Wij rapporteren hier, de lokalisatie van de immunofluorescentietest voor dynamin ter illustratie van de protocollen voor de detectie van eiwitten in paraffine-ingebedde muis epididymaire secties en die van een vereeuwigd epididymaire cellijn (mECap18). Ook beschrijven we de protocollen voor de isolatie van proteïnen van de secretoire van zowel epididymaire vloeistof en geconditioneerde cel media.
De zoogdieren bijbal genereert een van de meest complexe intraluminale vloeistoffen van een endocriene klier ter ondersteuning van de post-testiculaire rijping en opslag voor spermacellen. Dergelijke complexiteit ontstaat als gevolg van de gecombineerde secretoire en absorberend activiteit van de epitheliale cellen van de voering. Hier beschrijven we de technieken voor de analyse van epididymaire eiwitsynthese en secretie door zich te concentreren op het eiwit modelgezin van dynamin (DNM) mechanoenzymes; grote GTPases die de potentie hebben om reguleren bi-directionele membraan mensenhandel gebeurtenissen. Voor de studie van eiwit expressie in epididymaire weefsel beschrijven wij robuuste methodologie voor het labelen van de immunofluorescentie doel eiwitten in paraffine-ingebedde secties en de daaropvolgende opsporing van de ruimtelijke spreiding van deze eiwitten via immunofluorescentie microscopie. Ook beschrijven we geoptimaliseerde methodologie voor de isolatie en de karakterisering van exosome zoals blaasjes, bekend als epididymosomes, die worden uitgescheiden in de epididymaire lumen deelnemen aan intercellulaire communicatie met de rijping van de zaadcellen. Als een complementaire aanpak beschrijven we ook de immunofluorescentie detectie van doel eiwitten in een cellijn SV40-vereeuwigd muis caput epididymaire epitheliale (mECap18). Bovendien bespreken we het nut van de mECap18 cellijn als een model geschikt in vitro met voor het verkennen van de regulering van epididymaire secretoire activiteit. Voor dit doel beschrijven we de kweken eisen voor het onderhoud van de cellijn van mECap18 en het gebruik van selectieve farmacologische remming regimes die kunnen beïnvloeden hun secretoire eiwit-profiel. De laatste zijn beoordeeld gemakkelijk via het oogsten van geconditioneerde kweekmedium, concentratie van proteïnen toe secreted via trichloorazijnzuur/aceton neerslag en hun latere analyse via de SDS-pagina en immunoblotting. Wij betogen dat deze gecombineerde methoden geschikt voor de analyse van alternatieve epididymaire eiwit doelstellingen als een prelude zijn op de bepaling van hun functionele rol in de rijping van de zaadcellen en/of opslag.
De spermacellen van alle soorten zoogdieren verwerven de mogelijkheden om weer vooruit progressieve mobiliteit te geven en om een eicel te bevruchten tijdens hun langdurige afdaling via de bijbal, een zeer gespecialiseerde gebied van de mannelijke extra testiculaire kanaalsysteem, die kan 7-14 dagen om te navigeren (afhankelijk van de soort)1. Als gevolg van de extreme condensatie van de chromatine vaderlijke en het vergieten van de meerderheid van het cytoplasma die hoort bij de cytodifferentiation van spermacellen in de teelballen, wordt hun latere functionele rijping gedreven uitsluitend door hun interactie met de epididymaire communicatie. Dit milieu is, op zijn beurt, gemaakt door de secretoire en absorberend activiteit van de voering epididymaire soma en geeft een uitzonderlijke niveau van segment-segment variatie1. Dus, de meest actieve segmenten op het gebied van de eiwitsynthese en secretie zijn gelegen in het proximale deel van de bijbal (namelijk, de caput en corpus)2. Deze activiteit komt overeen met de functionele Profiel van spermatozoïden, met het eerste begin van de cellen weer te geven van de kenmerken van functionele bevoegdheid (dwz., progressieve mobiliteit en het vermogen om te binden aan zuur-ontbindend zona glycoproteïnen) volgende hun passage door het caput bijbal3. Deze functionele kenmerken blijven ontwikkelen vóór het bereiken van optimale niveaus als de zaadcellen de distale epididymaire segment (cauda) bereiken, waarin ze zijn opgeslagen in een rustige staat in gereedheid voor de ejaculatie. De vorming en het onderhoud van dit sperma opslag reservoir is ook nauw verbonden met de voering epitheel, die in de cauda wordt gedomineerd door sterke absorberend activiteit4,5. Hoewel anatomische verschillen gerapporteerde6,7,8 geweest, lijkt dergelijke regionale verdeling van de arbeid te zijn een kenmerk van de bijbal die wordt gedeeld door de meerderheid van de soorten zoogdieren studeerde tot nu toe, met inbegrip van onze eigen9,10. Inderdaad, vanuit klinisch oogpunt, het is bekend dat epididymaire dysfunctie levert een belangrijke bijdrage aan de etiologie van mannelijke factor onvruchtbaarheid11, dus wijzen op het belang van inzicht in de regulering van deze gespecialiseerde weefsel.
