Denne protokollen skisserer en rutinemessig metode for bruk av seriell blokk-ansikt skanning elektronmikroskopi (SBF-SEM), en kraftig 3D-bildeteknikk. Vellykket påføring av SBF-SEM hengsler på riktig fiksering og vevsfargingsteknikker, samt nøye vurdering av bildebehandlingsinnstillinger. Denne protokollen inneholder praktiske hensyn for hele denne prosessen.
Seriell blokk-ansikt skanning elektronmikroskopi (SBF-SEM) gjør det mulig å samle hundrevis til tusenvis av serieregistrerte ultrastrukturelle bilder, og tilbyr en enestående tredimensjonal visning av vevsmikroanatomi. Mens SBF-SEM har sett en eksponentiell økning i bruk de siste årene, er tekniske aspekter som riktig vevsforberedelse og avbildningsparametere avgjørende for suksessen til denne bildemodaliteten. Dette bildesystemet drar nytte av enhetens automatiserte natur, slik at man kan forlate mikroskopet uten tilsyn under bildebehandlingsprosessen, med den automatiserte samlingen av hundrevis av bilder mulig på en enkelt dag. Men uten passende vev forberedelse cellulær ultrastruktur kan endres på en slik måte at feil eller villedende konklusjoner kan trekkes. I tillegg genereres bilder ved å skanne blokksiden av en harpiks-innebygd biologisk prøve, og dette gir ofte utfordringer og hensyn som må tas. Opphopning av elektroner i blokken under avbildning, kjent som “vevslading”, kan føre til tap av kontrast og manglende evne til å sette pris på cellulær struktur. Videre, mens økende elektronstråleintensitet / spenning eller synkende stråleskanningshastighet kan øke bildeoppløsningen, kan dette også ha den uheldige bivirkningen av å skade harpiksblokken og forvrenge etterfølgende bilder i bildeserien. Her presenterer vi en rutinemessig protokoll for fremstilling av biologiske vevsprøver som bevarer cellulær ultrastruktur og reduserer vevslading. Vi tar også hensyn til bildebehandling for rask anskaffelse av seriebilder av høy kvalitet med minimal skade på vevsblokken.
Seriell blokk ansiktsskanning elektronmikroskopi (SBF-SEM) ble først beskrevet av Leighton i 1981 hvor han formet et skanningselektronmikroskop forsterket med en innebygd mikrotom som kunne kutte og bilde tynne deler av vev innebygd i harpiks. Dessverre begrenset tekniske begrensninger bruken til ledende prøver, da ikke-ledende prøver som biologisk vev akkumulerte uakseptable ladenivåer (elektronoppbygging i vevsprøven)1. Mens belegg blokk-ansikt mellom kutt med fordampet karbon redusert vev lading, dette sterkt økt bildebehandling oppkjøpstid og bildelagring forble et problem som datateknologi på den tiden var utilstrekkelig til å administrere de store filstørrelsene som er opprettet av enheten. Denne metodikken ble revidert av Denk og Horstmann i 2004 ved hjelp av en SBF-SEM utstyrt med et variabelt trykkkammer2. Dette tillot innføring av vanndamp til bildekammeret som reduserer lading i prøven, noe som gjør avbildning av ikke-ledende prøver levedyktig, om enn med tap av bildeoppløsning. Ytterligere forbedringer i vevsforberedelse og avbildningsmetoder tillater nå avbildning ved hjelp av høyt vakuum, og SBF-SEM-avbildning er ikke lenger avhengig av vanndamp for å spre lading3,4,5,6,7,8,9. Mens SBF-SEM har sett en eksponentiell økning i bruk de siste årene, er tekniske aspekter som riktig vevsforberedelse og avbildningsparametere avgjørende for suksessen til denne bildemodaliteten.
SBF-SEM tillater automatisert innsamling av tusenvis av serieregistrerte elektronmikroskopibilder, med planaroppløsning så liten som 3-5 nm10,11. Vev, impregnert med tungmetaller og innebygd i harpiks, er plassert i et skanningselektronmikroskop (SEM) som inneholder en ultramikrotomi utstyrt med en diamantkniv. En flat overflate er kuttet med diamantkniven, kniven trekkes tilbake, og overflaten av blokken skannes i et rastermønster med en elektronstråle for å skape et bilde av vev ultrastruktur. Blokken heves deretter en bestemt mengde (f.eks. 100 nm) i z-aksen, kjent som et “z-trinn”, og en ny overflate kuttes før prosessen gjentas. På denne måten produseres en 3-dimensjonal (3D) blokk med bilder når vevet kuttes bort. Dette bildesystemet drar ytterligere nytte av enhetens automatiserte natur, slik at man kan forlate mikroskopet uten tilsyn under bildebehandlingsprosessen, med den automatiserte samlingen av hundrevis av bilder mulig på en enkelt dag.
