De thoracale spinale hemisectie van de rat is een waardevol en reproduceerbaar model van eenzijdige ruggenmergletsel om de neurale mechanismen van het motorische herstel en de werkzaamheid van de behandeling te onderzoeken. Dit artikel bevat een gedetailleerde stapsgewijze handleiding om de hemisectie-procedure uit te voeren en om de prestaties van het bewegings vermogen in een open-veld Arena te beoordelen.
Ruggenmergletsel (SCI) veroorzaakt verstoringen in de motorische, sensorische en autonome functie onder het niveau van de laesie. Experimentele diermodellen zijn waardevolle hulpmiddelen om de neurale mechanismen die betrokken zijn bij het motorische herstel na SCI te begrijpen en om therapieën voor klinische populaties te ontwerpen. Er zijn verschillende experimentele Sci-modellen, waaronder contusie, compressie en transect blessures die worden gebruikt in een breed scala aan soorten. Een hemisectie omvat de eenzijdige transectie van het ruggenmerg en verstoort alle stijgende en dalende Tracts aan één kant. Spinale hemisectie produceert een zeer selectieve en reproduceerbare verwonding in vergelijking met contusie of compressietechnieken die nuttig is voor het onderzoeken van Neurale plasticiteit in gespaard en beschadigde trajecten in verband met functioneel herstel. We presenteren een gedetailleerd stap-voor-stap protocol voor het uitvoeren van een thoracale hemisectie op het T8-wervel niveau bij de rat, wat resulteert in een initiële verlamming van de achterledemaat aan de zijkant van de laesie met een cijfer spontaan herstel van de motorische functie over verschillende Weken. We bieden ook een locomotief scoring protocol om te beoordelen van functionele herstel in het open-veld. De motorische beoordeling biedt een lineair herstel profiel en kan zowel vroegtijdig als herhaaldelijk na letsel worden uitgevoerd om dieren nauwkeurig op te schermen voor geschikte tijdpunten om meer gespecialiseerde gedragstesten uit te voeren. De voorgestelde hemisectie-techniek kan gemakkelijk worden aangepast aan andere transect-modellen en-soorten, en de motorische beoordeling kan worden gebruikt in een verscheidenheid aan SCI-en andere verwondings modellen om de bewegings functie te scoren.
Ruggenmergletsel (SCI) wordt geassocieerd met ernstige verstoringen in de motorische, sensorische en autonome functie. Experimentele diermodellen van SCI zijn waardevolle hulpmiddelen om de anatomische en fysiologische gebeurtenissen die betrokken zijn bij SCI-pathologie te begrijpen, om de neurale mechanismen in herstel en herstel te onderzoeken, en om te scherm voor de werkzaamheid en veiligheid van potentiële therapeutische Interventies. De rat is de meest gebruikte soort in SCI Research1. Rat modellen zijn lage kosten, gemakkelijk te reproduceren, en een grote batterij van gedrags tests zijn beschikbaar voor het beoordelen van de functionele uitkomsten2. Ondanks enkele verschillen in Tract locaties, de rat ruggenmerg aandelen algemene soortgelijke sensorimotorische functies met grotere zoogdieren, met inbegrip van primaten3,4. Ratten delen ook analoge fysiologische en gedragsmatige gevolgen voor SCI die betrekking hebben op mensen5. Niet-menselijke primaten en grote diermodellen kunnen een nauwere benadering van humane SCI6 bieden en zijn essentieel om de veiligheid en werkzaamheid van de behandeling te bewijzen voorafgaand aan menselijke experimenten, maar worden minder vaak gebruikt als gevolg van ethisch en dierenwelzijn overwegingen, uitgaven en regelgevingsvereisten7.
