To obtain basic information on the sorption and recycling of gold from aqueous systems the interaction of Au(III) and Au(0) nanoparticles on S-layer proteins were investigated. The sorption of protein polymers was investigated by ICP-MS and that of proteinaceous monolayers by QCM-D. Subsequent AFM enables the imaging of the nanostructures.
In this publication the gold sorption behavior of surface layer (S-layer) proteins (Slp1) of Lysinibacillus sphaericus JG-B53 is described. These biomolecules arrange in paracrystalline two-dimensional arrays on surfaces, bind metals, and are thus interesting for several biotechnical applications, such as biosorptive materials for the removal or recovery of different elements from the environment and industrial processes. The deposition of Au(0) nanoparticles on S-layers, either by S-layer directed synthesis 1 or adsorption of nanoparticles, opens new possibilities for diverse sensory applications. Although numerous studies have described the biosorptive properties of S-layers 2-5, a deeper understanding of protein-protein and protein-metal interaction still remains challenging. In the following study, inductively coupled mass spectrometry (ICP-MS) was used for the detection of metal sorption by suspended S-layers. This was correlated to measurements of quartz crystal microbalance with dissipation monitoring (QCM-D), which allows the online detection of proteinaceous monolayer formation and metal deposition, and thus, a more detailed understanding on metal binding.
The ICP-MS results indicated that the binding of Au(III) to the suspended S-layer polymers is pH dependent. The maximum binding of Au(III) was obtained at pH 4.0. The QCM-D investigations enabled the detection of Au(III) sorption as well as the deposition of Au(0)-NPs in real-time during the in situ experiments. Further, this method allowed studying the influence of metal binding on the protein lattice stability of Slp1. Structural properties and protein layer stability could be visualized directly after QCM-D experiment using atomic force microscopy (AFM). In conclusion, the combination of these different methods provides a deeper understanding of metal binding by bacterial S-layer proteins in suspension or as monolayers on either bacterial cells or recrystallized surfaces.
Als gevolg van het toenemende gebruik van goud voor verschillende toepassingen, zoals elektronica, katalysatoren, biosensoren, of medische instrumenten, de vraag van dit edelmetaal is gegroeid in de afgelopen paar jaar tijd 6-9. Goud evenals vele andere edelstenen en zware metalen in het milieu via industrieel afvalwater in verdunde concentraties, door middel van mijnbouw, en afvalverwerking 7,8,10, hoewel de meeste milieuvervuiling door zware of edele metalen is een continu proces vooral veroorzaakt door de technologische activiteiten. Dit leidt tot een aanzienlijke verstoring van de natuurlijke ecosystemen en zou kunnen bedreigen de volksgezondheid 9. Weten deze negatieve uitkomsten bevordert het zoeken naar nieuwe technieken om metalen uit vervuilde ecosystemen en verbeteringen te verwijderen in de recycling van metalen uit industrieel afvalwater. Gevestigde fysisch-chemische methoden zoals neerslaan of ionenwisseling zijn niet zo effectief, vooral in de hogely verdunde oplossingen 7,8,11. Biosorptie, hetzij met levende of dode biomassa, is een aantrekkelijk alternatief voor de behandeling van afvalwater 10,12. Het gebruik van dergelijke biologische materialen kan het gebruik van giftige chemicaliën verminderen. Veel micro-organismen zijn beschreven ophopen of immobiliseren metalen. Bijvoorbeeld, cellen Lysinibacillus sphaericus (L. sphaericus) JG-A12 toonden hoge bindingscapaciteiten van edele metalen, bijvoorbeeld Pd (II), Pt (II), Au (III) en andere toxische metalen zoals Pb (II) of U (VI) 4,13, cellen van Bacillus megaterium van Cr (VI) 14, cellen van Saccharomyces cerevisiae van Pt (II) en Pd (II) 15 en Chlorella ordinaire voor Au (III) en U (VI) 16 , 17. De binding van de voorgaande metalen zoals Au (III), Pd (II) en Pt (II) is eveneens gerapporteerd Desulfovibrio desulfuricans 18 en L. sphaericus JG-B53 19,20. Toch niet all microben binden grote hoeveelheden metalen en hun toepassing als sorptieve materiaal is beperkt 12,21. Bovendien metaal bindingscapaciteit is afhankelijk van verschillende parameters, bijvoorbeeld celsamenstelling, de gebruikte bio-component, of milieu- en experimentele condities (pH, ionsterkte, temperatuur etc.). De studie van geïsoleerde celwandfragmenten 22,23, zoals membraanlipiden, peptidoglycaan, eiwitten of andere bestanddelen, helpt metaalbindingseigenschappen processen complex geconstrueerde gehele cellen 8,21 begrijpen.
