Hier präsentieren wir Ihnen ein Protokoll für die genetische Aufnahme von L-Dihydroxyphenylalanine Biosynthesized aus einfachen Ausgangsmaterialien und seine Anwendung auf Protein Konjugation.
L-Dihydroxyphenylalanine (DOPA) ist eine Aminosäure, die in die Biosynthese der Katecholamine bei Tieren und Pflanzen gefunden. Wegen seiner besonderen biochemischen Eigenschaften hat die Aminosäure Mehrfachnutzung in biochemische Anwendungen. Dieser Bericht beschreibt ein Protokoll für die genetische Einbeziehung der Biosynthesized DOPA und seine Anwendung auf Protein Konjugation. DOPA ist Biosynthesized durch ein Tyrosin Phenol-Lyase (TPL) von brenzcatechins, Pyruvat und Ammoniak und die Aminosäure ist direkt durch die genetische Einbau-Methode mit einem weiterentwickelten Aminoacyl-tRNA und Aminoacyl-tRNA synthestase Paar in Proteine aufgenommen. Dieses direkte Einbindung System beinhaltet effizient DOPA mit wenig Einbindung anderer natürlicher Aminosäuren und bessere Proteinausbeute als das Vorgängersystem genetische Aufnahme für DOPA. Protein-Konjugation mit DOPA-haltige Proteine ist effizient und ortsspezifische und zeigt seine Nützlichkeit für die verschiedensten Anwendungen. Dieses Protokoll bietet Protein Wissenschaftler mit detaillierten Verfahren für die effiziente Biosynthese von mutierten Proteine mit DOPA an gewünschten Standorten und deren Konjugation für industrielle und pharmazeutische Anwendungen.
DOPA ist eine Aminosäure, die in die Biosynthese der Katecholamine bei Tieren und Pflanzen beteiligt. Diese Aminosäure wird von Tyr von Tyrosin-Hydroxylase und molekularer Sauerstoff (O2)1synthetisiert. Da DOPA eine Vorstufe von Dopamin ist und die Blut – Hirn-Schranke durchdringen kann, hat es bei der Behandlung von Parkinson-Krankheit2eingesetzt. DOPA findet sich auch in Muschel Adhäsion Proteine (MAPs), die für die Hafteigenschaften der Muscheln in nassen Bedingungen3,4,5,6,7zuständig sind. Tyr wird zunächst an den Positionen kodiert, wo DOPA findet sich in Karten und dann durch tyrosinasen8,9in DOPA umgewandelt wird. Wegen seiner interessanten biochemischen Eigenschaften wurde DOPA in einer Vielzahl von Anwendungen eingesetzt. Die Dihydroxyl Gruppe von DOPA ist chemisch anfällig für Oxidation und die Aminosäure ist in L-Dopaquinone, ein Vorläufer der Melanine umfunktioniert. Aufgrund seiner hohen Electrophilicity wurden für Vernetzung und Konjugation mit Thiole und Amine10,11,12,13L-Dopaquinone und seine Derivate eingesetzt. 1,2-Chinonen können auch als ein Diens für Cycloaddition Reaktionen und werden durch Belastung gefördert Oxidation-gesteuerten Cyclooctyne-1,2-Quinone (SPOCQ) deutscher14seit biokonjugaten. Darüber hinaus die Dihydroxyl Gruppe kann Metallionen wie Fe3 + und Cu2 +Chelat und Proteine mit DOPA haben für Drug Delivery und Metallion sensing15,16verwendet worden.
DOPA hat genetisch in Proteine mit einem orthogonalen Aminoacyl-tRNA (aa-tRNA) und die Aminoacyl-tRNA synthestase (AaRS) Paar17 integriert und verwendet für Protein Konjugation und Vernetzung10,11, 12,13. In diesem Bericht werden experimentelle Ergebnisse und Protokolle für die genetische Einbeziehung von DOPA Biosynthesized von billige Ausgangsmaterialien und ihre Anwendungen auf biokonjugaten beschrieben. DOPA ist Biosynthesized mit einem TPL und ausgehend von brenzcatechins, Pyruvat und Ammoniak in Escherichia coli. Biosynthesized DOPA ist direkt integriert Proteine ein weiterentwickelter aa-tRNA und AaRS Pair für DOPA zum Ausdruck zu bringen. Darüber hinaus ist das Biosynthesized Protein mit DOPA mit einem fluoreszierenden Sonde und vernetzt zu produzieren Protein Oligomere ortspezifisch konjugiert. Dieses Protokoll wird nützlich sein für Protein Wissenschaftler zur Biosynthese mutierte Proteine mit DOPA und konjugieren der Proteine mit biochemischen Sonden oder Drogen für industrielle und pharmazeutische Anwendungen.
In diesem Protokoll werden die Biosynthese und die direkte Einbindung von DOPA beschrieben. Die Bakterienzelle verwendet bei dieser Methode kann eine weitere Aminosäure synthetisieren und verwenden es als unnatürlich Baustein für die Proteinsynthese. Die genetische Einbeziehung der unnatürlichen Aminosäuren ist eine Schlüsseltechnologie für die Entwicklung der unnatürlichen Organismus mit einer erweiterten genetischen Code gewesen.