Mens SBF-SEM-avbildning primært bruker backscattered elektroner for å danne et bilde av blokkflaten, genereres sekundære elektroner under bildebehandlingsprosessen12. Sekundære elektroner kan akkumuleres, sammen med backscattered og primærstråleelektroner som ikke unnslipper blokken, og produserer “vevslading”, noe som kan føre til et lokalisert elektrostatisk felt ved blokkflaten. Denne elektronakkumuleringen kan forvrenge bildet eller føre til at elektroner kastes ut av blokken og bidrar til signalet som samles inn av backscatter-detektoren, og reduserer signal-til-støy-forholdet13. Mens nivået av vevslading kan reduseres ved å redusere elektronstrålespenningen eller intensiteten, eller redusere strålebotiden, resulterer dette i et redusert signal-til-støy-forhold14. Når en elektronstråle med lavere spenning eller intensitet brukes, eller strålen bare har lov til å bo innenfor hvert pikselrom i en kortere periode, blir mindre backscattered elektroner kastet ut av vevet og fanget av elektrondetektoren, noe som resulterer i et svakere signal. Denk og Horstmann håndterte dette problemet ved å introdusere vanndamp i kammeret, og dermed redusere ladningen i kammeret og på blokkflaten på bekostning av bildeoppløsning. Med et kammertrykk på 10-100 Pa er en del av elektronstrålen spredt og bidrar til bildestøy og tap av oppløsning, men dette produserer også ioner i prøvekammeret som nøytraliserer ladningen i prøveblokken2. Nyere metoder for nøytralisering av ladningen i prøveblokken bruker brenngassinjeksjon av nitrogen over blokkflaten under avbildning, eller introduserer negativ spenning til SBF-SEM-trinnet for å redusere sondestråle-lading energi og øke signalet samlet inn6,7,15. I stedet for å introdusere stadiumskjevhet, kammertrykk eller lokalisert nitrogeninjeksjon for å redusere ladningsoppbyggingen på blokkoverflaten, er det også mulig å øke reduktiviteten til harpiksen ved å introdusere karbon til harpiksblandingen, noe som gir mer aggressive bildebehandlingsinnstillinger16. Følgende generelle protokoll er en tilpasning av Deerinck et al. protokollen publisert i 2010 og dekker modifikasjoner på vevsforberedelse og avbildningsmetoder vi fant nyttige for å minimere vevslading samtidig som vi opprettholder høyoppløselig bildeoppkjøp3,17,18,19. Mens den tidligere nevnte protokollen fokuserte på vevsbehandling og tungmetallimpregnering, gir denne protokollen innsikt i bildebehandlings-, dataanalyse- og rekonstruksjonsarbeidsflyten som er knyttet til SBF-SEM-studier. I vårt laboratorium har denne protokollen blitt vellykket og reprodusert påført et bredt utvalg av vev, inkludert hornhinne og fremre segmentstrukturer, øyelokk, lacrimal og hardere kjertel, netthinne og optisk nerve, hjerte, lunge og luftvei, nyre, lever, cremaster muskel, og cerebral cortex / medulla, og i en rekke arter, inkludert mus, rotte, kanin, marsvin, fisk, monolayer og stratifiserte cellekulturer, gris, ikke-menneskelig primat, samt menneskelig20,21,22,23. Selv om små endringer kan være verdt for spesifikke vev og applikasjoner, har denne generelle protokollen vist seg å være svært reproduserbar og nyttig i sammenheng med vårt kjerneavbildningsanlegg.
Hensikten med dette metodepapiret er å fremheve vevsforberedelses- og bildemetodikken som har gjort det mulig for laboratoriet vårt å fange opp høyoppløselige serielle elektronmikroskopibilder på en pålitelig måte, og å påpeke kritiske trinn som fører til dette resultatet, samt potensielle fallgruver som kan oppstå når du utfører SBF-SEM-avbildning.
