We adapted a permeable microporous membrane insert to mimic the tumor microenvironment (TME). The model consists of a mixed cell culture, allows simplified generation of highly enriched individual cell populations without using fluorescent tagging or cell sorting, and permits studying intercellular communication within the TME under normal or stress conditions.
Inzicht in de vroege heterotypische interactie tussen kankercellen en het omliggende niet-kanker stroma is belangrijk bij het ophelderen van de gebeurtenissen die tot stromale activatie en vestiging van de tumor micro-omgeving (TME). Verscheidene in vitro en in vivo modellen van de TME ontwikkeld; In het algemeen deze modellen niet gemakkelijk toe isolatie van afzonderlijke celpopulaties onder niet-storende omstandigheden voor verdere studie. Om dit probleem te omzeilen, hebben wij een in vitro TME model toegepast met een celgroei substraat bestaande uit een permeabele microporeuze membraan inzetstuk toelaat eenvoudige generatie hoog verrijkte celpopulaties innig gekweekt, maar afzonderlijk, aan weerszijden van het membraan het inzetstuk voor uitgebreide co -cultuur tijden. Door het gebruik van dit model, we zijn in staat om het genereren van sterk verrijkt met kanker geassocieerde fibroblast (CAF) populaties van normale diploïde humane fibroblasten volgendeco-kweek (120 uur) met zeer metastatische humane borstcarcinoom cellen, zonder het gebruik van fluorescerende tagging en / of celsortering. Bovendien, door het moduleren van de poriegrootte van het inzetstuk kunnen we controleren voor de wijze van intercellulaire communicatie (bijvoorbeeld, gap-junction communicatie uitgescheiden factoren) tussen de twee heterotypische celpopulaties, die onderzoek naar de mechanismen verantwoordelijk voor de ontwikkeling van de toestaat TME en de rol van gap-junction permeabiliteit. Dit model dient als waardevol instrument beter inzicht van de initiële gebeurtenissen die leiden tot kanker-stroma initiatie, de vroege ontwikkeling van het TME en de modulerende werking van de stroma van de reacties van kankercellen therapeutische middelen.
De tumor micro-omgeving (TME) is een zeer complex systeem omvat carcinoomcellen die naast elkaar bestaan en evolueren naast gastheer stroma. Deze stromale component bestaat typisch uit fibroblasten, myofibroblasten, endotheelcellen, verschillende immune componenten, en een extracellulaire matrix 1. Een belangrijk bestanddeel, vaak de meerderheid van dit stroma, geactiveerd fibroblasten, vaak aangeduid als kanker geassocieerde fibroblasten of-carcinoom geassocieerde fibroblasten (CAF) 2,3. In tegenstelling tot normale, niet-geactiveerde fibroblasten, cafe's bijdragen aan tumor initiatie, progressie, angiogenese, invasie, metastase en herhaling 4-11 in diverse carcinomen, zoals borst, prostaat, longen, pancreas, huid, darm, slokdarm, en eierstok 5,6,12-17. Toch blijft de precieze aard van de bijdrage van cafe's de hele kanker pathogenese slecht gedefinieerd. Voorts heeft klinische aanwijzingen voorspellende waarde van cafe's aangetoond, Correleren hun aanwezigheid aan high-grade maligniteiten, therapie falen, en over het algemeen een slechte prognose 10,18,19.
Het is duidelijk dat het verbeteren van ons begrip van de initiërende gebeurtenissen in CAF ontwikkeling, evenals de intercellulaire communicatie bemiddelen hun rol binnen de TME, kan opwindende nieuwe therapeutische targets en verbeterde strategieën die de patiënt resultaten kunnen verbeteren. Teneinde dit doel zijn verscheidene in vivo en in vitro modellen ontwikkeld. Terwijl in vivo benaderingen zijn meer een afspiegeling is van TME patiënten, ze bezitten beperkingen, met inbegrip van de enorme complexiteit en de heterogeniteit binnen en tussen tumoren. Bovendien tumormonsters van proefpersonen vertegenwoordigen vaak hoogontwikkelde TME en geen inzicht in de TME initiërende gebeurtenissen niet toe. Experimentele dierstudies bieden een aantal voordelen, maar veralgemening van gegevens over dieren naar de mens moet worden gedaan met de nodige voorzichtigheid te wijten aan verschillen in physiology tussen mensen en dieren zoals knaagdieren (bijvoorbeeld thiol chemie 20, metabolisme 21, stresstolerantie 22, etc.). Verder in tegenstelling tot de menselijke populatie, die genetisch heterogeen van aard, proefdieren worden typisch gekweekt tot homogeniteit. Ook is het vaak moeilijk om voorbijgaande fysiologische variaties en wijzigingen celfenotype onderzoeken, en om te controleren voor specifieke experimentele parameters met dieren zoals knaagdieren. Dus, in vitro 2- en 3-dimensionale (2D en 3D) weefselkweek modellen worden vaak gebruikt om de basiskennis van TME ontwikkeling bevorderen. Ondanks het ontbreken van een nauwkeurige weergave van de complexiteit van in vivo systemen, deze modellen bieden vele voordelen mechanistische onderzoeken aanzienlijk vergemakkelijken. In vitro apparaten kan een eenvoudiger, gerichte en kosteneffectieve analyse van TME, waarbij statistisch significante gegevens kunnen worden gegenereerdcellen vrij van systemische variaties die zich voordoen bij dieren.
