Dit protocol maakt gebruik van fluorescerende reporters en celsortering om knock-in experimenten in macrofaag- en T-cellijnen te vereenvoudigen. Twee plasmiden worden gebruikt voor deze vereenvoudigde knock-in experimenten, namelijk een CRISPR/Cas9- en DsRed2-tot expressie brengend plasmide en een homoloog recombinatiedonorplasmide dat EBFP2 tot expressie brengt, dat permanent is geïntegreerd op de Rosa26-locus in immuuncellen.
Functionele genomics studies van het immuunsysteem vereisen genetische manipulaties die zowel deletie van doelgenen als toevoeging van elementen aan eiwitten van belang omvatten. Identificatie van genfuncties in cellijnmodellen is belangrijk voor genontdekking en verkenning van celintrinsieke mechanismen. Genetische manipulaties van immuuncellen zoals T-cellen en macrofaagcellijnen met behulp van CRISPR / Cas9-gemedieerde knock-in zijn echter moeilijk vanwege de lage transfectie-efficiëntie van deze cellen, vooral in een rustige toestand. Om genen in immuuncellen te modificeren, worden medicijnresistentieselectie en virale vectoren meestal gebruikt om te verrijken voor cellen die het CRIPSR / Cas9-systeem tot expressie brengen, wat onvermijdelijk resulteert in ongewenste interventie van de cellen. In een eerdere studie ontwierpen we dubbele fluorescerende reporters gekoppeld aan CRISPR / Cas9 die tijdelijk tot expressie kwamen na elektroporatie. Deze technische oplossing leidt tot snelle gendeletie in immuuncellen; gen knock-in in immuuncellen zoals T-cellen en macrofagen zonder het gebruik van medicijnresistentieselectie of virale vectoren is echter nog uitdagender. In dit artikel laten we zien dat door celsortering te gebruiken om de selectie te ondersteunen van cellen die tijdelijk CRISPR / Cas9-constructies tot expressie brengen die gericht zijn op de Rosa26-locus in combinatie met een donorplasmide, gen-knock-in kan worden bereikt in zowel T-cellen als macrofagen zonder verrijking van medicijnresistentie. Als voorbeeld laten we zien hoe menselijke ACE2, een receptor van SARS-Cov-2, die verantwoordelijk is voor de huidige Covid-19-pandemie, tot expressie kan worden brengen in RAW264.7-macrofagen door knock-in-experimenten uit te voeren. Dergelijke gen knock-in cellen kunnen op grote schaal worden gebruikt voor mechanistische studies.
Immuuncellen zijn van cruciaal belang voor de verdediging tegen ziekteverwekkers. Zowel aangeboren als adaptieve immuniteit zijn vereist voor de klaring van infectanten en het behoud van weefselhomeostase1,2. Cellijnmodellen zijn essentiële hulpmiddelen voor het begrijpen van de moleculaire fundamenten van het immuunsysteem van zoogdieren; ze worden gebruikt in in vitro functionele assays, zoals die welke menselijke T-celactivatie modelleren, en bij het bepalen van de functie van genetische factoren bij het activeren of dempen van immuunresponsen3,4. Het is belangrijk op te merken dat het immuunsysteem van zoogdieren enorm heterogeen is en, even belangrijk, een groot aantal moleculen de differentiatie, migratie en functie van een bepaald celtyperegelen 5,6.
Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)/Cas9 genome editing tools maken genetische manipulatie van specifieke celtypen mogelijk, wat functionele annotatie van genen op een precieze maniervergemakkelijkt 7,8. Verschillende gepubliceerde protocollen hebben de levering van CRISPR / Cas9 beschreven in de vorm van Cas9-gids RNA-complexen die bekend staan als ribonucleoproteïnen (RSP’s) in HEK293-cellen, Jurkat-cellijnen, primaire T-cellen9,10,macrofagen11,12,13,stamcellen14en anderen15,16. In deze protocollen wordt gentagging meestal bereikt door een fluorescerende tag te fuseren met endogene eiwitten17,18. Er zijn echter weinig pogingen gedaan om dubbele fluorescerende reporters te gebruiken, die compatibel zijn met het sorteren van eencellige cellen, om knock-in experimenten19,20, met name in immuuncellen te vergemakkelijken.
Diepgaande mechanistische analyses gericht op het begrijpen van de functies van een nieuwe genetische factor in immuuncellen vereisen over het algemeen celtype specifieke deletie van een gen, genetische reddingsexperimenten en idealiter identificatie van de interactoren. Hoewel methoden voor optimalisatie van genetische deletie van genen in immuuncellen zijn gepubliceerd9,15,21,zijn er veel minder methoden gemeld voor het introduceren van knock-in allelen met veelzijdige functies om de immuunrespons te begrijpen. Daarom willen we in dit protocol in detail een efficiënt en zeer reproduceerbaar protocol beschrijven om een eiwit van belang (POI) tot expressie te brengen op de veiligehaven locus Rosa26 in zowel menselijke als muriene immuuncellijnen. We hebben een tweekleurig reportersysteem ontworpen om te verrijken voor cellen die zijn getransfecteerd met plasmiden die CRISPR / Cas9 (DsRed2) tot expressie brengen en een recombinant DNA-sjabloon (EBFP2), die kan worden geïsoleerd door celsortering. Volgens dit protocol verkregen we meerdere knock-in lijnen van de menselijke T-cellijn Jurkat en murine macrofaag RAW264.7 voor functionele analyses van slecht bestudeerde eiwitten.
Als voorbeeld laten we in dit protocol zien hoe je knock-in RAW264.7 macrofagen kunt verkrijgen die stabiel menselijke ACE2 (een receptor van SARS-Cov-2) tot expressie brengen22. Omdat aangeboren immuuncellen betrokken zijn bij de pathogenese van Covid-1923,24 en menselijk ACE2 wordt beschouwd als een belangrijke receptor die nodig is voor virale binnenkomst in cellen vóór replicatie, kunnen macrofagen met knock-in van humaan ACE2 dienen als een nuttig hulpmiddel voor mechanistische studies van virale vermenigvuldiging in macrofagen. Tegelijkertijd presenteren we ook een voorbeeld van knock-in van een gen op de menselijke ROSA26-locus om het RASGRP1-eiwit tot expressie te brengen, dat aan zijn amino-eindpunt was gefuseerd met een affiniteit Twin-Strep-tag (OST). T-cellen zijn belangrijke doelcellen in immuuntherapieën en een toenemend aantal studies heeft zich gericht op manipulatie van hun reactievermogen op kanker25,26. Aangezien Rasgrp1 bekend staat als een belangrijk signaalmolecuul stroomafwaarts van de T-celreceptor en de interactoren niet goed worden opgehelderd27, biedt het OST-RASGRP1 knock-in model de basis voor het identificeren van interactoren die de reactie van T-cellen op tumoren en infecties reguleren. Samen kunnen deze tools worden gebruikt voor Covid-19-studies en de ontdekking van nieuwe moleculen die interageren met Rasgrp1.
In onze experimenten hebben we aangetoond hoe we knock-in-bewerkingen in immuuncellen kunnen uitvoeren, van constructontwerp tot knock-in celscreening en validatie met behulp van menselijke Jurkat T-cellen en murine RAW264.7 macrofagen als voorbeelden.
