To obtain basic information on the sorption and recycling of gold from aqueous systems the interaction of Au(III) and Au(0) nanoparticles on S-layer proteins were investigated. The sorption of protein polymers was investigated by ICP-MS and that of proteinaceous monolayers by QCM-D. Subsequent AFM enables the imaging of the nanostructures.
In this publication the gold sorption behavior of surface layer (S-layer) proteins (Slp1) of Lysinibacillus sphaericus JG-B53 is described. These biomolecules arrange in paracrystalline two-dimensional arrays on surfaces, bind metals, and are thus interesting for several biotechnical applications, such as biosorptive materials for the removal or recovery of different elements from the environment and industrial processes. The deposition of Au(0) nanoparticles on S-layers, either by S-layer directed synthesis 1 or adsorption of nanoparticles, opens new possibilities for diverse sensory applications. Although numerous studies have described the biosorptive properties of S-layers 2-5, a deeper understanding of protein-protein and protein-metal interaction still remains challenging. In the following study, inductively coupled mass spectrometry (ICP-MS) was used for the detection of metal sorption by suspended S-layers. This was correlated to measurements of quartz crystal microbalance with dissipation monitoring (QCM-D), which allows the online detection of proteinaceous monolayer formation and metal deposition, and thus, a more detailed understanding on metal binding.
The ICP-MS results indicated that the binding of Au(III) to the suspended S-layer polymers is pH dependent. The maximum binding of Au(III) was obtained at pH 4.0. The QCM-D investigations enabled the detection of Au(III) sorption as well as the deposition of Au(0)-NPs in real-time during the in situ experiments. Further, this method allowed studying the influence of metal binding on the protein lattice stability of Slp1. Structural properties and protein layer stability could be visualized directly after QCM-D experiment using atomic force microscopy (AFM). In conclusion, the combination of these different methods provides a deeper understanding of metal binding by bacterial S-layer proteins in suspension or as monolayers on either bacterial cells or recrystallized surfaces.
På grund av den ökande användningen av guld för flera applikationer som elektronik, katalysatorer, biosensorer, eller medicinska instrument, har efterfrågan på denna ädelmetall ökat under de senaste åren "tid 6-9. Guld liksom många andra ädelstenar och tungmetaller släpps ut i miljön via industriutsläpp i utspädda koncentrationer, genom gruvdrift och avfallshantering 7,8,10, även om de flesta miljöförorening av tunga eller ädelmetaller är en pågående process orsakas huvudsakligen av teknisk verksamhet. Detta leder till en betydande störning av naturliga ekosystem och skulle kunna hota människors hälsa 9. Att känna till dessa negativa utfall främjar sökandet efter nya tekniker för att avlägsna metaller från förorenade ekosystem och förbättringar återvinna metaller ur industriellt avloppsvatten. Väletablerade fysikalisk-kemiska metoder såsom utfällning eller jonbyte är inte så effektiva, särskilt i högtly utspädda lösningar 7,8,11. Biosorption, antingen levande eller död biomassa, är ett attraktivt alternativ för rening av avloppsvatten 10,12. Användningen av sådana biologiska material kan minska förbrukningen av giftiga kemikalier. Många mikroorganismer har beskrivits att ackumulera eller immobilisera metaller. Till exempel, celler av Lysinibacillus sphaericus (L. sphaericus) JG-A12 har visat höga bindande kapacitet för ädelmetaller, t ex Pd (II), Pt (II), Au (III), och andra giftiga metaller som Pb (II) eller U (VI) 4,13, celler av Bacillus megaterium för Cr (VI) 14, celler av Saccharomyces cerevisiae för Pt (II) och Pd (II) 15, och Chlorella vulgärt för Au (III) och U (VI) 16 17. Bindningen av föregående metaller som Au (III), Pd (II), och Pt (II) har också rapporterats för Desulfovibrio desulfuricans 18 och för L. sphaericus JG-B53 19,20. Ändå inte all mikrober binder stora mängder metaller och deras tillämpning som sorberande material är begränsad 12,21. Vidare metallbindande kapacitet beror på olika parametrar, t.ex. cellkomposition, den använda bio-komponent eller miljömässiga och experimentella förhållanden (pH, jonstyrka, temperatur etc.). Studien av isolerade cellväggsfragment 22,23, som membranlipider, peptidoglykan, proteiner eller andra komponenter, hjälper till att förstå den metallbindande processer av komplexa konstruerade hela celler 8,21.
