We ontwierpen een continu kweken inrichting voor gebruik met systemen optogenetic aan kweken van microben en regelmatig afbeeldingscellen branden in het effluent met een omgekeerde microscoop. Het kweken, sampling, beeldvorming en beeldanalyse zijn volledig geautomatiseerd, zodat dynamische reacties op de verlichting kan worden gemeten over meerdere dagen.
Optogenetische systemen maken gebruik van genetisch gecodeerde eiwitten die conformatie veranderen als reactie op specifieke golflengten van licht om cellulaire processen veranderen. Er is behoefte aan het kweken en meetsystemen die gebruik geprogrammeerd verlichting en stimulering van optogenetic systemen. We presenteren een protocol voor het bouwen en gebruiken van een continue kweekapparaat microbiële cellen met geprogrammeerde doses van licht verlichten en automatisch verkrijgen en afbeeldingen van cellen in het effluent geanalyseerd. De werking van deze inrichting een chemostat kan de groeisnelheid en de cellulaire omgeving goed te regelen. Het effluent van de continue celkweek wordt regelmatig bemonsterd en de cellen worden afgebeeld door multi-channel microscopie. Het kweken, sampling, beeldvorming en beeldanalyse zijn volledig geautomatiseerd, zodat dynamische reacties in de fluorescentie-intensiteit en cellulaire morfologie van de cellen in de steekproef uit de kweek effluent worden gemeten over meerdere dagenzonder gebruikersinvoer. We tonen de bruikbaarheid van deze kweken inrichting door het dynamisch induceren eiwitproductie in een stam van Saccharomyces cerevisiae vervaardigd met een optogenetic systeem dat transcriptie activeert.
Optogenetische systemen gebruiken licht om een groeiende lijst van cellulaire processen, waaronder genexpressie, 1, 2, 3, 4, 5 eiwit lokalisatie, 6 eiwitactiviteit, 6, 7, 8 eiwitbinding, 8, 9, 10 en eiwitafbraak controleren. 11 Werkwijze voor het kweken van cellen in een gecontroleerde omgeving met geprogrammeerde optische stimulatie en het meten van hun reactie op biologisch relevante tijdschalen moet het potentieel van deze instrumenten benutten voor onderzoek in de celbiologie en biotechnologie. Onze methode maakt gebruik van chemostatische een constante celgroei handhaven in een goed gemengde, aegewaardeerd en temperatuurregeling glazen kweekvat 12, 13 dat is blootgesteld aan verlichting geprogrammeerd. We afbeelding individuele cellen in de kweek effluent met een omgekeerde microscoop om de respons van de cultuur geprogrammeerde belichting te meten. Het kweken, sampling, beeldvorming en beeldanalyse zijn volledig geautomatiseerd, zodat de fluorescentie-intensiteit en cellulaire morfologie van het effluent celkweek kan worden gemeten over meerdere dagen zonder input van de gebruiker.
Dit protocol kan in de meeste laboratoria vertrouwd groeiende celkweek en microscopie uitgevoerd, en de gebruikte apparatuur is goedkoop en van gemakkelijk beschikbare componenten. Een transparante kweekvat wordt boven een matrix van licht-emitterende diodes (LEDs) kan uitzenden 1 uW / cm 2 -10 mW / cm2 licht geplaatst. Microben worden gekweekt in het kweekvat continu; een peristaltische pomp wordt gebruikt om media aan deverdunning, andere wordt gebruikt om de cultuur intrekken een lager aan de microscoop, en het verschil ontsnapt via een overloop. Een verwarmingselement handhaaft de temperatuur. Lucht wordt continu gepompt in het kweekvat om een positieve druk te handhaven en te mengen en beluchten cultuur. Met uitzondering van de luchtpomp, wordt het vermogen deze worden geregeld door een microcontroller die ook ontvangt input van een thermometer en een aangesloten computer. Het effluent celkweek wordt gepompt naar een microfluïdische apparaat op het podium van een omgekeerde microscoop. Non-tl en tl-beelden worden automatisch overgenomen. De cellen in de afbeeldingen worden gekenmerkt door een algoritme dat elke cel lokaliseert een interessegebied (ROI) en meet de eigenschappen van elke ROI.
Als u een toepassing van dit protocol aan te tonen, we meten de respons op wisselende lichtintensiteiten van Saccharomyces cerevisiae cellen ontworpen met een blauw licht verantwoorve optogenetic systeem dat de transcriptie van fluorescerend eiwit regelt. S. cerevisiae, algemeen bekend als bakkersgist, werd geselecteerd omdat meerdere optogenetic systemen voor het reguleren van genexpressie in dit systeem bestaan 14, 15, 16.