De celcomponenten gericht in deze studie zijn S-layer eiwitten. S-layer eiwitten delen van de buitenste celwand van vele bacteriën en archaea, en zij vormen ongeveer 15 – 20% van de totale proteïnemassa van deze organismen. Als eerste interface naar de omgeving, deze mobiele verbindingen sterk beïnvloeden de bacteriële sorptie-eigenschappen 3. S-layer eiwitten met molecuulgewichten variërend van veertigtot honderden kDa wordt geproduceerd in de cel, maar buiten gemonteerd waar ze kunnen lagen op lipidemembranen of polymere celwand componenten vormen. Eenmaal geïsoleerd, bijna alle S-laag eiwitten hebben de intrinsieke eigenschap om spontaan zichzelf assembleren in suspensie, aan interfaces, of op oppervlakken vormen vlakke of buis-achtige structuren 3. De dikte van het eiwit monolaag afhankelijk van de bacteriën en ligt binnen een bereik van 5 – 25 nm 24. In het algemeen kan de gevormde S-layer eiwitstructuren een schuine (p1 en p2), vierkant (p4) of zeshoekig (p3 of p6) symmetrie roosterconstanten 2,5 tot 35 nm 3,24 hebben. Het rooster formatie lijkt in veel gevallen afhankelijk van tweewaardige kationen en voornamelijk Ca 2 + 25,26, Raff, J. et al. S-laag op basis van nanocomposieten voor industriële toepassingen op basis van eiwitten Engineered nanostructuren. (eds Tijana Z. Grove & Aitziber L. Cortajarena) (Springer, 2016 (ingediend)). Toch is de volledige reactie cascade monomeer vouwen, monomeer-monomeer interactie de vorming van een rooster, en de rol van verschillende metalen, vooral van divalente kationen zoals Ca2 + en Mg2 +, zijn nog niet volledig begrepen.
De gram-positieve stam L. sphaericus JG-B53 (hernoemd van Bacillus sphaericus na nieuwe fylogenetische classificatie) 27 werd geïsoleerd uit de uraniumwinning afval stapel "Haberland" (Johanngeorgenstadt, Saksen, Duitsland) 4,28,29. De functionele S-layer-eiwit (SLP1) bezit een vierkant rooster, een molecuulgewicht van 116 kDa 30 en een dikte van ≈ 10 nm op levende bacteriecellen 31. In eerdere onderzoeken werd de in vitro vorming van een gesloten en stabiel eiwit met een dikte van ongeveer 10 nm in minder dan 10 min 19 bereikt. De gerelateerde stam L. sphaericus JG-A12, ook een isolaat uit de "Haberland" pile, bezit hoge metaal binding capaciteiten, geïsoleerde S-layer-eiwit blijkt een hoge chemische en mechanische stabiliteit en goede sorptie bekijken van edelmetalen zoals Au (III), Pt (II), en Pd (II) 4,32,33. Deze binding van edelmetalen meer of minder specifiek voor bepaalde metalen en afhankelijk van de beschikbare functionele groepen op de buitenste en binnenste eiwit oppervlak van het polymeer en in zijn poriën, ionsterkte en pH. Relevante functionele groepen voor metalen interactie door de eiwitten COOH, NH 2 -, OH, PO 4 -, SO 4 – en SO. In principe, metaal bindende capaciteiten opent een breed spectrum van toepassingen, Raff, J. et al. S-laag op basis van nanocomposieten voor industriële toepassingen op basis van eiwitten Engineered nanostructuren. (eds Tijana Z. Grove & Aitziber L. Cortajarena) (Springer, 2016 (ingediend)). bv als biosorptive componenten voor de verwijdering of het herstelopgeloste toxische of waardevolle metalen, sjablonen voor synthese of gedefinieerd afzetting van regelmatig gestructureerde metalen nanodeeltjes (NP) voor katalyse en andere bio-engineered materialen zoals bio-zintuiglijke lagen 3,5,18,33. Regelmatig gerangschikte NP arrays zoals Au (0) -NPs zouden kunnen worden voor belangrijke toepassingen gaande van moleculaire elektronica en biosensoren, ultrahoge opslagdichtheid apparaten en katalysatoren voor CO-oxidatie 34-37. De ontwikkeling van dergelijke toepassingen en slimme ontwerp van deze materialen vereist een dieper begrip van het onderliggende metaal bindende mechanismen.
