Die Calciumbildgebung hat eine ausreichende räumliche und zeitliche Auflösung und ist eine wichtige Methode zur Beobachtung der Ca2+ Dynamik in lebenden Zellen12,13.
Derzeit gibt es zwei Hauptmethoden für die intrazelluläre Kalziumbildgebung in lebenden Zellen: Eine verwendet chemische kleinmolekulare Fluoreszenzsonden14und die andere ist der Genkodierungssensor (GECI)15,16. Angesichts der Schwierigkeit, ein stabiles transgenes System in Obstbäumen und einer längeren Fruchtentwicklung aufzubauen, ist GECIS für die Fluoreszenzbildgebung von Früchten Ca2+ ungeeignet.
Niedermolekulare Fluoreszenzsonden wie Fluo-4/AM haben einen besonderen Vorteil: Ihre AM-Esterform (zelldurchlässiges Acetoxymethylesterderivat) kann ohne Transfektion leicht in lebende Zellen geladen werden, was sie flexibel, schnell und nicht zytotoxisch macht17.