Cellkomponenterna fokuserade på i denna studie är S-skiktproteiner. S-skiktproteiner är delar av det yttre cellhölje av många bakterier och arkéer, och de utgör ca 15-20% av den totala proteinmassan av dessa organismer. Som det första gränssnittet för miljön, dessa cell föreningar starkt påverkar bakterie sorptionsegenskaper 3. S-skiktproteiner med molekylvikter som sträcker sig från fyrtiotill hundratals kDa produceras inuti cellen, men är monterade utanför där de kan bilda skikt på lipidmembran eller polymercellväggskomponenter. När isolerade, nästan alla S-skiktproteiner har den inneboende egenskapen att spontant själv montera i suspension, vid gränsytor, eller på ytor som bildar plana eller rörliknande strukturer 3. Tjockleken på proteinmonoskikt beror på bakterierna och är inom ett intervall av fem – 25 nm 24. I allmänhet kan de formade S-skiktproteinstrukturer har en sned (P1 eller P2), fyrkantig (p4), eller hexagonala (p3 eller p6) symmetri med gitterkonstanter på 2,5 till 35 nm 3,24. Den gallerbildning verkar vara i många fall beroende av divalenta katjoner och främst på Ca2 + 25,26, Raff, J. et al. S-lager baserade nanokompositer för industriapplikationer i Protein baserade Engineered nanostrukturer. (eds Tijana Z. Grove & Aitziber L. Cortajarena) (Springer, 2016 (lämnas)). Icke desto mindre, den fullständiga reaktionskaskad av monomer vikning, interaktion monomer-monomer, bildandet av ett gitter, och betydelsen av olika metaller, i synnerhet av divalenta katjoner, såsom Ca2 + och Mg2 +, är ännu inte helt klarlagda.
Den grampositiva stam L. sphaericus JG-B53 (bytt namn från Bacillus sphaericus efter nya fylogenetiska klassificeringen) 27 isolerades från uranbrytning avfallet högen "Haberland" (Johann, Sachsen, Tyskland) 4,28,29. Dess funktionella S-skikt protein (Slp1) besitter en kvadrat galler, en molekylvikt på 116 kDa 30, och en tjocklek på ≈ 10 nm på levande bakterieceller 31. I tidigare studier har bildningen in vitro av en sluten och stabilt protein-skikt med en tjocklek av ca 10 nm uppnås på mindre än 10 minuter 19. Den relaterade stammen L. sphaericus JG-A12, även ett isolat från "Haberland" högen, har hög metallbindningskapaciteter och dess isolerade S-skiktet proteinet har visat en hög kemisk och mekanisk stabilitet och goda sorptionsegenskaper priserna för ädelmetaller som Au (III), Pt (II), och Pd (II) 4,32,33. Denna bindning av ädelmetaller är mer eller mindre specifika för vissa metaller och beror på tillgängligheten av funktionella grupper på den yttre och inre protein ytan av polymeren och i dess porer, jonstyrka och pH-värdet. Relevanta funktionella grupper för interaktion metall av proteinerna är COOH, NH2 -, OH, PO 4 -, SO 4 – och SO-. I princip, metallbindande kapaciteten öppnar ett brett spektrum av tillämpningar, Raff, J. et al. S-lager baserade nanokompositer för industriapplikationer i Protein baserade Engineered nanostrukturer. (eds Tijana Z. Grove & Aitziber L. Cortajarena) (Springer, 2016 (lämnas)). t.ex. som biosorptive komponenter för avlägsnande eller återvinningav lösta giftiga eller värdefulla metaller, mallar för syntes eller definierade avsättning regelbundet strukturerade metalliska nanopartiklar (NPS) för katalys, och andra biologiskt konstruerade material som bio-sensoriska skikten 3,5,18,33. Regelbundet arrangeras NP arrayer som Au (0) -NPs skulle kunna användas för stora tillämpningar som sträcker sig från molekylär elektronik och biosensorer, ultrahög lagringskapacitet enheter och katalysatorer för CO-oxidation 34-37. Utvecklingen av sådana ansökningar och smart design av dessa material kräver en djupare förståelse av de underliggande metall bindande mekanismer.
