Most bacterial infections produce a biofilm. By virtue of their environment, biofilm associated bacteria are often phenotypically drug resistant. Novel antibacterial molecules that kill bacteria in biofilms are thus a high priority. We establish an assay to quickly screen for antimicrobial compounds that are effective at eradicating biofilms.
Most pathogenic bacteria are able to form biofilms during infection, but due to the difficulty of manipulating and assessing biofilms, the vast majority of laboratory work is conducted with planktonic cells. Here, we describe a peg plate biofilm assay as performed with Staphylococcus aureus. Bacterial biofilms are grown on pegs attached to a 96-well microtiter plate lid, washed through gentle submersion in buffer, and placed in a drug challenge plate. After subsequent incubation they are again washed and moved to a final recovery plate, in which the fluorescent dye resazurin serves as a viability indicator. This assay offers greatly increased ease-of-use, reliability, and reproducibility, as well as a wealth of data when conducted as a kinetic read. Moreover, this assay can be adapted to a medium-throughput drug screening approach by which an endpoint fluorescent readout is taken instead, offering a path for drug discovery efforts.
Pathogene bacteriën kunnen biofilms in vivo leidt tot niet-acute chronische infecties 1 vormen. Biofilm geassocieerde infectie is een ernstige risicofactor van medische procedures die de implantatie van vreemde voorwerpen (bijvoorbeeld kunstbot vervangingen, borstimplantaten) of de installatie van tracheale buizen of urine-katheters 2 te betrekken. In deze context, anti-infectieuze therapie is vrijwel altijd nodig als biofilm-gebaseerde infecties zelden gewist als zodanig, zelfs bij immuuncompetente individuen. Staphylococcus aureus is een van de meest voorkomende pathogenen betrokken in biofilm-gerelateerde complicaties optreden gedurende het gebruik van invasieve medische hulpmiddelen 3.
Helaas, de aard van biofilms als een beschermende barrière maakt ze beter bestand zijn tegen de behandeling dan plankton cellen 1,4, en de evaluatie van de voorspelde klinische werkzaamheid is een cruciaal onderdeel van zowel de initiëlegeneesmiddelontwikkeling en resistentie surveillance. Er is steeds meer erkenning dat laboratoriumomstandigheden, gericht op planktonische culturen, mag niet getrouw vertegenwoordigen real-world ziekte 5. Verdere compounding het probleem, replicatie van biofilm fenotypes is moeilijk, en de bestaande biofilm modellen zijn vervelend en lijden aan hoge inter- en intra-assay variabiliteit 6. Zo veel onderzoekers, noodgedwongen uit standaard planktonische celbepalingen drug gevoeligheid, waardoor mogelijk dat daarbij een belangrijk aspect van bacteriële virulentie en ziekte.
Hier beschrijven we het protocol voor het testen van bacteriën, in het bijzonder S. aureus, in pre-biofilms gegroeid onder toepassing van een 96 putjes gebaseerde biofilmsysteem 7,8. Terwijl het protocol voor de vorming en uitdaging biofilm volgt in wezen de aanbeveling van de fabrikant, presenteren we een alternatieve informatie-rijke methodologie voor het kwantificeren van de levensvatbaarheid van de BIOFilm na challenge. Kort bacteriën worden gekweekt in de penplaat, waarbij biofilms gevormd op de uitstekende pennen bevestigd aan het deksel van de plaat. Na de vorming van biofilm, worden de haringen zachtjes gedoopt in putjes van een nieuwe plaat gevuld met PBS om plankton cellen te verwijderen. De peg-deksel, met biofilms bevestigd, wordt overgebracht naar een nieuwe uitdaging plaat gemaakt met verschillende concentraties antibiotica te testen. Na een tweede incubatie worden deksels weer verwijderd, gewassen en overgebracht naar een plaat die herstel Resazurin kleurstof, waar een laatste incubatie. Resazurin conversie kan kinetisch worden opgenomen of genomen als een eindpunt lezen na een bepaalde herstelperiode. Deze dye-gebaseerde methode voor het kwantificeren van de levensvatbaarheid van biofilms verschilt aanzienlijk van de vervelende CFU (kolonievormende eenheden) count-gebaseerde methode in het oorspronkelijke protocol 7 beschreven. OD 600 metingen van de drug uitdaging bord en resazurin conversie kinetiek dienen als levensvatbaarheid gelezenouts van planktonische en biofilm cellen, respectievelijk, het aanbieden van een snelle, betrouwbare, informatie-rijke en technisch eenvoudige test voor biofilm te overleven.
Hier beschrijven we een gemodificeerde biofilm gekwantificeerd door activiteit van geteste remmers op S. aureus biofilms gericht op de metabole toestand van biofilm geassocieerde cellen. Terwijl de beschreven biofilm initiatie en beroepsprocedures veelal nagebootste aanbevelingen van de fabrikant, het gebruik van Resazurin kleurstof-biofilm geassocieerde cellen die een 24-uurs-remmer uitdaging overleven detecteren en te kwantificeren vereenvoudigt volgens de methode van de cel herstel (via waterbad sonificatie)…
