaea, og de utgjør ca 15 – 20% av den totale proteinmasse i disse organismer. Som det første grensesnittet til omgivelsene, disse celle forbindelsene en sterk innvirkning på de bakterielle sorpsjons-egenskaper 3. S-lags proteiner med molekylvekter i området fra førtitil hundrevis av kDa produseres inne i cellen, men er montert utenfor der de er i stand til å danne sjikt på lipidmembraner eller polymere celleveggkomponenter. Når isolert, nesten alle S-lags proteiner har den iboende egenskap å spontant selv montere i suspensjon, på grensesnitt, eller på flater som danner plane eller tube-lignende strukturer 3. Tykkelsen av proteinet monolaget avhenger av bakterier, og er innenfor et område på 5 – 25-nm 24. Generelt kan de dannede S-lag protein strukturer har en skrå (p1 og p2), square (p4) eller sekskantet (p3 eller P6) symmetri med gitter-konstanter på 2,5 til 35 nm 3,24. Gitterdannelse synes å være i mange tilfeller avhengig av divalente kationer og hovedsakelig på Ca 2+ 25,26, Raff, J. et al. Ikke desto mindre, den fullstendige reaksjonen kaskade av monomer folding, monomer-monomer interaksjon, dannelse av et gitter, og rollen til forskjellige metaller, spesielt divalente kationer så som Ca2 + og Mg2 +, er fortsatt ikke fullstendig forstått.
Den gram-positive belastning L. sphaericus JG-B53 (omdøpt fra Bacillus sphaericus etter ny fylogenetisk klassifikasjon) 27 ble isolert fra uran mining avfallet hoper "Haberland" (Johann, Sachsen, Tyskland) 4,28,29. Dets funksjonelle S-lag protein (Slp1) besitter en kvadratisk gitter, en molekylvekt på 116 kDa 30, og en tykkelse på 10 nm på ≈ levende bakterieceller 31. I tidligere studier, ble den in vitro dannelse av en lukket og stabil protein lag med en tykkelse på omtrent 10 nm oppnås på mindre enn 10 min 19. Den relaterte belastninger L. sphaericus JG-A12, også et isolat fra "Haber" haug, har høy metall-bindingskapasitet og dens isolerte S-lag protein har vist en høy kjemisk og mekanisk stabilitet og god absorpsjon prisene for edelmetaller som Au (III), Pt (II), og Pd (II) 4,32,33. Denne bindingen av edle metaller er mer eller mindre spesifikk for noen metaller, og er avhengig av tilgjengeligheten av funksjonelle grupper på den ytre og indre protein overflaten av polymeren og i dets porer, ionestyrke og pH-verdien. Aktuelle funksjonelle grupper for metall interaksjon med proteiner er COOH, NH2 -, OH-, PO 4 -, SO 4 – og SO. I prinsippet metallbindende kapasitet åpne et bredt spekter av applikasjoner, Raff, J. et al. S-lag basert nanocomposites for industrielle applikasjoner i Protein-baserte Engineered nanostrukturer. Cortajarena) (Springer, 2016 (innsendt)). f.eks, som biosorptive komponenter for fjerning eller gjenvinningoppløste giftige eller verdifulle metaller, maler for syntese eller definert deponering av regelmessig strukturerte metalliske nanopartikler (NPS) for katalyse og andre bio-konstruerte materialer som bio-sensorisk lag 3,5,18,33. Jevnlig arrangert NP arrays som Au (0) -NPs kunne brukes til større applikasjoner som spenner fra molekylær elektronikk og biosensorer, ultrahøy tetthet lagringsenheter, og katalysatorer for CO-oksidasjon 34-37. Utviklingen av slike programmer og smart design av disse materialene krever en dypere forståelse av de underliggende metall bindende mekanismer.
En forutsetning for utviklingen av slike bio-baserte materialer er pålitelig gjennomføring av et grensesnittsjikt mellom biomolekylet og den tekniske overflaten 38,39. For eksempel, polyelektrolytter montert med lag-på-lag (LbL) 40,41 teknikken har blitt brukt som et grensesnittlag for omkrystallisering av S-lag protein 39 </sup>. Et slikt grensesnitt har en forholdsvis enkel måte å utføre den proteinbelegg på en reproduserbar og kvantitativ måte. Ved å utføre ulike eksperimenter med og uten endringer med selvklebende arrangører, er det mulig å lage uttalelser om belegg kinetikk, stabilitet lag og samspill av metaller med biomolekyler 19,42, Raff, J. et al. S-lag basert nanocomposites for industrielle applikasjoner i Protein-baserte Engineered nanostrukturer. (eds Tijana Z. Grove & Aitziber L. Cortajarena) (Springer, 2016 (innsendt)). Imidlertid er den komplekse mekanismen for protein adsorpsjon og protein-interaksjon overflate ikke er helt forstått. Spesielt informasjon om konformasjon, mønster orientering, og belegg tettheter er fortsatt savnet.
Kvartskrystall mikro med ødsling overvåking (QCM-D) teknikken har tiltrukket seg oppmerksomhet i de siste årene som et verktøy for å studere protein adsorpsjon, belegg kinetikk, og samhandling proprosesser på nanometerskala 19,43-45. Denne teknikken gjør det mulig for den detaljerte påvisning av masse adsorpsjon i sanntid, og kan benyttes som en indikator for proteinet selvsammen prosess og kobling av funksjonelle proteinmolekyler på gittere 19,20,42,46-48. I tillegg QCM-D-målinger åpne muligheten til å studere metall interaksjonsprosesser med det proteinholdige lag under naturlige biologiske forhold. I en fersk undersøkelse, samspillet av S-lag protein med utvalgte metaller som Eu (III), Au (III), Pd (II), og Pt (II) har blitt studert med QCM-D 19,20. Det adsorberte protein Laget kan tjene som en forenklet modell av en cellevegg av gram-positive bakterier. Studiet av denne enkeltkomponent kan bidra til en dypere forståelse av metall interaksjon. Men ikke utelukkende QCM-D eksperimenter ikke tillate uttalelser om overflatestrukturer og påvirkninger av metaller til protein. Andre teknikker er nødvendig for å oppnå slik informasjon. En poslighet for bildebehandling bio-nanostrukturer og innhente informasjon om strukturelle egenskaper er atomkraften mikroskopi (AFM).
Målet med den fremlagte studien var å undersøke sorpsjon av gull (Au (III) og Au (0) -NPs) til S-lag protein, spesielt Slp1 av L. sphaericus JG-B53. Forsøkene ble utført med proteiner suspendert i batch skala i et pH-område på 2,0 – 5,0 ved hjelp av ICP-MS og med immobiliserte S-lag ved hjelp av QCM-D. I tillegg, ble innflytelsen av metallsaltløsning på gitteret stabilitet undersøkt med påfølgende AFM studier. Kombinasjonen av disse teknikkene bidrar til en bedre forståelse av in vitro metall samhandlingsprosesser som et verktøy for å lære mer om forpliktende aktiviteter hele bakterieceller om konkrete metall slektskap. Denne kunnskapen er ikke bare avgjørende for utviklingen av gjeldende filtermaterialer for utvinning av metaller for miljøvern og bevaring av rekilder 49, men også for utvikling av matriser av svært bestilt metalliske NPs for ulike tekniske applikasjoner.
I dette arbeidet studert binding av Au til S-lag protein ble undersøkt ved hjelp av en kombinasjon av forskjellige analytiske metoder. Spesielt binding av Au er meget attraktivt, ikke bare for utvinning av Au fra gruvedrift farvann eller prosessløsninger, men også for bygging av materialer, f.eks sensoriske overflater.