Een voorwaarde voor de ontwikkeling van dergelijke materialen op biologische basis is de betrouwbare uitvoering van een tussenlaag tussen het biomolecuul en de technische oppervlak 38,39. Bijvoorbeeld, polyelektrolyten samengevoegd met de laag-voor-laag (LbL) techniek 40,41 zijn gebruikt als interface laag voor herkristallisatie van S-layer eiwitten 39 </sup>. Dergelijke interface biedt een relatief eenvoudige manier de proteïne bekleding te voeren op een reproduceerbare en kwantitatieve wijze. Door uitvoeren van verschillende experimenten met en zonder modificatie met lijm promoters, is het mogelijk om uitspraken betreffende deklaag kinetiek, layer stabiliteit en interacties van metalen met biomoleculen 19,42, Raff, J. et al maken. S-laag op basis van nanocomposieten voor industriële toepassingen op basis van eiwitten Engineered nanostructuren. (eds Tijana Z. Grove & Aitziber L. Cortajarena) (Springer, 2016 (ingediend)). Echter, het complex mechanisme van eiwitadsorptie en interactie eiwit-oppervlak niet volledig begrepen. Vooral informatie over de conformatie, patroon oriëntatie en coating dichtheden wordt nog steeds vermist.
Kwartskristalmicrobalans met dissipatie bewaking (QCM-D) techniek heeft de aandacht getrokken in de afgelopen jaren als een instrument voor het bestuderen eiwitadsorptie, coating kinetiek en interactie proprocessen op de nanometerschaal 19,43-45. Deze techniek maakt het mogelijk om gedetailleerde detectie van massa adsorptie in real-time, en kan worden gebruikt als indicator voor het eiwit zelfassemblerende proces op proteïne roosters 19,20,42,46-48 koppeling van functionele moleculen. Daarnaast QCM-D metingen opent de mogelijkheid om metalen interactie processen te bestuderen met de proteïneachtige laag onder natuurlijke biologische omstandigheden. In een recente studie, de interactie van de S-layer-eiwit met bepaalde metalen zoals Eu (III), Au (III), Pd (II) en Pt (II) is onderzocht met QCM-D 19,20. Het geadsorbeerde eiwit laag kan dienen als een vereenvoudigd model van een celwand van Gram-positieve bacteriën. Het onderzoek van deze ene component kan bijdragen tot een beter begrip van metaal interactie. Echter, alleen QCM-D experimenten geen uitspraken over oppervlaktestructuren en invloeden van metalen eiwit mogelijk te maken. Andere technieken zijn nodig om dergelijke informatie te verkrijgen. Een poswoordelijkheid voor bio-imaging nanostructuren en verkrijgen van informatie over structurele eigenschappen is de atomaire kracht microscopie (AFM).
De doelstelling van dit onderzoek was om de sorptie van goud (Au (III) en Au (0) -NPs) S-layer eiwitten te onderzoeken, met name SLP1 van L. sphaericus JG-B53. Experimenten werden uitgevoerd met charge gesuspendeerd eiwitten op schaal in een pH traject van 2,0 – 5,0 behulp van ICP-MS en geïmmobiliseerd S-lagen gebruikt QCM-D. Bovendien is de invloed van metaalzoutoplossing aan het rooster stabiliteit onderzocht latere AFM studies. De combinatie van deze technieken draagt bij aan een beter begrip van de in vitro metaal interactie processen als een instrument voor het leren meer over bindende gebeurtenissen op de hele bacteriecellen over specifieke metalen affiniteiten. Deze kennis is niet alleen cruciaal voor de ontwikkeling van de toepasselijke filter materialen voor de recycling van metalen voor de bescherming van het milieu en het behoud van de rebronnen 49, maar ook voor de ontwikkeling van arrays van sterk geordende metalen NP voor diverse technische toepassingen.
In dit werk onderzochte de binding van Au S-laag-eiwitten werd onderzocht met behulp van een combinatie van verschillende analysemethoden. Vooral de binding van Au is zeer aantrekkelijk, niet alleen voor de terugwinning van Au uit de mijnbouw water of procesoplossingen, maar ook voor de constructie van materialen, bijvoorbeeld sensorische oppervlakken. Voor studies van de interactie Au (Au (III) en Au (0) -NPs) met onderbroken en herkristalliseerd monolaag van SLP1 het eiwit moesten worden geïsoleerd. Daarom h…