Rat transect Sci modellen worden uitgevoerd door de gerichte onderbreking van het ruggenmerg met een selectieve laesie met behulp van een dissectie mes of iridectomie schaar na een laminectomie. Vergeleken met een volledige transect, resulteert partiële transect in de rat in een minder ernstige verwonding, gemakkelijker postoperatieve dierenverzorging, spontaan bewegings herstel, en meer in de gaten in de Sci-modellen bij mensen die overwegend onvolledig zijn met gedeeltelijke kaliumsparende van weefsel dat het ruggenmerg en de supraspinale structuren verbindt8. Een eenzijdige hemisectie verstoort alle stijgende en dalende Tracts aan één kant, en produceert kwantificeerbare en zeer reproduceerbare motorische tekorten, waardoor de verkenning van de onderliggende biologische mechanismen wordt versterkt. De meest prominente functionele consequentie van de hemisectie is een verlamming van de initiële ledemaat aan dezelfde kant en onder het niveau van de laesie met Graded spontaan herstel van de bewegings functie gedurende meerdere weken9,10, 11 , 12. het hemisectie-model is met name nuttig voor het onderzoeken van Neurale plasticiteit van beschadigde en residuele Tracts en circuits geassocieerd met functioneel herstel9,11,12, 13,14,15,16,17,18. In het bijzonder is hemisectie uitgevoerd op het thoracale niveau, d.w.z. boven de spinale circuits die de achterste ledematen beheersen, bijzonder nuttig voor het onderzoeken van veranderingen in de bewegingscontrole. Omdat er een niet-lineaire relatie bestaat tussen laesie-ernst en bewegings herstel na SCI19, is het van cruciaal belang om de functionele uitkomsten te beoordelen in experimentele modellen.
Er is een uitgebreide reeks gedragstesten beschikbaar om specifieke aspecten van functioneel bewegings herstel in de rat2,20te beoordelen. Veel bewegings tests bieden geen betrouwbare maatregelen vroeg na SCI omdat ratten te gehandicapt zijn om hun lichaamsgewicht te ondersteunen. Een maat voor de spontane prestaties van het bewegingsapparaat die vroeg na de verwonding gevoelig zijn voor tekorten en waarvoor geen preoperatieve training of gespecialiseerde apparatuur nodig is, is nuttig om het bewegings herstel te bewaken voor geschikte tijdpunten om supplement gespecialiseerde gedragstesten. De Martinez open-Field Assessment score10, oorspronkelijk ontwikkeld voor het evalueren van de motorische prestaties na cervicale SCI in de rat, is een 20-punts ordinale Score die de mondiale motorische prestaties beoordeelt tijdens spontane bovengrondse-motoriek in een Open-Field. Scores worden afzonderlijk voor elke ledemaat uitgevoerd met behulp van een rubriek die specifieke parameters evalueert van een reeks bewegings maatregelen, waaronder de bewegingen van de gewrichts ledematen, gewichtsondersteuning, cijfer positie, stappen mogelijkheden, coördinatie van de forelimb-hindledematen en staart Positie. De beoordelingsscore is afgeleid van de open-veldbeoordelings schaal Basso, Beattie en Bresnahan (BBB) die is ontworpen om de prestaties van het bewegings vermogen na thoracische contusie21te evalueren. Het is aangepast aan het nauwkeurig en betrouwbaar evalueren van zowel de forelimb en de hindledemaat motorische functie, maakt onafhankelijke beoordeling van de verschillende scoring parameters die niet vatbaar is voor de hiërarchische scoring van de BBB, en biedt een lineair herstel profiel10. Bovendien, in vergelijking met de BBB, de assessment Score is gevoelig en betrouwbaar in ernstiger letsel modellen10,11,20,22. De beoordelingsscore is gebruikt voor het beoordelen van motorische stoornissen bij ratten na cervicale10,12 en thoracale9 Sci alleen en in combinatie met traumatisch hersenletsel23.
We presenteren hier een gedetailleerd stap-voor-stap protocol voor het uitvoeren van een thoracale hemisectie SCI op de T8 Vertebrale niveau in de vrouwelijke Long-Evans rat, en voor het beoordelen van hindledemaat motorische herstel in het open-veld.
Een belangrijke sterkte van de hemisectie-techniek is de selectiviteit en reproduceerbaarheid van de laesie, wat leidt tot een verminderde variabiliteit in histologische en gedragsfenotypes tussen dieren25.