Het is daarom betreurenswaardig dat ons begrip van epididymaire fysiologie, en de mechanismen die de opeenvolgende fasen van rijping van de zaadcellen en opslag binnen dit weefsel, reguleren blijven worden volledig opgelost. De bijdragende factoren behoren beperkende voorschotten in epididymaire onderzoek tot de algehele complexiteit van dit weefsel en kennis van de mechanismen die het uitoefenen van regelgevende controle over haar luminal communicatie. Anatomisch, weten we dat buiten het onderscheid van caput, corpus en cauda segmenten, de bijbal kan verder worden onderverdeeld in verschillende zones (figuur 1A), elk gescheiden door septa12 en gekenmerkt door aparte profielen van gene/eiwit expressie13,14,15,16,17,18. Inderdaad, op basis van gedetailleerde transcriptionele profilering van gesegmenteerde genexpressie in de bijbal, maar liefst 6 en 9 verschillende epididymaire zones zijn gemeld in de muis en rat modellen, respectievelijk19,20. Dergelijke complexiteit vermoedelijk weerspiegelt de samenstelling van de epididymaire soma, een pseudostratified epitheel bestaat uit talrijke verschillende soorten cellen; elke verschillen met betrekking tot hun overvloed, distributie en secretoire/absorberend activiteiten langs de lengte van het darmkanaal. Belangrijkste cellen zijn dus veruit de meest voorkomende epididymaire cel type vormen meer dan 80% van alle epitheliale cellen. Dienovereenkomstig, belangrijkste cellen zijn verantwoordelijk voor het grootste deel van epididymaire eiwit biosynthese en secretie5. Daarentegen zijn de cel wissen bevolking, die gerangschikt als de tweede meest voorkomende celtype binnen de epididymaire soma, voornamelijk betrokken bij selectieve absorptie van luminal componenten en de verzuring van deze communicatie-5. Een andere laag van complexiteit toe te voegen, uitoefenen androgenen en andere lumicrine factoren van testiculaire oorsprong differentiële controle over elk van deze epididymaire celtypes afhankelijk van hun plaats langs de tractus.
Ondanks de beperkingen opgelegd door dergelijke complexiteit, blijven belangrijke inbreuk worden gemaakt in het oplossen van de mechanistische basis van epididymaire functie. Een sleutel tot deze studies is de toepassing van geavanceerde massaspectrometrie strategieën om grootschalige voorraden van de epididymaire Proteoom, samen met gedetailleerde analyses van individuele eiwitten gekozen uit deze eerste onderzoeken geweest. Een illustratie van deze benadering is onze recente karakterisering van de DNM-familie van mechanoenzymes in de muis model21. Onze eerste interesse in DNM werd aangewakkerd door haar dubbele werking in de koppeling van exo- en endocytotic processen. Voortbouwend op deze opmerkingen, konden we aantonen dat de drie canonieke isoforms van DNM (DNM1 – DNM3) zijn sterk in de bijbal muis uitgedrukt en op de juiste manier gepositioneerd om te voldoen aan regelgeving rollen in eiwit secretie en absorptie21 . Bovendien konden we duidelijk onderscheid elke DNM-isovorm op basis van hun cellulaire en sub cellulaire lokalisatie, dus suggereren dat zij elkaar aanvullen, in tegenstelling tot de redundante, activiteit binnen de epididymaire epitheel-21bezitten.
Hier beschrijven we de experimentele methodologie werkzaam voor de studie van DNM expressie in de bijbal muis met de hoop dat deze informatie zal ruimere toepassing in de karakterisering van alternatieve epididymaire eiwitten vinden en aldus bijdragen aan onze inzicht in de functie van dit belangrijke element van het mannelijk voortplantingssysteem. Specifiek, beschrijven we de ontwikkeling van robuuste methodologie voor het labelen van de immunofluorescentie doel eiwitten in paraffine-ingebedde epididymaire secties en de daaropvolgende opsporing van de ruimtelijke spreiding van deze eiwitten via immunofluorescentie microscopie. Documenteren we verder onze onlangs geoptimaliseerde protocollen22 voor de isolatie en de karakterisering van epididymosomes; kleine exosome-achtige blaasjes die vormen belangrijke elementen van het epididymaire secretoire profiel en lijken te houden een prominente rol bij de bevordering van sperma rijping23. Als een complementaire aanpak beschrijven we ook de immunofluorescentie detectie van doel eiwitten in een cellijn vereeuwigd muis caput epididymaire epitheliale (mECap18) en het gebruik van deze hulpbron als een model om te verkennen van de regulering van de epididymaire secretoire activiteit in vitro.
Deze studies opgenomen het gebruik van Bouin van vaste epididymaire weefsel dat paraffine insluiten en vectorafbeeldingsbestanden standaardprotocollen had ondergaan. Bouin van kleefpoeders oplossing bestaat uit een mengsel van formaldehyde, picrinezuur en azijnzuur, met elke component die een specifieke en complementaire functie hebben. Dus, formaldehyde reageert met primaire amines vormen van eiwit cross-links pikrinezuur langzaam dringt het weefsel zouten vormen en vandaar stolling van fundamentele eiwitten en omgekeer…