Er zijn verschillende soorten in vitro systemen. De twee meest gebruikte TME in vitro modellen bestaan uit gemengde monolaag of sferoïde celculturen. Beide kweekmethoden zijn voordelig voor fundamentele studies van intercellulaire interacties (bijvoorbeeld normale cellen met tumorcellen) en voor de analyse van verschillende TME specifieke veranderingen cel fenotype (bijv ontstaan van kanker-geassocieerde fibroblasten van normale fibroblasten). Bovendien kunnen de sferoïden kunnen een reflecterende weefsel structuur van het TME maken en kan representatief tumor heterogeniteit 23 zijn. Echter sferoïden leveren vaak zeer uiteenlopende zuurstofspanning gradiënten over lagen, die experimentele conclusies 24 kan bemoeilijken. Jammer genoeg, zijn beide modellen zeer beperkt in hun vermogen om pure cel populaties voor verdere karakterisering en studie volgende samenwerking te isolerencultuur. Anders zou zij ten minste één celtype worden fluorescent-gelabeld of gemerkt met een identificerend maker, en vervolgens onderwerpen van de gemengde co-kweek uitgebreide verwerking en celsortering de celpopulaties te scheiden. Terwijl een cel sorteerder kan isoleren van een vrij zuivere celpopulatie is, moet men zich bewust van cellulaire stress en mogelijke microbiële contaminatie risico 25.
Om het begrip van intercellulaire communicatie te vergemakkelijken, zijn grote inspanningen gewijd aan de ontwikkeling en het optimaliseren van in vitro systemen die nauw nabootsen van de in vivo milieu, terwijl het toelaten van een vereenvoudigde benadering. Een dergelijk instrument is de doorlaatbare microporeuze insert, een membraan substraat dat voor het eerst werd ontwikkeld in 1953 26 en vervolgens aangepast voor uiteenlopende toepassingen en studies (bijvoorbeeld cel polariteit 27, 28 endocytose, drug transport 29, weefsel modelleren 30, FERTilization 31, bystander effect 32,33, etc.). Dit systeem maakt de groei van cellen met in vivo-achtige anatomische en functionele differentiatie en expressie van vele in vivo tellers 34,35 die niet indien gekweekt op ondoordringbare plasticware waargenomen. Bovendien is de zeer dunne poreuze membraan (10 urn dik) maakt snelle diffusie van moleculen en verevening tijden, die de in vivo nabootst en maakt onafhankelijke cellulaire functioneren zowel de apicale en basolaterale domeinen cel. Een bijkomend voordeel van het inzetstuk nut als een TME systeem is de fysische scheiding van twee heterotypische celpopulaties gekweekt op beide zijden van het membraan dezelfde omgevingsomstandigheden, met behoud van verschillende wijzen van intercellulaire communicatie via het membraan poriën. Hoewel fysisch gescheiden, worden de twee celpopulaties metabolisch gekoppeld via uitgescheiden onderdelen en in dehier beschreven, ook door middel van gap-junctie kanalen. Bovendien, door het handhaven van de inzetstukken op de in vivo partiële zuurstofdruk (PO2), het model vermindert de complicaties van zuurstof en chemische gradiënten waargenomen in andere systemen. Eerder, verhoogt het begrip van natuurlijke mechanismen die de TME. Met name kunnen de twee celpopulaties eenvoudig worden geïsoleerd met hoge zuiverheid, zonder fluorescerende tagging en / of celsortering na langere co-cultuur.
Hier beschrijven we een in vitro TME protocol bestaat uit humaan borstcarcinoom cellen en humane fibroblasten gekweekt, respectievelijk aan weerszijden van een permeabele microporeuze membraan insert, maar toch continu bidirectionele communicatie via het membraan poriën. We tonen aan dat door membranen met verschillende poriegroottes, de bijdrage van een bepaald type intercellulaire communicatie (bijvoorbeeld uitgescheiden versus factoren gap junctions) de ontwikkelingvan het TME kan worden onderzocht.