En förutsättning för utvecklingen av sådana biobaserade material är tillförlitlig genomförandet av ett gränsskikt mellan biomolekylen och den tekniska ytan 38,39. Exempelvis polyelektrolyter monterad med lager-för-skikt (LBL) teknik 40,41 har använts som ett gränsskikt för omkristallisation av S-skiktproteiner 39 </sup>. Ett sådant gränssnitt erbjuder ett relativt enkelt sätt att utföra proteinbeläggningen på ett reproducerbart och kvantitativt sätt. Genom att utföra olika experiment med och utan ändringar med självhäftande promotorer, är det möjligt att göra uttalanden om beläggnings kinetik, lager stabilitet och samverkan mellan metaller med biomolekyler 19,42, Raff, J. et al. S-lager baserade nanokompositer för industriapplikationer i Protein baserade Engineered nanostrukturer. (eds Tijana Z. Grove & Aitziber L. Cortajarena) (Springer, 2016 (lämnas)). Emellertid är den komplexa mekanismen för proteinadsorption och interaktion protein-yta inte fullständigt klarlagd. Speciellt information om konforma, mönster orientering och beläggningstäthet saknas fortfarande.
Kvartskristallmikrovåg med förlustövervakning (QCM-D) teknik har uppmärksammats de senaste åren som ett verktyg för att studera proteinadsorption, beläggnings kinetik och interaktion proprocesser på nanometerskala 19,43-45. Denna teknik gör det möjligt att den detaljerade detektering av mass adsorption i realtid, och kan användas som en indikator för proteinsjälvsammansättningsprocessen och koppling av funktionella molekyler på proteingitter 19,20,42,46-48. Dessutom QCM-D mätningar öppnar möjlighet att studera interaktionsmetallprocesser med det proteinskikt under naturliga biologiska förhållanden. I en nyligen genomförd studie, interaktionen av S-skiktet protein med utvalda metaller såsom Eu (III), Au (III), Pd (II), och Pt (II) har studerats med QCM-D 19,20. Det adsorberade proteinskikt kan tjäna som en förenklad modell av en cellvägg av grampositiva bakterier. Studiet av denna enda komponent kan bidra till en djupare förståelse av interaktion metall. Dock inte enbart QCM-D experiment tillåter inte påståenden om strukturer och influenser av metaller till protein yta. Andra tekniker är nödvändiga för att erhålla sådan information. En postagande för avbildning bio-nanostrukturer och få information om strukturella egenskaper är atomkraftsmikroskop (AFM).
Målet för den presenterade studien var att undersöka sorption av guld (Au (III) och Au (0) -NPs) till S-skiktproteiner, särskilt Slp1 L. sphaericus JG-B53. Experiment gjordes med suspenderade proteiner på satsskala i ett pH-intervall av 2,0-5,0 med hjälp av ICP-MS och med immobiliserade S-skikt med användning av QCM-D. Dessutom var påverkan av metallsaltlösningen på gitterstabilitet undersöktes med efterföljande AFM studier. Kombinationen av dessa tekniker bidrar till en bättre förståelse av in vitro interaktionsmetallprocesser som ett verktyg för att lära sig mer om bindande händelser hela bakterieceller avseende specifika metall tillhörighet. Denna kunskap är inte bara avgörande för utvecklingen av tillämpliga filtermaterial för återvinning av metaller för miljöskydd och bevarande av rekällor 49, men också för att utveckla uppsättningar av högt ordnade metall NP för olika tekniska tillämpningar.
I detta arbete studerades bindningen av Au till S-skiktproteiner undersöktes med användning av en kombination av olika analysmetoder. I synnerhet bindningen av Au är mycket attraktiv inte bara för återvinning av Au från gruvvatten eller processlösningar, men också för byggandet av material, t.ex. sensoriska ytor.
