Vi har utformat en kontinuerlig odlingsapparat för användning med optogenetic system för att belysa kulturer av mikrober och regelbundet bildceller i utflödet med ett inverterat mikroskop. Odlingen, provtagning, bildbehandling och bildanalys är helt automatiserad så att dynamiska svar på belysning kan mätas under flera dagar.
Optogenetic system utnyttjar genetiskt kodade proteiner som ändrar konforma som svar på specifika våglängder av ljus för att förändra cellulära processer. Det finns ett behov för odling och mätsystem som innehåller programmerade belysning och stimulering av optogenetic system. Vi presenterar ett protokoll för att bygga och använda en kontinuerlig odlingsapparat för att belysa mikrobiella celler med programmerade doser av ljus, och automatiskt förvärva och analysera bilder av celler i utflödet. Driften av denna apparat som kemostat tillåter tillväxttakten och den cellulära miljön att hårt kontrollerad. Utflödet av det kontinuerliga cellkulturen regelbundet samplas och cellerna avbildas av flerkanals mikroskopi. Odlingen, provtagning, bildbehandling och bildanalys är helt automatiserad så att dynamiska svar i fluorescensintensitet och cellulär morfologi av celler samplade från odlings utflöde mäts över flera dagarutan användarinmatning. Vi visar användbarheten av detta odlingsapparat genom att dynamiskt inducera proteinproduktion i en stam av Saccharomyces cerevisiae konstruerad med ett optogenetic system som aktiverar transkription.
Optogenetic system använder ljus för att styra en växande lista av cellulära processer, inklusive genexpression, 1, 2, 3, 4, 5 proteinlokalisering, 6-proteinaktivitet, 6, 7, 8-protein-bindning, 8, 9, 10 och proteinnedbrytning. 11 En metod för odling av celler i en kontrollerad miljö med programmerad optisk stimulering och för att mäta deras svar över biologiskt relevanta tidsskalor är nödvändig för att utnyttja potentialen hos dessa verktyg för forskning inom cellbiologi och bioteknik. Vår metod drar fördel av chemostasis att bibehålla en konstant celltillväxthastighet i en välblandad, aeklassad, och temperaturreglerad glasodlingskärl 12, 13 som är utsatt för programmerad belysning. Detta protokoll kan implementeras i de flesta labb bekanta med växande cellkultur och mikroskopi, och apparaten som används är billig och gjord av lätt tillgängliga komponenter. En transparent odlingskärl är placerat ovanför en matris av Ijusemitterande dioder (LED) som kan avge ett iW / cm 2 -10 mW / cm 2 av ljus. Mikrober odlas i odlingskärlet kontinuerligt; en peristaltisk pump används för att lägga till media iutspädningshastighet, en annan används för att dra tillbaka kulturen till en lägre hastighet till mikroskopet, och skillnaden försvinner genom ett bräddavlopp. En värmedyna upprätthåller temperaturen. Utflödet cellodling pumpas till en mikrofluidanordning på scenen av ett inverterat mikroskop. Icke-fluorescerande och fluorescerande bilder förvärvas automatiskt. Cellerna i bilderna kännetecknas av en algoritm som lokaliserar varje cell som ett område av intresse (ROI) och mäter egenskaperna för varje ROI.
För att demonstrera en tillämpning av detta protokoll, mätte vi svaret på varierande ljusintensitet av Saccharomyces cerevisiae-celler modifierade med en blåljus ansvave optogenetic system som kontrollerar transkriptionen av fluorescerande protein. S. cerevisiae, allmänt känd som bagerijäst, valdes på grund multipla optogenetic system för reglering genuttryck i detta system redan finns 14, 15, 16.
