Wir haben eine kontinuierliche Kultivierungsvorrichtung zur Verwendung mit optogenetische Systeme Kulturen von Mikroben zu beleuchten und regelmäßig Bildzellen in dem Abwasser mit einem umgekehrten Mikroskop. Die Kultivierung, Probenahme, Bildgebung und Bildanalyse sind voll automatisiert, so dass dynamische Reaktionen auf Beleuchtung kann über mehrere Tage gemessen werden.
Optogenetische Systeme verwenden genetisch kodierten Proteine, die Konformation in Reaktion auf bestimmte Wellenlängen des Lichts verändern zellulärer Prozesse zu verändern. Es besteht ein Bedarf für die Kultivierung und Messsysteme, die Beleuchtung und die Stimulation von optogenetische Systeme programmiert einzuarbeiten. Wir stellen ein Protokoll für den Aufbau und die Verwendung eines kontinuierlichen Kultivierungsvorrichtung mikrobiellen Zellen mit programmierter Dosen von Licht zu beleuchten, und automatisch erfassen und die Bilder der Zellen in dem Ausfluß zu analysieren. Der Betrieb dieser Vorrichtung als Chemostat ermöglicht die Wachstumsrate und der Zellumgebung streng kontrolliert werden. Der Austrag der kontinuierlichen Zellkulturen werden regelmäßig Proben genommen und die Zellen werden durch Mehrkanal-Mikroskopie abgebildet. Die Kultivierung, Probenahme, Bildverarbeitung und Bildanalyse sind vollständig automatisiert, so dass dynamische Reaktionen in der Fluoreszenzintensität und zellulärer Morphologie von aus der Kultur Abstrom abgetastete Zellen über mehrere Tage gemessenohne Benutzereingabe. Wir zeigen die Nützlichkeit dieser Kultivierungsvorrichtung durch dynamisches Protein – Produktion in einem Stamm von Saccharomyces cerevisiae konstruiert mit einem optogenetische System induziert die Transkription aktiviert.

Der Abstrom Zellkultur wird auf eine Mikrofluidik-Vorrichtung auf der Bühne eines umgekehrten Mikroskops gepumpt. Nicht fluoreszierende und Fluoreszenzbilder werden automatisch erfasst. Die Zellen in den Bildern durch einen Algorithmus gekennzeichnet, daß jede Zelle als eine Region von Interesse (ROI) lokalisiert und mißt die Eigenschaften der einzelnen ROI.
Um eine Anwendung dieses Protokolls zu demonstrieren, maßen wir die Antwort auf unterschiedlichen Lichtintensitäten von Saccharomyces cerevisiae – Zellen entwickelt , mit einem Blaulicht Verantve optogenetische System, das die Transkription von fluoreszierenden Proteins steuert. S. cerevisiae, wie Bäckerhefe allgemein bekannt ist , wurde ausgewählt , weil mehrere optogenetische Systeme für die bereits in diesem System Genexpression Steuerung 14 vorhanden ist , 15, 16. Darüber hinaus ist dieses Modellorganismus für Studien in der Systembiologie 17 und als Fahrgestell für biotechnologische Anwendungen 18, 19, 20 verwendet.
